Small cell lung cancer (SCLC), considered a mortal recalcitrant cancer, is a severe healthcare issue because of its poor prognosis, early metastasis, drug resistance and limited clinical treatment options. In our previous study, we established a MRP1-targeted antibody-IR700 system (Mab-IR700) for near infrared photoimmunotherapy (NIR-PIT) which exhibited a promising therapeutic effect on drug resistant H69AR cells both in vitro and in vivo, though the tumor growth suppression effect did not last long with a single round of PIT treatment. To achieve a better anticancer effect, we have combined Mab-IR700-mediated NIR-PIT with liposomal doxorubicin (Doxil®) and investigated the in vitro and in vivo cytotoxicity by using a H69AR/3T3 cell co-culture model in which 3T3 cells were used to mimic stromal cells. Cytotoxicity experiments demonstrated the specificity of Mab-IR700 to H69AR cells, while cytotoxicity and flow cytometry experiments confirmed that H69AR cells were doxorubicin-resistant. Compared with Mab-IR700-mediated PIT or Doxil-mediated chemotherapy, the combination therapy exhibited the best cell killing effect in vitro and superior tumor growth inhibition and survival prolongation effect in vivo. Super enhanced permeability and retention (SUPR) effect was observed in both co-culture spheroids and tumor-bearing mice. Owing to an approximately 9-fold greater accumulation of Doxil within the tumors, NIR-PIT combined with Doxil resulted in enhanced antitumor effects compared to NIR-PIT alone. This photoimmunochemotherapy is a practical strategy for the treatment of chemoresistant SCLC and should be further investigated for clinical translation.Biofiber is a new generation of highly absorbent, and textured bandage with patented fiber technology. A growing number of non-coding RNAs (ncRNAs), for instance, microRNAs, have been identified whose abnormal expression patterns have been implicated in HNSCC. However, more recently, several seminal research has shown that piwi-interacting RNAs (piRNAs), a promising and young class of small ncRNA, are linked to the emergence and progression of cancer. They can regulate transposable elements (TE) and gene expression through multiple mechanisms, making them potentially more powerful regulators than miRNAs. Hence, they can be more promising ncRNAs candidates for cancer therapeutic intervention. Here, we surveyed the roles and clinical implications of piRNAs and their PIWI proteins partners in tumorigenesis and associated molecular processes of cancer, with a particular focus on HNSCC, to offer a new avenue for diagnosis, prognosis, and therapeutic interventions for the malignancy, improving patient's outcomes.The potential effect of gestational exposure to phthalates on the lung function levels during childhood is unclear. Therefore, we examined this association at different ages (from 4 to 11 years) and over the whole childhood. Specifically, we measured 9 phthalate metabolites (MEP, MiBP, MnBP, MCMHP, MBzP, MEHHP, MEOHP, MECPP, MEHP) in the urine of 641 gestating women from the INMA study (Spain) and the forced vital capacity (FVC), forced expiratory volume in 1 s (FEV1) and FEV1/FVC in their offspring at ages 4, 7, 9 and 11. We used linear regression and mixed linear regression with a random intercept for subject to assess the association between phthalates and lung function at each study visit and for the overall childhood, respectively. We also assessed the phthalate metabolites mixture effect on lung function using a Weighted Quantile Sum (WQS) regression. We observed that the phthalate metabolites gestational levels were consistently associated with lower FVC and FEV1 at all ages, both when assessed individually and jointly as a mixture, although most associations were not statistically significant. Of note, a 10% increase in MiBP was related to lower FVC (-0.02 (-0.04, 0)) and FEV1 z-scores (-0.02 (-0.04, -0.01) at age 4. Similar significant reductions in FVC were observed at ages 4 and 7 associated with an increase in MEP and MnBP, respectively, and for FEV1 at age 4 associated with an increase in MBzP. WQS regression consistently identified MBzP as an important contributor to the phthalate mixture effect. We can conclude that the gestational exposure to phthalates was associated with children's lower FVC and FEV1, especially in early childhood, and in a statistically significant manner for MEP, MiBP, MBzP and MnBP. Given the ubiquity of phthalate exposure and its established endocrine disrupting effects in children, our findings support current regulations that limit phthalate exposure.Heavy metal contamination and low use efficiency of phosphorus (P) fertilizers are worldwide issues. Alkaline lignin is expected to decrease the heavy metal risk and enhance the P availability in heavy-metal-contaminated soils. A 120-day incubation study examined the effects of alkaline lignin on Cd, Pb and P bioavailability and transformation in Cd or Cd/Pb co-contaminated red and cinnamon soils and elucidated the associated mechanisms. A pot experiment further tested Cd accumulation in lettuce (Lactuca sativa L.) grown in the Cd-contaminated red soil. The amendment of alkaline lignin increased the concentrations of bioavailable Cd by 13-20% in the acid red soil and 97-107% in the alkaline cinnamon soil, respectively, due to the increase of dissolved organic C concentrations. Meanwhile, it also increased the concentrations of available P in both soils, Al-P in the red soil and Ca2-P in the cinnamon soil. Consequently, alkaline lignin amendment increased lettuce biomass of shoots by 8-23% and of roots by 56-71%, P uptake by 37-50% in shoots and by 28-62% in roots, and limited Cd transport from root to shoot which decreased Cd concentrations by 26% in lettuce shoot (edible part). The results suggest that alkaline lignin increases plant growth and decreases Cd bioaccumulation in the shoot through restricting Cd translocation from the root to shoot and increasing soil P availability but not Cd immobilization, and hence may have potential to reduce vegetable Cd contamination risk.The intestine is not only the main accumulation organ of microplastics (MPs), but also the intestinal environment is very conductive to the release of additives in MPs. However, the kinetics of release process, influence factors, and the related effects on gut microbiota remain largely unknown. In this study, a mucosal-simulator of the human intestinal microbial ecosystem (M-SHIME) was used to investigate the influence of gut microbiota on the release of phthalates (PAEs) from MPs and the effects of MPs on the intestinal luminal microbiota and mucosal microbiota. We found that di-(2-ethylhexyl) phthalate (DEHP), di-n-butyl phthalate (DBP), and dimethyl phthalate (DMP) were the dominant PAEs released in the gut. Gut microbiota accelerated the release of PAEs, with the time to reach the maximum release was shortened from 7 days to 2 days. Moreover, MPs induced differential effects on luminal microbiota and mucosal microbiota. Compared with mucosal microbiota, the luminal microbiota was more susceptible to the leaching of PAEs from MPs, as evidenced by more microbiota alterations. MPs also inhibited the metabolic activity of intestinal flora based on the reduced production of short chain fatty acids (SCFA). These effects were mainly contributed by the release of PAEs. Acidaminococcus and Morganella were simultaneously correlated to the release of PAEs and the inhibition of metabolic activity of intestinal microbiota and can be used as indicators for the intestinal exposure of MPs and additives.Glyphosate-based herbicides are used worldwide, and glyphosate's primary metabolite (aminomethylphosphonic acid AMPA), is globally retrieved in surface waters. AMPA induces various adverse effects on aquatic wildlife, including selective mortality, which suggests that glyphosate exposure may have selected for AMPA-resistant individuals. We tested this hypothesis using spined toads (Bufo spinosus), an amphibian found in a variety of habitats, from AMPA-exposed agricultural lands to AMPA-free forested areas. We predicted that the offspring of individuals originating from agricultural habitats would develop AMPA-resistance - and be less prone to develop adverse effects from- AMPA exposure. To investigate this question, we performed a common garden brood-rearing experiment. The embryos and larvae of 40 spined toad pairs captured in agricultural and forest ponds were exposed either to an environmental relevant concentration of AMPA (0.4 μg L-1) or to control conditions (n = 8160 embryos, n = 240 tadpoles). We monitored development durations, developmental abnormalities and morphology, measured across key developmental stages. Although we observed significant effects of AMPA on fitness parameters in each group, these effects were not exacerbated in individuals from AMPA-free habitats. We suggest that temporal and/or spatial dynamics of contamination, as well as gene flow between exposed and preserved populations, may hinder adaptive divergence between populations. Yet, we show strong adverse effects of AMPA exposure at early developmental stages. AMPA could therefore be one of the numerous causes of declining wild amphibian populations.
