V.Immunohistochemical localisation of indoleamine 2,3-dioxygenase was studied in order to better understand the pathophysiology of placenta accreta spectrum. In the decidua staining for indoleamine 2,3-dioxygenase was found in the glandular epithelium with some additional positive cells. Extravillous cytotrophoblast invasion was present in the myometrium which was not covered by the decidual tissue whereas myometrial invasion of cytotrophoblasts was absent where this tissue lay deep to decidua. These results suggest that indoleamine 2,3-dioxygenase expression in the decidua may normally control trophoblast invasion and absence of its expression where decidua is absent may be involved in the pathogenesis of the over-invaded placenta. The free radicals play an important role to understand direct/indirect transformation mechanisms of organic pollutants. However, very few efforts have been made to elucidate the radicals produced by direct photolysis. In this study, the short-lived radicals generated under simulated sunlight irradiation from representative halogenated phenols (HPs), monobromophenols, were investigated by electron paramagnetic resonance (EPR). The results showed that three radicals, carbon-centered radical (C), hydrogen radical (H) and hydroxyl radical (OH), were generated from the direct irradiation of HPs. Compared to other substitutions, halogenated atom at para-position led to the highest production of these radicals which is in accordance with the energies calculated by density functional theory. Based on the analyses of the reactive species and corresponding intermediate adducts, the possible reaction pathways for these radicals were tentatively proposed. Dissolved organic matters (DOM) could enhance the photodegradation of HPs by directly affecting the radicals' formation, mainly due to generation of excited triplet DOM (3DOM*). A positive correlation was found between the concentrations of hydrated electron and the steady state 3DOM* from different DOM. Our findings provided insights into environmental photochemical fate of HPs through their direct photolysis and will help more accurately understand their phototransformation mechanisms in the environment. Histamine, one of the most toxic and commonly encountered biogenic amines (BA) in food, is produced by the microbial decarboxylation of histidine. It may accumulate at high concentrations in fish and fermented food. Cheese has some of the highest histamine concentrations, the result of the histidine-decarboxylase activity of certain lactic acid bacteria (LAB). The present work describes the nucleotide sequence and transcriptional organization of the gene cluster responsible for histamine biosynthesis (the HDC cluster) in Lactobacillus vaginalis IPLA 11064 isolated from cheese. The influence of histidine availability and pH on histamine production and the expression of the HDC cluster genes is also examined. As expected, the results suggest that the production of histamine under acidic conditions improves cell survival by maintaining the cytosol at an appropriate pH. However, the transcriptional regulation of the HDC cluster is quite different from that described in other dairy histamine-producing LAB, probably due to the lack of a termination-antitermination system in the histidyl-tRNA synthetase gene (hisS). Micronutrients provide a potentially interesting alternative to fungicides for the protection of crops against fungal pathogens. Here we studied the effect of foliar-applied manganese (Mn) in the form of MnSO4 on severity of anthracnose disease, caused by Colletotrichum lagenarium in cucumber (Cucumis sativus L.) plant. The study was done aimed to characterize the optimum dose and application time of Mn fertilizer on disease suppression as well as to identify the defense mechanisms by which Mn-treated plants resist to fungal disease. In preliminary tests, Mn was applied at different concentrations (1.8, 4.5 and 7.2 mM) and various time points (three days before or two hours before inoculation, or three days after inoculation). Results showed that application of Mn either before or after inoculation suppressed the fungal infection in leaves and cotyledons, with a higher efficiency when applied three days prior to inoculation. However, all applied concentrations of Mn equally reduced the disease severity. Mn treatment in the absence of the pathogen promoted lignification and reactive oxygen species (ROS) accumulation. Also, pre-inoculation Mn treatment enhanced pathogen-induced lignification, callose or ROS production and reduced pathogen-induced cell death. The increase of lignin, callose and ROS induction by Mn application were 34, 30 and 31 % compared to control, respectively. Together, the results suggested the effectiveness of Mn treatments on anthracnose alleviation in cucumber plants. The findings here have a practical importance in plant physiology studies to identify the resistance-relevant mechanisms to pathogens and in sustainable agriculture to control the fungal diseases by a safe method. BACKGROUND AND OBJECTIVE Vascular access is the "lifeline" of end-stage renal disease patients, which is surgically constructed to remove blood-waste and return artificially filtered blood into circulation. The arteriovenous shunting causes an abrupt change in blood flow and results in increased fluidic stress, which predisposes to access stenosis and thrombosis. While access flow is crucial to evaluate interventional endpoint, application to measure flow using digital angiogram is not yet available. The goal of this study was to determine the feasibility of flow quantification in dialysis access using a software tool and to guide the design of an imaging protocol. METHODS 173 digital subtraction angiographic (DSA) images were retrospectively analyzed to evaluate access flow in a custom-programming environment. Four bolus transit time algorithms and a distance calculation method were assessed for flow computation. Gamma variate function was applied to remove secondary flow and intensity outliers in the bolus te. V.BACKGROUND Transcranial B-mode sonography (TCS) can detect hyperechogenic speckles in the area of the substantia nigra (SN) in Parkinson's disease (PD). These speckles correlate with iron accumulation in the SN tissue, but an exact volumetric localization in and around the SN is still unknown. Areas of increased iron content in brain tissue can be detected in vivo with magnetic resonance imaging, using quantitative susceptibility mapping (QSM). METHODS In this work, we i) acquire, co-register and transform TCS and QSM imaging from a cohort of 23 PD patients and 27 healthy control subjects into a normalized atlas template space and ii) analyze and compare the 3D spatial distributions of iron accumulation in the midbrain, as detected by a signal increase (TCS+ and QSM+) in both modalities. RESULTS We achieved sufficiently accurate intra-modal target registration errors (TRE less then 1 mm) for all MRI volumes and multi-modal TCS-MRI co-localization (TRE less then 4 mm) for 66.7% of TCS scans. In the caudal part of the midbrain, enlarged TCS+ and QSM+ areas were located within the SN pars compacta in PD patients in comparison to healthy controls. More cranially, overlapping TCS+ and QSM+ areas in PD subjects were found in the area of the ventral tegmental area (VTA). CONCLUSION Our findings are concordant with several QSM-based studies on iron-related alterations in the area SN pars compacta. They substantiate that TCS+ is an indicator of iron accumulation in Parkinson's disease within and in the vicinity of the SN. Furthermore, they are in favor of an involvement of the VTA and thereby the mesolimbic system in Parkinson's disease. Rehabilitation of heavy metals contaminated soils using association between legumes and beneficial rhizospheric microorganisms such as plant growth-promoting bacteria (PGPR) is a major challenge in agronomy. The present study focuses on assessing the impact of field inoculation with I1 (Rhizobium leguminosarum (M5) + Bacillus simplex + Luteibacter sp. + Variovorax sp.) and I5 (R. leguminosarum (M5) + Pseudomonas fluorescens (K23) + Luteibacter sp. + Variovorax sp.) on growth and phytoremediation potential of Lathyrus sativus plants as well as soil quality and fertility. The experimentation was carried out in mine tailings of northern Tunisia. Obtained Results indicated that the in situ inoculation with I1 and I5 significantly increased the shoots (47% and 22%) and roots dry weights (22% and 29%), as well as nodules number (48% and 31%), respectively, compared to uninoculated plants. The maximum Pb accumulation in the above-ground tissue was recorded in plants inoculated with I5 (1180.85 mg kg-1 DW). At the same time, we noticed a reduction in total Pb and Cd in the rhizosphere of inoculated plots mainly in those inoculated with I5 reaching 46% and 61%, respectively, compared to uninoculated plots. Likewise, I5 inoculum significantly enhanced soil total nitrogen (35%) and available phosphorus (100%), as well as β-glucosidase (16%), urease (32%) and alkaline phosphatase (12%) activities. Here we demonstrate the usefulness of L. sativus inoculated with I5 inoculum formed by mixing efficient and heavy metals resistant PGPR to boost an efficient reclamation of Cd and Pb contaminated soils and, ultimately, to improve their quality and fertility. Eight labdane diterpenoids, including two new labdane diterpenoids, named forsyshiyanins A-B (2-3), along with six known ones (1, 4-8), were isolated from the fruits of Forsythia suspensa. The new structures including their absolute configurations were elucidated by extensive spectroscopic analyses, X-ray diffraction and computational calculation. In vitro, eight labdane diterpenoids showed anti-inflammatory activities, with the inhibition rates of release of β-glucuronidase from polymorphonuclear leukocytes of rats being in the range 46.8-51.0% at concentrations of 10 µM, as well as anti-viral activities against influenza A (H1N1) virus and respiratory syncytial virus (RSV), with the IC50 values in the range 18.4-26.2 μM and EC50 values in the range 10.5-14.4 μM, respectively. INTRODUCTION The optimal management of elderly patients (pts) with Hodgkin's lymphoma is not yet defined. click here The aims of the present study were 1) to evaluate clinical and laboratory characteristics of elderly pts; 2) to indentify risk factors for unfavorable outcome. PATIENTS AND METHODS The outcome of 182 pts ≥ 60 years (y) was retrospectively analyzed (median age, 67y). Mixed cellularity histology was diagnosed in 49.5 %, advanced stage of disease was in 68.7 % pts, CIRS > 3 in 35.7 %, ECOG PS ≥ 2 in 22.9 % (60-69y) of pts. Chemotherapy (CMT) alone was used in 69.2 % and combination of CMT and radiotherapy in 26.9 % of pts. Anthracycline-based CMT received 83.5 % of pts. The median follow-up was 4.5y. RESULTS The overall response/complete remission rate was 85.6/70.7 %. The median progression free survival (PFS) and overall survival (OS) were 10y and 11.3y, respectively. Estimated 5-y PFS and 5-y OS were 65.7 % (in contrast to 98.2 % in pts 3, lymphopenia less then 8 % and anthracycline-free chemotherapy appear to be significant for unfavorable outcome.
