Any Gut-Ex-Vivo Method to examine Gut Infection Linked to Inflamed Bowel Condition (IBD).
A microbial co-occurrence analysis revealed a strong positive correlation between Nitrospira and several OTUs, some of which, such as Anaerolinea OTU, have been found to co-exist with comammox Nitrospira in the biofilms of water treatment systems. Given that these OTUs were abundant only on microbe-attached carriers in the system, their co-existence within the biofilm could be beneficial to stabilize the Nitrospira abundance, possibly by physically preventing oxygen exposure as well as cell spillage. The broad application of perfluoroalkyl acids (PFAAs) has attracted global concern regarding their adverse environmental effects. The possible removal processes of PFAAs in constructed wetlands were excavated and quantified using two typical submerged macrophytes (rooted Potamogeton wrightii and rootless Ceratophyllum demersum). Our results showed that 33.59-88.99% of PFAAs could be removed via not only sediment sorption or phytoextraction but also by the bioaccumulation of microbiota. The sediment acts as a vital sink for PFAAs, preloading 23.51-50.09% and 16.65-52.18% of PFAAs in treatments with P. wrightii (Pw1) and C. demersum (Cd1), respectively. C. demersum showed a better capacity to accumulate PFAAs (0.91-32.03%) than P. wrightii ( less then 10%). Considerable PFAAs were observed to be distributed in microbes, underlining the non-negligible role of microbiota in bioaccumulating PFAAs. The contributions of planktonic microbes, biofilm microbes, and extracellular polymeric substances in biofilms were 0.39-20.96%, 0.03-7.95%, and 0.39-14.15% in Pw1 and 0.23-15.68%, 0.01-15.68%, and 0.53-26.77% in Cd1, respectively. The adsorption/uptake was significantly correlated with the perfluoroalkyl chain length (p less then 0.05), except for the uptake of biofilms in C. demersum. Furthermore, PFAAs and submerged macrophytes could decrease the richness of microbiota but increase the relative abundance of some strains in Betaproteobacteriales, Sphingomonadales, and Cytophagales. Our results were helpful for understanding the removal processes of PFAAs in constructed wetlands and their linkages with PFAA properties, thus further providing insight into the management and removal of emerging organic contaminants.Engineered nanoparticles are observed to be released into the environment and ended up in wastewater treatment plants. It has been reported that these nanoparticles in sewage might have a toxic effect on microorganisms, and thus affect anaerobic microbial fermentation. However, the mechanisms involved in nanoparticles-induced effects on the anaerobic acidification process and its related bacterial metabolism are still unclear. This work indicated that copper nanoparticles (Cu NPs) were able to cause cell membrane oxidative damage and inhibit the growth and metabolism of Moorella thermoacetica (a model acetogen). The OD600 and acetic acid production of M. thermoacetica in the presence of 1 mg/L of Cu NPs were decreased to 29.2% and 40.7% of the control, respectively. The key mechanism of the inhibitory effect was governed by the fact that Cu NPs significantly reduced the glucose consumption, and led to the decreased pyruvate metabolism levels. Additionally, Cu NPs inhibited the gene expressions and catalytic activities of the key enzymes related to acetic acid production. It was identified that the relative activities of phosphofructokinase, pyruvate kinase, phosphotransacetylase, and acetate kinase of M. thermoacetica in the presence of 1 mg/L of Cu NPs decreased to only 70.1%, 69.3%, 50.1%, and 65.2% of the control, respectively. These results demonstrated that the release of Cu NPs in the environment could pose risks to anaerobic fermentation processes via regulating microbial transcriptional response and enzyme activity. The article presented by Wang et al. (2020) intends to elucidate the possible ecological effects of low (0.05-25 μg L-1) and higher (100 μg L-1) concentrations of methamphetamine on adult Oryzias latipes through a battery of assays, including histopathology. However, we found several mistakes and inaccuracies in the findings by means of this method. Given the increasing research effort in the field, the authors' paper may become highly influential in future toxicological research. Despite the authors' undoubted effort invested in their experiment, they did not employ standardized methods for histopathological assessment in the key laboratory experiment presented in their paper.Particulate organic carbon (POC) sources, which regulate dissolved organic carbon, sediment organic carbon, and inorganic carbon via deposition, degradation, and mineralization, play an important role in lake ecosystems. Linear or Bayesian algorithms on isotope and n-alkanes have been widely used to identify the source proportion of organic carbon. However, the applicability of these methods is ambiguous because of the unilateral advantages of each model and trace factors. To test the applicability of the various methods for identifying POC sources, we analyzed dual isotopes and n-alkanes in surface water samples of Lake Taihu, and Multi-source mixing model and Bayesian mixing model were used to distinguish between endogenous and exogenous contributions. Carbon isotope presented a clear advantage in West Taihu (-21.85 ± 0.78‰) and Southwest Taih (-22.61 ± 1.35‰); nitrogen isotope also showed high values in Meiliang Bay (9.76 ± 0.92‰). The majority of the lake was dominated by short-chain n-alkanes, except for East Taihu Lake (dominated by medium-chain n-alkanes) and areas with riverine input (dominated by long-chain n-alkanes). Different principles between the Bayesian mixing model (based on the Markov Chain Monte Carlo algorithm) and the Multi-source mixing model (based on linear estimation) caused discrepancies in the estimations of source contributions. read more But the fraction of chemical compounds during the migration process, and the overlap of potential sources play important role in the inconsistency of results. The estimations from the different models were consistent in indicating the dominance of endogenous organic carbon in Lake Taihu (mean of 60.18 ± 20.26%), particularly in the north and western regions (West Taihu, Meiliang Bay, and Southwest Taihu). This was likely due to algal aggregation influenced by human activities and climatic factors.Enhanced covalent immobilization of xylanase from Chaetomium globosum (XylCg) onto SiO2 nanoparticles was achieved by the modification of surface residues. The mutation of surface residues to lysine by site-directed mutagenesis increased the immobilization efficiency (IE) and immobilization yield (IY). The immobilized mutant XylCg (N172K-H173K-S176K-K133A-K148A) exhibited an IY of 99.5% and IE of 135%, which were 1.8- and 4.3-fold higher than immobilized wildtype (WT). Regarding the catalytic properties, the kcat and kcat/Km values were 1850 s-1 and 2030 mL mg-1 s-1 for the immobilized mutant, and 331 s-1 and 404 mL mg-1 s-1 for the immobilized WT, respectively. Additionally, the immobilized mutant exhibited four times higher thermal stability than the immobilized WT at 60 °C. These results suggest that surface-mutated lysine residues confer good stability and orientation on the support matrix, thus improving the overall performance of xylanase.Two-phase high-solid digestion is conducive to the degradation of food waste. In this study, Fe/C was added in high-solid digestion in different acidification and/or methanogenic phase. The experimental results indicated that it significantly increased the cumulative yield of biomethane. When Fe/C was added to the acidification phase only and both the acidification and methanogenic phases, the biomethane yield reached 474.07 ± 7.03 and 475.47 ± 4.68 mL·g VS -1, respectively, and the biodegradation rate reached 87.30% and 87.58%, respectively, indicating that Fe/C had mainly effect on the performance of acidification phase. In a two-phase anaerobic fermentation system, the activity of dehydrogenases and the concentration of coenzyme F420 were 2.23-2.95 mg·g-1·h-1 and 0.0063-0.0294 mol·g-1 volatile solids, respectively. Additionally, the archaeal communities production pathway of methane from using acetic acid to using hydrogen as the reactant. The major share of phosphate in biogas digestate is inorganic. For optimized nutrient recovery, inorganic phosphate must be analyzed adequately. Therefore, the photometric spectra of extracts from Hedley fractionation were measured and analyzed for their peaks using the molybdenum blue method. The ideal wavelength was 709 nm. The lower calibration limit needed to be raised from 15 µg L-1 to 50 µg L-1 to avoid underestimation of phosphate concentration. Drying digestate before extraction increased H2O-P by 78.4% and NaHCO3-P by 44.9% compared to undried digestate. The filter paper of the filtration between extractions was added to the next extraction to avoid phosphate losses. This made it necessary to rinse the samples with 30 mL deionized H2O after the H2O extraction, with 60 mL NaHCO3 after NaHCO3 extraction and 60 mL NaOH after the NaOH extraction. Ultimately, the results showed that the phosphate concentration in extracts was independent of extraction time.Solid state anaerobic digestion (SSAD) of lignocellulosic biomass may be attractive solution for its valorisation. Compared to liquid state anaerobic digestion (LSAD), SSAD can handle higher organic loading rates (OLR), requires a less water and smaller reactor volume. It may require lower energy demand for heating or mixing and has higher volumetric methane productivity. Besides numerous benefits of SSAD processes and progress in system design, there are still obstacles, which need to be overcome for its successful implementations. This review aims to compile the recent trends in enhancing the bioconversion of agricultural stubbles in SSAD. Several pretreatment procedures used to breaking lignin and cellulose complex, method to overcome carbon to nitrogen ratio imbalance, use of carbon-based conducting materials to enhance Volatile Fatty Acids (VFA) conversion and additives for achieving nutrient balance will be discussed in this review. Leachate recirculation and its impacts on SSAD of agricultural stubbles are also discussed. The Arabian Peninsula's advantageous climate, availability of non-arable land, access to seawater and CO2-rich flue gas, make it an attractive location for microalgae biomass production. Despite these promising aspects, the region has seen very few studies into the commercial feasibility of algae-based value chains. This work aims to address this gap through a techno-economic feasibility study of algae biomass production costs, comparing different photobioreactor types, locations, and production scales. Flat panel and raceway pond cultivation systems were found to be the most economically attractive cultivation systems, with biomass production costs as low as 2.9 €·kg-1. Potential cost reductions of up to 42.5% and 25% could be accomplished with improvements in photosynthetic efficiencies and increased culture temperatures, respectively. As of such, efforts to source local thermo- and photo- tolerant strains could be the key to unlock the potential of the region for algae commercialization, linking into food, feed and nutraceutical industries.
