Donor surgery was mainly done by robotic-assisted laparoscopy, involving dissections of the utero-vaginal fossa, arteries, and ureters. The last part of surgery was by laparotomy. Recipient laparotomy included vascular anastomoses to the external laparoscopy in uterus transplantation and is thereby a proof-of-concept of use of minimal invasive surgery in this new type of transplantation. Evolving evidence indicates that circular RNA Tau tubulin kinase 2 (circ-TTBK2) is a cancer-associated gene. Therefore, this study was to explore the potential role of circ-TTBK2. MATERIALS AND METHODS Levels of circ-TTBK2, microRNA (miR)-761, and integrin subunit beta 8 (ITGB8) were determined by adopting quantitative real-time polymerase chain reaction (qRT-PCR) or Western blot. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was performed to detect cell viability, and the invaded cells were distinguished utilizing transwell assay. Iron and lipid reactive oxygen species (ROS) assays were implemented to examine the iron (total iron and ferrous iron) and lipid-based ROS in glioma cells, respectively. Besides, dual-luciferase reporter assay was administrated to illustrate the interaction between miR-761 and circ-TTBK2 or ITGB8. The role of circ-TTBK2 was identified via xenograft tumor model. RESULTS Levels of circ-TTBK2 and ITGB8 were upregulated, whereas miR-761 level was low-expressed in glioma tissues and cells. Circ-TTBK2 was a sponge of miR-761 to modulate ITGB8. Additionally, circ-TTBK2 knockdown or miR-761 increase could retard cell proliferation, invasion, and promote ferroptosis in glioma cells. Interestingly, miR-761 inhibitor could abolish the repressive impact of circ-TTBK2 silencing on cell growth in vitro. Also, the influence of miR-761 mimic on cell phenotypes was regained after ITGB8 upregulation. Meanwhile, circ-TTBK2 deficiency caused the decrease of tumor growth. CONCLUSIONS Circ-TTBK2 regulated cell proliferation, invasion and ferroptosis via targeting ITGB8 by sponging miR-761 in glioma, providing a promising biomarker for the clinical therapy of human glioma.An outbreak of a novel coronavirus disease-19 (nCoV-19) infection began in December 2019 in Wuhan, China, and now involved the whole word. Hence, biliary dysbiosis may be a primary trigger in the pathogenesis of biliary diseases. In particular, recent studies have suggested that microorganisms could play a significant role in the development of gallstones, pathogenesis of autoimmune cholangiopathies and biliary carcinogenesis. Moreover, the intimate connection between the biliary tract, liver and pancreas, could reveal hidden influences on the development of diseases of these organs. Further studies are needed to deepen the comprehension of the influence of the biliary microbiota in human pathology. This knowledge could lead to the formulation of strategies for modulating the biliary microbiota in order to treat and prevent these pathological conditions.OBJECTIVE Phosphorylation of insulin receptor substrate (IRS) 1 by tumor necrosis factor alpha (TNF-α) has been implicated as a factor contributing to insulin resistance. Administration of IL-15 reduces adipose tissue deposition in young rats and stimulates secretion of adiponectin, an insulin sensitizing hormone that inhibits the production and activity of TNF-α. We aimed at investigating the effects of age life-long moderate calorie restriction (CR) on IL-15 and TNF-α signaling in rat white adipose tissue (WAT). MATERIALS AND METHODS Thirty-six 8-month-old, 18-month-old, and 29-month-old male Fischer344´Brown Norway F1 rats (6 per group) were either fed ad libitum (AL) or calorie restricted by 40%. The serum levels of IL-15 and IL-15 receptor α-chain (IL-15Rα) were increased by CR controls regardless of age. An opposite pattern was detected in WAT. In addition, CR reduced gene expression of TNF-α and cytosolic IRS1 serine phosphorylation in WAT, independently from age. RESULTS IL-15 signaling in WAT is increased over the course of aging in AL rats compared with CR rodents. Protein levels of IL-15Rα are greater in WAT of AL than in CR rats independently from age. This adaptation was paralleled by increased IRS1 phosphorylation through TNF-α-mediated insulin resistance. Adiponectin decreased at old age in AL rats, while no changes were evident in CR rats across age groups. CONCLUSIONS IL-15 signaling could therefore represent a potential target for interventions to counteract metabolic alterations and the deterioration of body composition during aging.OBJECTIVE Recent studies indicated that long non-coding RNA is involved in the formation of atherosclerosis, which is the pathological basis of coronary heart disease. Here, we reported the function and regulatory mechanism of RMRP in coronary atherosclerosis. MATERIALS AND METHODS qPCR was used to investigate the expression of IL-6, IL-8, RMRP, and miR-128-1-5P in coronary atherosclerosis and human vascular smooth muscle cells. Luciferase reporter assay confirmed the direct target effect of RMRP with miR-128-1-5P and miR-128-1-5P with Gadd45g on HEK293T. Western blot was used to detect protein expression in coronary atherosclerosis and human vascular smooth muscle cells. RESULTS RMRP expression and Gadd45g protein level were up-regulated in coronary atherosclerosis and human vascular smooth muscle cells, while miR-128-1-5P was down-regulated. RMRP downregulation remarkably inhibited the expression of IL-6, IL-8, and apoptosis related protein in human vascular smooth muscle cells after ox-LDL treatment. CONCLUSIONS Liraglutide could curb the expression of inflammatory cytokines and apoptosis related protein in coronary atherosclerosis by regulating RMRP/miR-128-1-5P/Gadd45g signaling pathway, providing a new potential strategy for the treatment of coronary atherosclerosis.OBJECTIVE Uric acid is considered a biomarker for cardiovascular risk. Only a few studies have investigated the effect of aspirin on serum uric acid (SUA) levels with contradictory results. The present study evaluated the effect of aspirin on SUA levels in Chinese individuals over 60 years of age. PATIENTS AND METHODS Subjects over 60 with coronary artery disease or multiple cardiovascular risk factors were enrolled in a multicentre randomized clinical trial. Eligible subjects were randomized to receive 50 mg or 100 mg aspirin daily. Levels of arachidonic acid-induced platelet aggregation performed by light transmission aggregometry (LTA-AA) and SUA were measured at randomization and two weeks thereafter. In this subanalysis, subjects without aspirin use prior to enrolment were chosen. RESULTS A total of 446 subjects were analysed, of which 151 subjects took 50 mg aspirin, and 295 took 100 mg aspirin. Hyperuricaemia was present in 23.3% (104/446) of subjects at baseline. LTA-AA levels were significantly reduced in subjects after taking aspirin for two weeks (both 50 mg and 100 mg, p 0.05), while slightly decreased in hyperuricaemic subjects (429 μmol/L vs. 392 μmol/L, p less then 0.001). CONCLUSIONS Our study showed that both 50 mg and 100 mg aspirin significantly inhibited platelet aggregation. Aspirin treatment for two weeks showed no hyperuricaemic effect in people over 60. SUA levels were unchanged after taking aspirin in normouricaemic subjects but decreased in hyperuricaemic subjects. This trial was registered at www. chictr.org.cn as ChiCTR1800018517.OBJECTIVE Acute uncomplicated diverticulitis is an important clinical condition usually managed in clinical practice with antibiotic therapies and hospitalization in ward. In this setting, recent papers and guidelines suggest to limit the use of antibiotics in selected cases and encourage an early discharge in low-risk patients. The purpose of this retrospective study is to identify serological inflammatory markers and CT findings of acute uncomplicated diverticulitis (AUD) at the onset of the disease and the correlation with the need for in-patient or out-patient management. PATIENTS AND METHODS It was used a database drawn from the collection of the patients admitted to our Emergency Room from January 2016 to 2019 and undergoing urgent abdominal CT-scan for suspicious of acute diverticulitis. For each patient we considered biochemical and radiological parameters at the onset of the disease and if patients were managed as in-patients (hospitalization in ward) or as out-patient (early discharged or after observation in Short Stay Unit). RESULTS Among patients with early diagnosis of AUD, 108 (65%) were hospitalized in ward with mean time of in-stay of 6.94 days, while only 58 (35%) patients with same diagnosis were managed as out-patient and early discharged from emergency room or after observation in short stay unit with a mean time of in-stay significantly shorter (3.39 days, p-value 0.0007). Higher levels of C reactive protein and the length of colon involved considered as percentage (%) in comparison with the entire colon were significantly related to the need for hospitalization (p-value 0.03). CONCLUSIONS Biochemical parameters and a more advanced radiological evaluation, as the length (%) of colon involved, could allow a stratification of patients with diagnosis of AUD at the admission and help physicians in the early management.
