They can be constructed using various CAD/CAM materials that can provide a modulus of elasticity similar to that of teeth. The ability to use of such materials in composite blocks that can be easily repaired is also an advantage, provided appropriate bonding performance is ensured. This study, therefore, evaluated the marginal gap and fracture resistance of two CAD/CAM endocrown materials using two bonding protocols. Thirty-two mandibular molars were evaluated in two groups based on the material type a Cerasmart group (GC America Inc; n = 16) acting as the control and a Ceramill COMP group (Amann Girrbach, Germany; n = 16). These groups were then classified according to the bonding protocol used a total-etch bonding protocol (n = 8) and a self-etch bonding protocol (n = 8) implemented using RelyX ultimate adhesive resin cement (3M ESPE). The samples were then subjected to aging by simulating a 1-year thermo-mechanical process. The marginal gap results were statistically insignificant across the material and bonding protocol groups before thermo-mechanical aging. Thermo-mechanical aging significantly reduced the marginal gap distance for Ceramill COMP endocrowns cemented using the total-etch protocol (p = 0.002). No statistically significant difference was recorded for the fracture resistance in either the material or bonding protocol groups (p ≥ 0.05). Both materials and bonding protocols can, therefore, be used in the posterior region providing conservative treatment, adequate marginal gap and fracture resistance. © 2020 The Author(s). Remedial actions to resolve the corresponding lead in water crisis included reconnection to the original Lake Huron source water with orthophosphate, implementing enhanced corrosion control by dosing extra orthophosphate, a "Flush for Flint" program to help clean out loose leaded sediment from service lines and premise plumbing, and eventually lead service line replacement. Independent sampling over a period of 37 months (January 2016-February 2019) was conducted by the United States Environmental Protection Agency and Virginia Tech to evaluate possible human exposure via normal flow (∼2-3 L/min) sampling at the cold kitchen tap, and to examine the status of loose deposits from the service line and the premise plumbing via high-velocity flushing (∼12-13 L/min) from the hose bib. The sampling results indicated that high lead in water persisted for more than a year in two Flint homes due to a large reservoir of lead deposits. The effects of a large reservoir of loose lead deposits persisted until the lead service line was completely removed in these two anomalous homes. As water conservation efforts are implemented in many areas of the country, problems with mobile lead reservoirs in service lines are likely to pose a human health risk. © 2020 The Authors.Background & Aims Metabolism supports cell proliferation and growth. Surprisingly, the tumor suppressor miR-22 is induced by metabolic stimulators like bile acids. Thus, this study examines whether miR-22 could be a metabolic silencer. Methods The relationship between miR-22 and the expression of fibroblast growth factor 21 (FGF21) and its receptor FGFR1 was studied in cells and fatty livers obtained from patients and mouse models. We evaluated the effect of an miR-22 inhibitor alone and in combination with obeticholic acid (OCA) for the treatment of steatosis. Results The levels of miR-22 were inversely correlated with those of FGF21, FGFR1, and PGC1α in human and mouse fatty livers, suggesting that hepatic miR-22 acts as a metabolic silencer. Indeed, miR-22 reduced FGFR1 by direct targeting and decreased FGF21 by reducing the recruitment of PPARα and PGC1α to their binding motifs. In contrast, an miR-22 inhibitor increases hepatic FGF21 and FGFR1, leading to AMPK and ERK1/2 activation, which was effective i of fatty liver. Consistent with this finding, an miR-22 inhibitor effectively reversed both alcohol- and diet-induced fatty liver; miR-22 inhibition is a promising therapeutic option which could be used in combination with obeticholic acid. © 2020 The Author(s).Background & Aims End-ischemic hypothermic oxygenated machine perfusion (HOPE) of the donor liver for 1-2 h mitigates ischemia-reperfusion injury during subsequent liver transplantation. Extended preservation time may be preferred to facilitate difficult recipient hepatectomy or to optimize logistics. We therefore investigated whether end-ischemic dual HOPE (DHOPE) could extend preservation time for up to 24 h using a porcine liver reperfusion model. Methods Following 30 min warm ischemia, porcine livers were subjected to 2 h static cold storage (SCS), followed by 2 h, 6 h, or 24 h DHOPE (n = 6 per group). Subsequent normothermic reperfusion was performed for 4 h using autologous blood. Two livers preserved by 24 h SCS served as additional controls. A proof of principle confirmation was carried out in 2 discarded human livers subjected to extended DHOPE. Hepatocellular and cholangiocyte injury and function were assessed. Oxidative stress levels and histology were compared between groups. This extended procedure could be used globally to facilitate transplantation and expand the donor pool. © 2020 The Author(s).Background & Aims The inverse association between non-alcoholic fatty liver disease (NAFLD) and diets rich in fruit and vegetables has been demonstrated, but the specific compounds that may be responsible for this association need to be elucidated. The aim of this study was to test the association between phenolic acid consumption, NAFLD, and insulin resistance (IR). Methods A cross-sectional cohort of individuals included in a metabolic screening program was studied. Liver steatosis was evaluated by ultrasonography and quantified by the hepatorenal index (HRI); fibrosis was assessed by FibroTest; IR by the sample upper quartile of the homeostatic model assessment score. Dietary intake was measured by a food frequency questionnaire. The phenolic acid content of food was calculated according to Phenol-Explorer. Results A total of 789 individuals were included (52.6% men, age 58.83 ± 6.58 years). Higher (above the upper median) phenolic acid intake was inversely associated with the presence of NAFLD (odds ratiof hydroxycinnamic acids, another class of phenolic acids, is associated with a lower prevalence of insulin resistance. © 2020 The Author(s).The NIR excitation and large anti-Stokes shift of upconversion nanoparticles (UCNPs) have made them an ideal choice as biological nanoprobes. A key challenge in the field is to confer biorecognition units to UCNPs so that they can be used to probe specific targets in biological systems. While various agents have been combined with UCNPs to meet such a challenge, most studies are limited to small molecules, while biomolecules such as metalloproteins that possess much higher affinity and selectivity for metal ions have not been explored. Herein we demonstrate that fusion of zwitterion-coated UCNPs with azurin, a member of a family of redox-active copper proteins called cupredoxins that play important roles in diverse biological functions, can serve as an ideal platform for the label-free upconversion luminescence sensing of Cu2+ with a ratiometric response. The selectively binding of apo-azurin with Cu2+ induce a significant absorbance at about 625 nm, and hence decreases the red emission of the UCNPs. In contrast, the green emission of the UCNPs remains constant and acts as an internal standard reference for the ratiometric sensing of Cu2+. This approach opens a new window for the development of assays for biosensing based on a combination of specific metalloproteins with UCNPs.Background Nightshift work is a plausible risk factor for hematologic cancer, but epidemiological evidence remains sparse, especially for individual subtypes. We prospectively examined the association of rotating nightshift work with hematopoietic cancer risk. Methods This cohort study included US women from the Nurses' Health Study (NHS n = 76 846, 1988-2012) and Nurses' Health Study II (NHSII n = 113 087, 1989-2013). Rotating nightshift work duration was assessed at baseline (both cohorts) and cumulatively updated (NHSII). Fasiglifam Cox regression was used to estimate hazard ratios (HRs) and 95% confidence intervals (CIs) for overall hematopoietic cancer and specific histologic subtypes. Dans l’etude actuelle, les propriétés psychométriques d’une échelle d’évaluation environnementale ont été évaluées de manière empirique à l’aide de données provenant d’une étude menée au niveau communautaire au Nigeria. Différentes mesures d’évaluation environnementale ont été regroupées dans une seule échelle d’évaluation et administrées à 1 858 personnes. La moyenne (type écart) et les proportions ont été utilisées pour décrire la distribution des données continues et discrètes, respectivement. La fiabilité de l’échelle a été évaluée à l’aide du coefficient alpha de Cronbach. Différents modèles hypothétiques de l’AE ont été évalués à l’aide d’une analyse factorielle exploratoire (AFE) et d’une analyse factorielle confirmatoire (AFC). Les données ont été analysées avec IBM SPSS version 20 et AMOS version 21 à un niveau significatif de 5%. Le score moyen global et le coefficient alpha de la mesure de l’EA combinée étaient respectivement de 381,7 (49,0) et 0,928. Une structure à 3 facteurs représentant une variance cumulée de 36% de l’élément d’échelle a été extraite dans un EFA. Un modèle à 3 facteurs non orthogonal constituait une amélioration significative par rapport au modèle original à 5 facteurs sans corrélation [χ2 (1344) = 29216,33, p less then 0,001; CAIC = 9816.870]. La conceptualisation de l’AE en tant que structure à 3 facteurs non orthogonale offre un meilleur ajustement aux données actuelles. La structure à 3 facteurs est conseillée au Nigeria et dans des contextes similaires.
