Despite the decades-long use of HCQ in the treatment of diseases, such as malaria and autoimmune disorders, the exact mechanisms of action of this drug are only beginning to be understood. To date, no data are available on the genotoxic potential of HCQ in vitro or in vivo. The present study is the first investigation of the DNA damaging- and mutagenic effects of HCQ in mammalian cells in vitro, at concentrations that are comparable to clinically achievable doses in patient populations. We demonstrate significant induction of a representative oxidative DNA damage (8-oxodG) in primary mouse embryonic fibroblasts (MEFs) treated with HCQ at 5 and 25 μM concentrations (P = 0.020 and P = 0.029, respectively), as determined by enzyme-linked immunosorbent assay. Furthermore, we show significant mutagenicity of HCQ, manifest as 2.2- and 1.8-fold increases in relative cII mutant frequency in primary and spontaneously immortalized Big Blue® MEFs, respectively, treated with 25 μM dose of this drug (P = 0.005 and P = 0.012, respectively). The observed genotoxic effects of HCQ in vitro, achievable at clinically relevant doses, are novel and important, and may have significant implications for safety monitoring in patient populations. The ingestion of this mycotoxin represents a risk for both human and animal health. The development of F. verticillioides is associated with environmental conditions, especially carbon sources. We developed a reliable and fast reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay and determined fum1 gene expression upon growth of two F. verticillioides strains isolated from maize and wheat in Czapek's medium containing four different sugars as sole carbon sources. Fumonisin B1 (FB1) production was determined by LC-MS/MS analysis. High growth and production of FB1 were observed in fructose-containing medium for the strain that originated from maize. Less production of FB1 occurred using maltose as sole carbon source for both strains. The fum1 gene expression started between 2 and 4 days of incubation, and positive signals were detected prior to the initial production of FB1. The RT-LAMP assay was effective in the detection of fum1 gene expression at very early stages of F. verticillioides growth and allowed the prediction of FB1 formation.

There are many methods of early detection of IHD which can help prevent death, but few are able to determine the configuration and severity of this disease. Our study aims to determine the severity and configuration of ischemic zones by implementing the reaction-diffusion analysis of cardiac excitation in a model of the left ventricle of the human heart. Initially, this model is applied to compute twenty thousand in-silico ECG signals with stochastic distribution of ischemic parameters. Furthermore, generated data is effectively (r2=0.85) implemented for training a one-dimensional convolutional neural network to determine the severity and configuration of ischemia using only two lead surface ECG. Our results readily demonstrate that using a minimally configured portable ECG system can be instrumental for monitoring IHD and allowing early tracking of acute ischemic events.

T-helper (Th)1 cells, a process we have shown may be mitigated by long-chain (LC) ω-3 polyunsaturated fatty acids (PUFAs). Our objective was to study pivotal mediators of interactions between Th1 cells and adipocytes as potential mechanisms underlying the antiinflammatory effects of LC ω-3 PUFAs.

Using an in vitro model, 3T3-L1 adipocytes were cocultured with purified splenic CD4
T cells from C57BL/6 mice consuming one of two isocaloric high-fat (HF) diets (60% kcal fat), containing either 41.2% kcal from lard+18.7% kcal from corn oil (control, HF) or 41.2% kcal from lard+13.4% kcal from corn oil+5.3% kcal from fish oil (HF+FO). Cocultures were stimulated for 48 h with lipopolysaccharide (10 ng/mL).

Compared with HF cocultures, HF+FO reduced Th1-cell markers (including secreted interferon-γ) and increased Th2-cell markers, consistent with reduced expression of genes related to major histocompatibility complex II (P < 0.05). HF+FO also blunted markers of priming and activity of the NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome (P < 0.05). In confirmatory work, 3T3-L1 adipocyte pretreatment with the LC ω-3 PUFA docosahexaenoic acid (100 μM, 24 h) blunted interferon-γ-induced (5 ng/mL, 24 h) expression of genes related to major histocompatibility complex II and priming and activity markers of the NLRP3 inflammasome compared with control (P < 0.05).
