Using transgenic mouse models, we previously showed that abrogation of radiation-induced senescence, or apoptosis, can partially rescue neurogenesis in the subventricular and hippocampus regions. Here, we evaluate whether the injection of recombinant epidermal growth factor (rEGF) or mesenchymal stromal cells (MSC) engineered to secrete EGF (MSC-EGF) can preserve neurogenesis. Using doublecortin (Dcx) expression and BrdU incorporation assays, we found that the injection of rEGF into the subventricular zone (SVZ) promotes neurogenesis, despite increasing apoptosis, in the brain of irradiated mice. The effect of rEGF was mostly localized, as Dcx expression was not induced in the hippocampus region and limited in the contralateral SVZ. Surprisingly, the injection of bone marrow-derived MSC alone, or secreting EGF, did not result in increased neurogenesis despite the fact that part of the MSC survived a few weeks after injection. Our results suggest that only a supraphysiological concentration of rEGF can promote neurogenesis, likely through a direct mitogenic effect.Carbon monoxide (CO) poisoning presents an interesting challenge for postmortem toxicology laboratories. A comparable alternative that is quick and simple is analysis using a standard UV-Vis spectrophotometer which offers sufficient precision and accuracy for the measurement of % carboxyhemoglobin (COHb) in postmortem blood. The hurdle for laboratories then becomes the selection of an appropriate spectrophotometric method since a variety of procedures have been published over the years. Four methods were evaluated based on literature findings and/or the appropriateness for postmortem testing. These methods are based on 1) relating %COHb to the ratio of COHb and hemoglobin (Hb) absorbance of a two-component system (two wavelength), 2) multicomponent analysis of all hemoglobin species, 3) multicomponent analysis of a two-component system, and 4) derivative spectroscopy. While all four methods performed similarly in terms of typical validation requirements, the ability to more effectively handle decomposed samples and the ease of sample preparation afforded 3) multicomponent analysis of a two-component system the most suitable for routine postmortem testing.A flexible method to image unmodified transcripts and transcription in vivo would be a valuable tool to understand the regulation and dynamics of transcription. Here, we present a novel approach to follow native transcription, with fluorescence microscopy, in live C. elegans. By using the fluorescently tagged Argonaute protein NRDE-3, programmed by exposure to defined dsRNA to bind to nascent transcripts of the gene of interest, we demonstrate transcript labelling of multiple genes, at the transcription site and in the cytoplasm. This flexible approach does not require genetic manipulation, and can be easily scaled up by relying on whole-genome dsRNA libraries. Cell therapy is a potential treatment for radiation-induced skin injury. The stromal vascular fraction (SVF) is a newer material in stem cell therapy that is made up of stem cells harvested from adipose tissue, which has been shown to promote the healing of refractory wounds of different causes. In this study, SVF was isolated from patients with radiation-induced skin injury. Adipose-derived stem cells (ADSCs) accounted for approximately 10% of the SVF by flow cytometry. Compared with the control group of rats, administration with SVF attenuated the skin injury induced by electron beam radiation. The effect of SVF on the human skin fibroblast microenvironment was determined by proteomic profiling of secreted proteins in SVF-co-cultured human skin fibroblast WS1 cells. Results revealed 293 upregulated and 1,481 downregulated proteins in the supernatant of SVF-co-cultured WS1 cells. WS1 co-culture with SVF induced secretion of multiple proteins including collagen and MMP-1. In the clinic, five patients with radiation-induced skin injury were recruited to receive SVF transfer-based therapy, either alone or combined with flap transplantation. Autogenous SVF was isolated and introduced into a multi-needle precision electronic injection device, which automatically and aseptically distributed the SVF to the exact layer of the wound in an accurate amount. After SVF transfer, wound healing clearly improved and pain was significantly relieved. The patients' skin showed satisfactory texture and shape with no further wound recurrence. Our findings suggest that transplantation of SVF could be an effective countermeasure against severe radiation-induced skin injury.
Sequence similarity is the most frequently used procedure in biological research, as proved by the widely used BLAST program. The consecutive seed used by BLAST can be dramatically improved by considering multiple spaced seeds. Finding the best seeds is a hard problem and much effort went into developing heuristic algorithms and software for designing highly sensitive spaced seeds.

We introduce a new algorithm and software, ALeS, that produces more sensitive seeds than the current state-of-the-art programs, as shown by extensive testing. We also accurately estimate the sensitivity of a seed, enabling its computation for arbitrary seeds.

The quantification of long-range correlation of electroencephalogram (EEG) signals is an important research direction for its relevance in helping understanding the brain activity. Epileptic seizures have been studied in the past years where different non-linear statistical approaches have been employed to understand the relationship between the EEG signal and the epileptic discharge. One of the most widely used method for to analyse long memory processes is the detrended fluctuation analysis (DFA). However, no objective and pragmatic methods have been developed to detect crossover points and reference channels in DFA.

In this article, we propose (i) two automatic approaches that successfully detect crossover points in DFA related methods on the log-log plot and (ii) a criteria to choose the reference channel for the log-amplitude function. Moreover, the DFA is applied to EEG signals of 10 epileptic patients collected from the CHB-MIT database, being the log-amplitude function used to compare the different brain hemispheres by making use of the methodology proposed in the article. The existence of long-range power-law correlations is demonstrated and indicates that the EEG signals of epileptic patients present three well-defined regions with the first region showing a 1/f noise (pink noise) for seven subjects and a random walk behaviour for three subjects. The second and third regions show anti-persistence behaviour. Moreover, the results of the log-amplitude function were divided in two groups the first, including seven subjects, where the increase in the scales results in an increase in the fluctuation in the frontal channels and the second, included three subjects, where the fluctuation for large scales are greater for the parietal channels.

Herein, we report the crystal structure of a quaternary complex of AcrIIA14 bound SauCas9-sgRNA-dsDNA at 2.22 Å resolution, revealing the molecular basis for AcrIIA14 recognition and inhibition. Our structural and biochemical data analysis suggest that AcrIIA14 binds to a non-conserved region of SauCas9 HNH domain that is distinctly different from AcrIIC1 and AcrIIC3, with no significant effect on sgRNA or dsDNA binding. Further, our structural data shows that the allostery of the HNH domain close to the substrate DNA is sterically prevented by AcrIIA14 binding. In addition, the binding of AcrIIA14 triggers the conformational allostery of the HNH domain and the L1 linker within the SauCas9, driving them to make new interactions with the target-guide heteroduplex, enhancing the inhibitory ability of AcrIIA14. Our research both expands the current understanding of anti-CRISPRs and provides additional culues for the rational use of the CRISPR-Cas system in genome editing and gene regulation.Urea recycling occurs in all mammalian species and represents an important source of ruminal nitrogen (N) for ruminants fed protein-restricted diets. However, its importance for cattle fed adequate amounts of protein and energy remains unclear. Six Nellore feedlot steers fed concentrate-based diets were used in a 6 × 6 Latin square design with a 3 × 2 factorial arrangement of treatments to evaluate ruminal fermentation, urea kinetics, and N excretion. Treatments consisted of 3 protein sources (PS soybean meal plus urea [SU], corn gluten meal [CGM], and dry distillers grains [DDG]) and 2 inclusion levels (PL; 11 and 14%). Steers were adapted to the diets for 14 d followed by 8 d of sample collection. Feed intake, fecal output, and urine production were measured from d 18 to d 22 of each period. Blood samples were collected every 6 h on d 18. [ 15N- 15N]-urea was infused into the jugular vein for 82 h over d 19 to d 22, and measurement of 15N in background (d 18) and enriched feces and urine (d 21) were used toundegradable protein (RUP) feed sources (CGM or DDG) compared to diets with SU markedly increased NUE, while maintaining microbial protein (MICP) synthesis. Results from this study suggest that the equation adopted by NASEM (2016) was not accurate in estimating the urea-N used for anabolism (UUA) in Nellore feedlot cattle fed concentrate-based diets.
