We propose a comparison method that uses the 3D models of a, computed tomography-generated (CT-generated), preoperative plan and a postoperative CT to obtain the discrepancy in the six possible degrees of freedom. We ran a prospective study (level 2 evidence) of 30 patients undergoing uncemented THA to quantify the discrepancy between planned and achieved femoral stem orientation and position. The discrepancy was low for femoral stem vertical position and leg length, and varus-valgus and anterior-posterior orientation. The discrepancy was higher for femoral version with a mean (±SD) of -1.5 ± 7.8 deg. Surgeons should be aware of the variability of the eventual position of uncemented stems in THA and acknowledge the risk of achieving a less-than-optimal femoral version, different from the preoperative 3D CT plan. © 2020 Orthopaedic Research Society. Published by Wiley Periodicals, Inc.BACKGROUND Lactic acid in the stratum corneum contributes to skin flexibility, making it a useful indicator of skin conditions. It is this useful to devise a simple technique to measure the lactic acid in the stratum corneum. To this aim, a printed chip biosensor was developed to analyze lactic acid in tape stripped (TS) stratum corneum samples. MATERIALS AND METHODS Lactic acid was extracted from TS stratum corneum samples. A normal lactic acid sensor was prepared by applying lactate oxidase (LOD) to a printed chip. Another lactic acid sensor was prepared using LOD and a mediator osmium polymer immobilized on a printed chip. The amount of lactic acid in the extracted solutions was quantified using either the prepared biosensors or an existing analysis method. RESULTS The results measured using the normal lactic acid sensor show low correlation with the results measured using an existing analytical method as a comparison, but those of the mediator osmium lactic acid sensor show high correlation. CONCLUSIONS The amount of lactic acid in samples extracted from the stratum corneum using a simple TS technique can be simply analyzed with high accuracy using a printed chip biosensor. © 2020 John Wiley & Sons A/S. Published by John Wiley & Sons Ltd.Subcutaneous injection of azacitidine (AZA) is an important treatment option for myelodysplastic syndrome (MDS), which improves overall survival. In hematology, the incidence of AZA-induced cutaneous adverse events (AE) has been known to be relatively high, which has not been well recognized by dermatologists. Discontinuation of AZA can result in the deterioration of MDS disease activity. Therefore, on dermatological consultation, precise evaluation of AE severity and careful consideration is required for post-AE medication management. To enhance our understanding of AZA-induced cutaneous AE, we report four cases with two representative cutaneous AE subtypes and summarize the clinicopathological phenotypes and courses of the cases in the published work. Case 1, a 71-year-old man, developed neutrophilic dermatosis involving the dermis and subcutaneous tissue. The other three cases, a 75-year-old man, a 78-year-old woman and a 68-year-old man, presented injection-site erythema associated with flare-up reaction. Discontinuation of AZA was necessary for case 1 alone. The published work review delineated three major subtypes of AZA-induced cutaneous AE systemic cutaneous reaction, neutrophilic dermatosis type and erythematous type injection-site reaction. Histologically, the first two subtypes are mostly characterized by neutrophil infiltration, while the third subtype presents lymphocytic cell infiltration. Neither AZA discontinuation nor intensive interventions were required for the erythematous type injection-site reaction, while AZA termination or systemic treatments, represented by corticosteroid administration, were preferentially conducted for the systemic cutaneous reaction or the neutrophilic dermatosis type injection-site reaction subgroup. These observations support the necessity of subtype-dependent treatment strategies for the management of AZA-induced cutaneous AE. © 2020 Japanese Dermatological Association.RATIONALE Ambient mass spectrometry (AMS) techniques are very required in forensic chemistry to evaluate evidence with less analytical interference, high confidence, and accuracy. However, traditional methodologies, such as paper spray ionization (PSI), have been shown to present low sensitivity in the analysis of illicit drugs from biological matrices. METHODS Fiber spray ionization mass spectrometry (FSI-MS) was developed using a capillary polypropylene (PP) hollow fiber. Seized samples of drugs, i.e., a tablet, blotter paper, hashish, and cocaine powder, were analyzed. Cocaine was quantified from whole urine by dipping the fiber directly into the solution. FSI-MS was tested for the analysis of a sample of urine obtained from a drug abuse suspect. RESULTS The FSI(+) analysis showed the detection of different types of synthetic drugs in tablet and blotter paper samples, e.g., amphetamine, cathinones, phenethylamines, and opioids, while pure cocaine and different types of coca alkaloids were identified from csample, which makes this a very promising AMS method and relevant to forensic chemistry. However, a little number of miRNAs have been reported in the muscle development of Chinese native pig breeds. In this study, the longissimus dorsi transcripts of Chinese native Rongchang pig at weaning and slaughter time points were analysed for miRNA-seq. The results showed that 19 novel and 186 known miRNAs involved in the Rongchang pig skeletal muscle development were identified. Based on these findings, we further confirmed that porcine miR-127, miR-299 and miR-432-5p were obviously down-expressed in adult pig (287 days of age), while miR-7134-3p and 664-5p were significantly up-expressed in weaning pig (35 days of age). In other words, these miRNAs could be the potential molecular markers and play vital roles in the muscle development process. Moreover, we found miR-127 could inhibit the proliferation and myogenesis of porcine satellite cells in longissimus dorsi muscle. Our findings will provide deep insight into miRNA function for pork quality research with Chinese indigenous pig breeds. © 2020 Blackwell Verlag GmbH.The objective was to evaluate the effect of inclusion of 2.5% and 5% ovine serum, enriched with vitamin E (Vit E) and fish oil (FO), in human sperm freezing medium. Serum samples were prepared from sixteen rams (n = 4) feeding on a without supplemented diet, and diets supplemented with Vit E, FO and Vit E + FO. Semen samples, from 60 normozoospermic men, were frozen in (I) a commercial freezing medium (SpermFreeze™; control medium), (II) the commercial freezing medium containing foetal bovine serum, (III) the commercial freezing medium + nonenriched serum (serum group), (IV) the commercial freezing medium + Vit E enriched serum (Vit E group), (V) the commercial freezing medium + FO enriched serum (FO group) and (VI) the commercial freezing medium + Vit E + FO enriched serum (Vit E + FO group). Sperm total and progressive motility, morphology, viability and plasma membrane integrity were significantly higher (p ≤ .05) in Vit E and Vit E + FO groups compared with the control group. Mitochondrial membrane potential did not differ between treatments (p > .05). It was concluded that ovine serum enriched with vitamin E and vitamin E + FO improved the quality of human spermatozoa but enriched serum containing FO could not improve the sperm cryo-injuries. © 2020 Blackwell Verlag GmbH.BACKGROUND Abomasitis is a syndrome affecting young milk-fed calves. The current veterinary literature describes mainly its necropsy findings. OBJECTIVES To describe the clinical presentation, complementary tests, treatments, and case-fatality rate of calves with a clinical diagnosis of abomasitis and to identify potential factors associated with outcome. METHODS Observational retrospective cohort study (2006-2016). Review of the medical records of calves less then 3 months of age presented with abdominal and abomasal distension for less then 7 days that were clinically diagnosed with abomasitis at the Faculty of Veterinary Medicine of the Université de Montréal. A follow-up examination was conducted by telephone interview. ANIMALS Twenty-three calves clinically diagnosed with abomasitis. RESULTS Median age of presentation was 3 days (range, 0-62 days). The typical duration of the clinical course was less then 24 hours (15/23). On admission, the 2 most common clinical signs were anorexia (13/14) and positive succussion (13/14). Hyper-l-lactatemia (15/16) and increased γ-glutamyl-transferase activity (13/14) were the most common laboratory abnormalities. Hypoproteinemia (19/22) and a left shift (15/18) of the neutrophils also were observed. The short-term case-fatality rate was 52% (12/23). The clinical diagnosis was confirmed on all necropsied calves. Clostridium spp. and Escherichia coli were the most frequently isolated bacteria. Based on univariate statistical analysis, the surviving calves were significantly (P  less then  .05) less hypothermic, less acidemic, less hyper-l-lactatemic, and had lower serum creatinine concentrations on admission than did the deceased calves. CONCLUSIONS AND CLINICAL IMPORTANCE In our study, abomasitis was associated with a guarded prognosis. © 2020 The Authors. However, the functional role of TNXB and causal variants had not been interrogated in that study. In the present study, we explored the effects of TNXB expression in the development of ESCC and searched for functional variants in this gene. We found TNXB was downregulated in ESCC tumors. Using small interfering RNAs and CRISPR-Cas9 methods, we identified that both knockdown and knockout of TNXB significantly promoted ESCC cell growth in vitro, suggesting a tumor suppressor role of this gene in ESCC. Through further fine-mapping analysis, we identified that a noncoding variant in the promoter of TNXB, rs411337, predisposed to ESCC risk (odds ratio = 1.36, 95% confidence interval 1.22-1.51, P = 9.10 × 10-9 ). These findings revealed the functional mechanism of TNXB in the development of ESCC and may contribute to the prevention and treatment of this disease in the future.
