After 48 h treatment, botryosphaeran induced nitric oxide production (NO), caused mitochondrial membrane hyperpolarization, increased reactive oxygen species (ROS), and accumulation of lipid vesicles in promastigotes, resulting in apoptosis, necrosis and autophagy, and was accompanied by morphological and ultrastructural changes. The range of concentrations used did not alter the viability of peritoneal macrophages from BALB/c mice and erythrocytes of sheep. Botryosphaeran was able to reduce the number of infected macrophages and the number of amastigotes per macrophage at 12.5 μg/mL (50.75% ± 6.48), 25 μg/mL (55.66% ± 3.93) and 50 μg/mL (72.9% ± 6.98), and IC50 9.3 μg/mL (±0.66) for intracellular amastigotes forms. The leishmanicidal effect was due to activation of NF-κB and promoted an increase in pro-inflammatory cytokines (TNF-α and IL-6), iNOS and microbial-derived ROS and NO, in addition to decreasing the levels of SOD. Based upon the data obtained, we infer that botryosphaeran exerted an active leishmanicidal and immunomodulatory effect, acting on promastigotes through autophagic, apoptotic and necrosis processes, and in the intracellular amastigote form, through the action of ROS and NO.
To examine patient laboratory testing compliance by tracking time to submission of laboratory requisitions in Southern Alberta, Canada as part of a quality improvement initiative.

Data was collected retrospectively from patients from the Chinook Primary Care Network in Alberta, Canada, who received a laboratory requisition consisting of a complete blood count (CBC) test order between September 1, 2016 and August 31, 2017. To allow for all laboratory requisitions created to be submitted within one year, the study collection period was from September 1, 2016 to August 31, 2018. Patient age, sex, and dates of laboratory requisition creation and submission were collected. The days-to-test-submission served as a marker of compliance. Association of age, sex, and clinic location with time to laboratory requisition completion was determined using Cox regression analysis.

During the study period, 70.4% (n=1607) of laboratory requisitions created were completed within one year, and over half (50.5%) of the laborly manner.

Among CECs are pharmaceuticals like 17 α-ethinylestradiol (EE2), which is a synthetic hormone with high estrogenic potency. EE2 has been increasingly found in different aquatic systems but few studies addressed its potential toxicity to marine wildlife, in particular to bivalves. Therefore, the aim of the present study was to evaluate the influence of temperature (17 °C-control and 21 °C) on the potential effects of EE2 on the mussel Mytilus galloprovincialis. For this purpose, mussels were exposed to different concentrations of EE2 (5.0; 25.0; 125.0 and 625 ng/L), resembling low to highly polluted sites. Mussels exposed to each concentration were maintained under two temperatures, 17 and 21 °C, wh predicted warming conditions will enhance the impacts, which in a near future might result in impairments at the population and community levels.This study aims to investigate the coronavirus disease (COVID-19) pandemic effects and associated restrictive rules on ship activities and pollutant emissions (CO2, SOX, NOX, PM, CO, CH4) in four major seaports, namely the Ports of Singapore, Long Beach, Los Angeles, and Hamburg. We used 2019 as the baseline year to show the business-as-usual emission and compared with the estimated quantity during the July 2020-July 2021 pandemic period. We also project future ship emissions from August 2021-August 2022 to illustrate two potential port congestion scenarios due to COVID-19. The results show that the ship emissions in all four ports generally increased by an average of 79% because of the prolonged turnaround time in port. Importantly, majority of ship emissions occurred during the extended hoteling time at berth and anchorage areas as longer operational times were needed due to pandemic-related delays, with increases ranging from 27 to 123% in the total emissions across ports. The most affected shipping segments were the container ships and dry bulk carriers which the total emissions of all pollutants increased by an average of 94-142% compared with 2019. Overall, the results of this study provide a comprehensive review of the ship emission outlook amid the pandemic uncertainty.
While evidence suggests that daily ambient temperature exposure influences stroke risk, little is known about the potential triggering role of ultra short-term temperature.

We examined the association between hourly temperature and ischemic and hemorrhagic stroke, separately, and identified any relevant lags of exposure among adult New York State residents from 2000 to 2015. Cases were identified via ICD-9 codes from the New York Department of Health Statewide Planning and Reearch Cooperative System. We estimated ambient temperature up to 36h prior to estimated stroke onset based on patient residential ZIP Code. We applied a time-stratified case-crossover study design; control periods were matched to case periods by year, month, day of week, and hour of day. Additionally, we assessed effect modification by leading stroke risk factors hypertension and atrial fibrillation.

We observed 578,181 ischemic and 164,755 hemorrhagic strokes. However, available information on heavy metals in the containers is extremely limited and the associated health risks remain poorly understood. We investigated Cd, Cr, Pb, Sb, Mn, Ni and Co in the popular take-out food containers from China and found the concentrations of these metals were moderate in comparison to the concentrations reported in other food contact materials. The metal concentrations in sampled containers by material type differed significantly, and higher concentrations of Cd, Pb, Sb, Mn and Co were observed in expanded polystyrene samples. The metals in the containers likely originated from intentional addition and/or contamination of materials. The potential release of the metals from containers was simulated and found the median leaching rates of Cd, Pb, Sb, Ni and Co in the range of 0.36-4.80% under typical conditions, which depended largely on the material types. Based on the observed leaching rates, we estimated that the summed carcinogenic risks of Cd, Pb, Ni and Co were unacceptable under specific exposure frequency, although the total non-carcinogenic risks from metal intake were low.Exposure to air pollution, including criteria pollutants such as fine particulate matter (PM2.5) and ozone (O3), has been associated with morbidity and mortality in mammals. As a genetically homogenous population that is closely monitored for health, dairy cattle present a unique opportunity to assess the association between changes in air pollution and mammalian health. Milk yield decreases in the summer if temperature and humidity, measured by the Temperature Humidity Index (THI). As O3 levels increase with warmer temperatures, and summer PM2.5 may increase with wildfire smoke, dairy cows may serve as a useful sentinel species to evaluate subacute markers of inflammation and metabolic output and ambient pollution. Over two years, we assessed summertime O3 and PM2.5 concentrations from local US EPA air quality monitors into an auto-regressive mixed model of the association between THI and daily milk production data and bulk tank somatic cell count (SCC). In unadjusted models, a 10 unit increase THI was associated with 28,700 cells/mL (95% CI 17,700, 39,690) increase in SCC. After controlling for ambient air pollutants, THI was associated with a 14,500 SCC increase (95% CI 3,400, 25,680), a 48% decrease in effect compared to the crude model. Further, in fully adjusted models, PM2.5 was associated with a 105,500 cells/mL (95% CI 90,030, 121,050) increase in SCC. Similar results were found for milk production. Results were amplified when high PM2.5 days (95th percentile of observed values) associated with wildfire smoke were removed from the analyses. Our results support the hypothesis that PM2.5 confounds the relationships between THI and milk yield and somatic cell count. The results of this study can be used to inform strategies for intervention to mitigate these impacts at the dairy level and potentially contribute to a model where production animals can act as air quality sentinels.
