Cementless tibial components have shown improvements in clinical performance compared to predicate designs, though evidence supporting mid-term performance and fixation is scarce. The purpose of this study is to determine the mid-term survivorships, revision rates, and reasons why 3-dimensional printed cementless tibial baseplates (3DTKAs) failed compared to other cementless as well as cemented tibial baseplates reported from the American Joint Replacement Registry (AJRR) data.

All primary total knee arthroplasty (TKA) cases performed in patients 65 years of age or older within the AJRR from January 2, 2012 through June 30, 2020 were queried. A total of 28,631 3DTKAs were identified from 428 institutions. These were compared to all other "aggregated cementless tibia" (n=7,577) and "aggregated cemented tibia" (n= 550,133) cases. Centers for Medicare & Medicaid Services data over the same time period were merged with AJRR data to determine survivorship and patient-timed incident revision rates per 1,000 years. Failure reasons were tracked during this study period.

At 60 months, Kaplan-Meier implant survivorship was 98.9% (CI 98.7-99.0), 98.3% (CI 97.9-98.6), and 98.4% (CI 98.4-98.5) in the 3DTKA, aggregate cementless, and cemented knee cohorts, respectively (P< .0001). Patient-timed incident revision rates were 3.11 (CI 2.75-3.53), 3.99 (CI 3.34-4.76), and 3.35 (CI3.28-3.42) for those cohorts, which corresponds to a revision rate of 0.31%, 0.40%, and 0.34% per year.

Quercetin (QT) is the most plentiful flavonoid in vegetables and fruits, with antioxidant activities. This study aimed to evaluate the protective role of QT on testicular toxicity induced by COV. Twenty-four adult male Wistar rats were randomly divided into four groups (n = 6) including control, quercetin (QT) (50 mg/kg), crude oil vapors (COV), and COV + QT. The inhalation method was used to expose the rats to crude oil vapors for 5 h daily, and QT was administered orally. After 30 days, the rats were euthanized, then, the testes were removed for gonadosomatic index (GSI), sperm parameters, H&E staining, the activity of the antioxidant enzymes, and apoptotic gene expression assessments. The COV statistically significantly (P less then 0.05) reduced GSI, sperm count, motility, viability, and sperm normal morphology, histological indexes, and antioxidant enzyme activities than control. Also, COV statistically significantly (P less then 0.05) increased the expression of caspase-3, p-53, and Bax genes and decreased Bcl-2 gene expression. Co-administration of QT + COV caused a statistically significant (P less then 0.05) decrease in Bax gene expression and increased antioxidant enzyme activities, Bcl-2 gene expression, and reproductive parameters than the COV group. ALA is also reported to promote cytochrome c oxidase (COX) activity, which can generate ROS itself. Therefore, this study focused on the effect of ALA during PDT. In addition, in the previous study, sodium ferrous citrate (SFC) is reported to increase COX activity. So, this study also aims to improve the COX activity by the addition of SFC that can promote ROS generation, which has a cytotoxic effect.

However, the underlying mechanisms are not fully understood and require further investigation. In the present study, we established a PFOS-exposed Sprague-Dawley (SD) rat kidney injury model by intraperitoneal injection of PFOS (1 mg/kg and 10 mg/kg body weight) every alternate day for 15 days and cytotoxicity models of normal rat kidney epithelial (NRK52E) and human renal proximal tubular (HK2) cells exposed to PFOS (20 μM and 60 μM) for 24 h to reveal the mechanisms underlying PFOS-induced nephrotoxicity. Data showed that PFOS increased the kidney index and induced nephrotoxicity in rats. Furthermore, PFOS significantly increased malondialdehyde (MDA) levels, decreased GSH peroxidase (GSH-PX) activity in kidney tissues, and increased intracellular reactive oxygen species (ROS) levels in NRK52E and HK2 cells. Following PFOS treatment, mitochondrial damage in the renal tubular epithelial cells of rats was observed and the mitochondrial membrane potential (ΔΨm) was decreased in NRK52E cells. PFOS upregulated apoptosis of tubular epithelial cells and expression of Connexin 43 (Cx43) in vitro and in vivo. The Cx43 inhibitor gap26 attenuated the apoptosis of tubular epithelial cells. In conclusion, our findings reveal that PFOS may trigger renal tubular cell apoptosis through oxidative stress and upregulation of Cx43, resulting in PFOS-induced nephrotoxicity.

Cell counting kit-8 (CCK8) and lactate dehydrogenase (LDH) assays revealed time- and dose-dependent toxicity of DZN against mouse hippocampus-derived HT22 cells. Acetylcholinesterase (AChE) activity assay suggested that DZN inhibited the AChE activity, and TUNEL staining revealed that DZN increased the apoptotic rate.
