If mice were treated with total body irradiation, they showed significant apoptosis in bone marrow monocytes, but not in peritoneal macrophages. This was also observed after treatment with the methylating anticancer drug temozolomide, resulting in high death rate of monocytes, but not macrophages. Monocyte monocytes appears to be a general property of this cell type.
Ethical decisions about an allowance for animal experiments need to be based on scientifically sound information about the burden and distress associated with the experimental procedure and models. Thereby, species differences need to be considered for recommendations regarding evidence-based severity assessment and refinement measures.

A comprehensive analysis of behavioral patterns and corticosterone or its metabolites in serum and feces was completed in kindled mice. The impact of kindling via two different stimulation sites in the amygdala and hippocampus was determined. Data were compared to those from naive and electrode-implanted groups.

Amygdala and hippocampus kindled mice exhibited comparable behavioral patterns with increased activity in the open field, reduced anxiety-associated behavior in the elevated-plus maze, and increased anhedonia-associated behavior in the saccharin preference test. In addition, repeated stimulation of the hippocampus caused a reduction in burrowing behavior and an ir the clinical management of respective psychiatric comorbidities in patients.
Taken together the findings suggest a severity classification for the mouse kindling paradigms as moderate regardless of the stimulation site. The outcome of the species comparison provides valuable guidance for species selection for studies exploring behavioral comorbidities. In this context, it is emphasized that the mouse kindling paradigms seem to be well suited for studies exploring the link between ictal events and network alterations on the one hand, and hyperactivity and anhedonia-associated behavior on the other hand. Moreover, the underlying pathophysiological mechanisms and the impact of therapeutic interventions on these behavioral alterations can be studied in these paradigms providing guidance for the clinical management of respective psychiatric comorbidities in patients.The objective assessment of biomechanical asymmetries during movement tasks is used to monitor rehabilitation following anterior cruciate ligament reconstruction (ACLR). Marker placement is an important source of methodological variability within human motion analysis. It is currently unclear how marker placement error effects the interpretation of biomechanical asymmetries throughout post ACLR rehabilitation. The aim of this investigation was to determine the effect of random marker placement variation on the interpretation of inter-limb differences during a change of direction (CoD) task. Forty-seven participants 9 months post-ACLR and fifty uninjured controls completed a 90° CoD task on both limbs. Inter-limb differences in kinematic and kinetic metrics during the CoD stance phase were calculated for both groups using the Vicon Plug-in Gait model, and ACLR subjects were classified as having 'normal' or 'abnormal' inter-limb differences relative to the control group. Simulated random marker displacements based on published marker placement error ranges were then repeatedly applied to the lateral thigh, femoral epicondyle and tibia markers. ACLR inter-limb differences were recalculated each time, allowing the estimation of 95% confidence intervals and minimal identifiable between-session changes. ACLR subjects were also reclassified relative to the control group after each simulation and the percentage of participants to change classification was calculated. Marker displacements caused large deviations in inter-limb difference measures in several variables including hip rotation angle, knee abduction angle and knee abduction moment, thus limiting the ability to identify participants with large inter-limb differences relative to a control group. Furthermore, this sensor was successfully employed to monitor Sn2+ and Cu2+ in practical samples.This work investigates the nature of the molecular interactions between water vapor and polymers using time series Fourier transform infrared (FTIR) spectroscopy. A detailed analysis of the frequency shifts and relative peak intensities led to the conclusion that polyvinyl alcohol (PVOH) sorbed a large quantity of water vapor molecules, resulting in swelling and dissolving of polymer crystallites. Difference spectra were calculated to investigate spectral changes occurring upon sorption by dividing the spectra of polymers during the sorption time series by the spectrum of the dry sample and subsequently subtracting the water vapor spectrum. Based on the absorbance area of the OH stretching vibration region (4000-3000 cm-1) in difference spectra, the amount of water sorbed was significantly higher in poly-L-lactic acid (PLLA) and polyvinyl chloride (PVC) than in polyethylene (PE) and polytetrafluoroethylene (PTFE), increasing with the hydrophilicity of the surface. The OH stretching band of difference spectra shifted from 3499 cm-1 for PVC, to 3416 cm-1 for PE and finally to 3387 cm-1 for PTFE, indicating a more strengthened hydrogen-bonding network in the PTFE matrix upon water vapor sorption.We present a UVRR spectroscopy setup which is equipped with a picosecond pulsed laser excitation source continuously tunable in the 210-2600 nm wavelength range. This laser source is based on a three-stage optical parametric amplifier (OPA) pumped by a bandwidth-compressed second harmonic output of an amplified YbKGW laser. It provides less then 15 cm-1 linewidth pulses below 270 nm, which is sufficient for resolving Raman lines of samples in condensed phase studies. For demonstrating the capability of this tunable setup for UVRR spectroscopy we present its application to the artificial ligand guanidiniocarbonyl pyrrole (GCP), a carboxylate binder used in peptide and protein recognition. A UVRR excitation study in the range 244-310 nm was performed for identifying the optimum laser excitation wavelength for UVRR spectroscopy of this ligand (λmax = 298 nm) at submillimolar concentrations (400 µM) in aqueous solution. The optimum UVRR spectrum is observed for laser excitation with λexc = 266 nm. Only in the relatively narrow range of λexc = 266-275 nm UVRR spectra with a sufficiently high signal-to-noise ratio and without severe interference from autofluorescence (AF) were detectable. At longer excitation wavelengths the UVRR signal is masked by AF. At shorter excitation wavelengths the UVRR spectrum is sufficiently separated from the AF, but the resonance enhancement is not sufficient. A label-free sensing assay based on the enzyme-mimicking property of Cytidine-Au nanoclusters (Cy-AuNCs) was demonstrated for colorimetric detection of GSH, GSSG and glutathione reductase (GR). Firstly, obvious blue color accompanied with an absorption peak at 652 nm was observed due to the high peroxidase-like activity of Cy-AuNCs toward 3,3',5,5'-tetramethylbenzidine (TMB). Then, in the presence of target, the mimetic activity of Cy-AuNCs could be strongly inhibited and used to achieve the visualization detection. The inhibition effect arose from the surface interaction between GSH and Cy-AuNCs. Interestingly, Hg2+ is discerned by opening the rhodamine spirocycle and by generating a new special cycle for the quinoline unit. Resultantly, there were two UV-Vis absorption bands at around 365 nm and 560 nm, which were accompanied by fluorescence turn-on. Moreover, the chemosensor can quantitatively detect Cu2+ and Hg2+ by off-on fluorescence and ratiometric UV-Vis absorption changes, respectively.
