Background In the past decade, there has been an increase in prescription opioid related harms. These include dependence, non-fatal and fatal overdose. Pharmacists play a an important role in safe opioid supply. As most opioids are supplied through pharmacies, pharmacists are in a prime position to reduce harms associated with opioid use. Development of specific core competencies for pharmacists may facilitate consistent and safer opioid supply. Objective To reach consensus on which competency items identified by the Association of Faculties of Pharmacy of Canada's Opioid Working Group are considered core competencies for Australian pharmacists in opioid supply and assess expert pharmacists' perceptions of how well these competencies are currently met by practicing pharmacists. Setting Expert pharmacists in the area of opioid supply from across Australia. Method A series of questionnaires were presented to Australian opioid expert pharmacists via a modified Delphi study, with the aim to reach consensus on whibe considered core competencies for Australian pharmacists in opioid supply. Given the large number of items identified, further research may help determine priorities for training and education.Cystic echinococcosis (CE) is a disease of livestock and humans caused by the larval stage of the Echinococcus granulosus sensu lato (s.l.) species complex and occurs especially in areas where sheep breeding is widespread. This survey evaluates, after more than a decade since the last epidemiological survey, the current situation and trend of CE in sheep of the highly endemic island of Sardinia (Italy). From 2012 to 2018, out of a total of 1416 sheep, 65.3% were found positive for CE. Cyst fertility has increased slightly from 10.3% in 2006 (Scala et al. 2006) to 11.7% in the present survey (χ2 = 1.07; P = 0.30018) with a peak of 14.0% in the province of Sassari. Overall, 6815 hydatids were found and classified as sterile (26.2%), fertile (14.4%), caseous (5.3%), or calcified (54.1%) based on their morphology. All isolates were identified as E. granulosus sensu stricto (former G1/G3, strains). The objective of this work is to study the effect of host sex and age class, and the annual variation on the prevalence and mean abundance of Antarctophthirus lobodontis in crabeater seals from the Antarctic Peninsula. During three consecutive field-seasons, we collected lice from 41 crabeater seals (23 females, 16 males, 2 indeterminate, being 24 adults, and 17 juveniles). We investigated this effect on the prevalence and mean abundance by a generalized linear model formulation in a Bayesian framework. According to the lowest Deviance Index Criterion model, sex host does not affect prevalence nor mean abundance. We found that juveniles present greater abundance and prevalence than adults, possibly due to foraging habits. They spent more time on the ice than adults in groups of dozens of animals. This behavior would favor both egg development and lice transmission. We do not find adult females with lice, which suggests that transmission of A. lobodontis should be horizontal. The high mean abundance of lice in 2014 could be associated with an unusual increase in Lobodon carcinophaga population, probably related to the pack-ice availability and zooplankton abundance.This is the first study showing an in vivo microautophagy upregulation by Leishmania infantum in dogs. Both Leishmania amastigotes and promastigotes were detected in the cytoplasm of many professional and nonprofessional phagocytic cells of popliteal lymph node of three dogs suffering from chronic cutaneous leishmaniasis. Ultrastructurally, parasites appeared to be wrapped by lysosomes and/or multivesicular bodies. Neither phagophores nor double-membraned vacuoles consistent with autophagosomes were observed. Transcription factor EB (TFEB), a key factor involved in lysosome biogenesis, showed a statistically significant increase in the total component when examined by western blot in samples from leishmaniotic dogs compared with samples from healthy dogs. Instead, phosphorylated TFEB showed unmodified expression levels both in leishmaniotic and healthy dogs. Furthermore, Hsc70 and endosomal sorting complex required for transport (ESCRT)-I, which are known to play a role in microautophagy, showed no variation in expression levels both in diseased and healthy animals. Vps4A/B, an evolutionary conserved ATPase responsible for ESCRT-I complex disassembly and MVB maturation, was statistically significantly overexpressed in lymph nodal samples from leishmaniotic dogs. Bag3 was downregulated in diseased dogs whereas CHIP, p62, and LC3-II did not show any variation in expression levels. The altered expression profile of Bag3 suggested an altered interaction of Bag3 with Hsc70 and CHIP, which usually form a molecular complex involved in autophagosome-lysosome pathways. Ultrastructural and molecular findings suggested that the microautophagy pathway is upregulated in lymph nodes of dogs suffering from a chronic natural infection by Leishmania infantum.To enhance specific or total sophorolipids (SLs) production by Starmerella bombicola for specific application, mutant library consisting of 106 mutants from 7 batches was constructed via atmospheric and room-temperature plasma (ARTP). When compared to the wild strain, 11, 36 and 12 mutants performed increases over 30% in lactonic, acidic or total SLs production. Genetic stability investigation showed that 8, 7, and 4 mutants could maintain the improved SLs production capacity. Mutants of A6-9 and A2-8 were selected out for enhanced specific SLs and total SLs production in fed-batch cultivation in flask. Without optimization, A6-9 obtained the highest reported lactonic SLs production of 51.95 g/l and A2-8 performed comparable acidic and total SLs production of 68.75 g/l and 100.33 g/l with all the reported stains. The structural composition of the obtained SLs was analyzed by HPLC and LC/MS, and the results confirmed the enhancement of SLs and certain SL components. These mutants would be important in industrial applications because the production and purification costs of SLs could be greatly reduced. Besides, the acquisition of these mutants also provided materials for the investigation of regulation mechanism of SLs biosynthesis for further genetic engineering of S. bombicola. Furthermore, critical micelle concentration (CMC), minimum surface tension (STmin) and hydrophilic-lipophilic balance (HLB) of the SLs obtained from the wild and mutant strains were also examined and compared. These results demonstrated the feasibility of obtaining SLs with different properties from different strains and the high efficiency of mutation breeding of S. bombicola by ARTP.A purification system was constructed with the N-segment of the Npu DnaE split intein as an affinity ligand immobilized onto an epoxy-activated medium and the C-segment used as the cleavable tag fusing target protein. The affinity properties of C-tagged proteins adsorbed on IN affinity chromatography medium were studied with GFP as a model target protein. The saturated adsorption capacity and dynamic adsorption capacity reached 51.9-21.0 mg mL-1, respectively. With this system, two model proteins, GFP and alcohol dehydrogenase (ADH), has been successfully taglessly purified with regulation of Zn2+ and DTT. The yield, purification factor and purity of purified tagless GFP reached 39, 11.7 and 97%, respectively; while these values for purified tagless ADH were 38.2, 6.8 and 91%, respectively. These results showed that the system for Npu DnaE split intein-mediated affinity adsorption and in situ cleavage is a potential platform for recombinant protein production.In this work, white wine lees (WWL), cheese whey (CW), and glycerol (GLY) were used as carbon (C) sources to mixotrophically support the production of the microalga Nannochloropsis salina, replacing CO2 supply. In doing so, the alga was allowed to grow on C sources dosed at 2 g L-1, 3 g L-1, and 4 g L-1 of C, in the presence and absence of CO2 supply. WWL and CW were not able to support the algal growth due to a fungal contamination that was genomically identified, while GLY gave interesting results in particular with 3 g L-1 of C. GLY-C was able to replace CO2-C completely when the latter was omitted, showing an algal biomass production similar to those obtained in autotrophy. If CO2-C was provided jointly with GLY-C, biomass production and lipid contents increased more than 30% and 23%, respectively, compared to autotrophy.Background Attention is usually given to inferior steepening on corneal topography in the evaluation of a patient's suitability for LASIK surgery. The aim of this study is to investigate long-term refractive results with superior steepening. Methods Patients who underwent LASIK surgery between 2015 and 2019 in our refractive surgery department were retrospectively reviewed. The patients with a ≥ 1.0 D superior-inferior (S-I) quadrant difference in the tangential map, using a Scheimpflug camera with a Placido disc topographer (Sirius), were included in the study. Preoperative and postoperative best-corrected and uncorrected visual acuity (Snellen), cylindrical refraction values, and spherical equivalent (SE) values were compared. Adverse events were recorded. Results Fifty eyes of 28 patients participated in the study. The mean age of the patients was 27.5 ± 8.0 (19-59). Sixteen patients were female (57.1%), and 12 (42.8%) patients were male. The average follow-up time was 29.8 ± 11.1 months (12-61). Average central corneal thickness was 549.4 ± 26.0 (498-602) μm. Average minimal corneal thickness was 549.1 ± 26.9 (497-598) μm. Preoperative S-I quadrant difference (D) was 1.87 ± 0.7 (1.0-3.99). Posterior elevation (Kvb) was 11.2 ± 1.9 (9-17) μm. The preoperative SE value was - 1.7 ± 2.1 (- 6.25-3.25) and improved to - 0.3 ± 0.44 D (- 1.25-0.75) (p less then 0.001). Preoperative cylindrical refraction values were - 2.04 ± 1.7 (- 6.25-0), and postoperative values were - 0.47 ± 0.4 (- 2-0) D (p less then 0.001). Uncorrected visual acuity was median 1.0 (0.4-1.0) with 38 eyes (76%) having 20/20 postoperative uncorrected visual acuity. No sight threatening complications or ectasia findings were observed during the 2 years postoperative follow-up time. Conclusions Abnormal corneal topographies with (S-I) asymmetry result in predictable results after LASIK.
