Median age was 38 years (range 16-82), 73% were male, and 43% had stage II. With a median follow-up of 10.5 years for all patients, 5-year progression free survival (PFS) and overall survival (OS) were 93% and 97%, respectively, with no difference by treatment era (PFS p=0.13; OS p=0.35). For the 49 patients that had a PET2 scan 86% were PET-negative and 14% were PET-positive by Deauville criteria with a 5-year PFS of 92% and 80% (p=0.70), respectively. This represents the largest experience of a PET-adapted approach in NLPHL and supports that ABVD alone may be a viable option in select patients with a PET2-negative scan, with consideration of acute and long-term toxicities.Tyrosine kinase inhibitors (TKI) have dramatically changed the survival of chronic myeloid leukemia (CML) patients and treatment-free remission (TFR) has recently merged as a new goal of CML treatment. The aim of this work was to develop recommendations for TKI discontinuation in Latin America (LA), outside clinical trials. A working group of CML experts from LA discussed 22 questions regarding TFR and reached a consensus for TFR recommendations in the region. TFR is indicated in patients in first CP, with typical BCR-ABL transcripts, under TKI treatment for a minimum of 5 years, in sustained deep molecular response (DMR MR4.5) for 2 years. Sustained DMR must be demonstrated on at least 4 IS qPCR tests, separated by at least 3 months, in the immediate prior 2 years. After 2nd line therapy, TFR is indicated in previously intolerant patients, not resistant. Molecular monitoring is recommended monthly the first 6 months, every 2-3 months from months 7 to 12, and every 3 months during the second year, indefinitely. Exclusion of antibody carriers from the donor pool has decreased the number of cases significantly, but TRALI remains leading cause of transfusion-related morbidity and mortality in industrialized countries. Here, we demonstrate that proteins released from donor cells during processing of blood components are capable of inducing a new type of reverse TRALI when transfused to pre-immunized recipients. First, we show that soluble neutrophil surface protein CD177 in complex with proteinase 3 (sCD177/PR3) is not only present in human plasma, but also in packed red blood cell (PRBC) supernatant. Filtration or storage enhances the concentration of sCD177/PR3 in PRBCs. Second, we demonstrate that sCD177/PR3 specifically binds to PECAM-1 on stimulated (but not on unstimulated) endothelial cells (EC). Third, we provide evidence that the sCD177/PR3/PECAM-1 complex is functional. In the presence of monoclonal or human antibodies against CD177 or PR3, ECs produce reactive oxygen species and become apoptotic. Albumin flux through an EC monolayer increases significantly whenever antibodies and the cognate antigens are present. Finally, we present a clinical case in which anti-CD177 present in a transfusion recipient precipitated TRALI after the transfusion of CD177 positive, but not negative, PRBCs. In conclusion, we introduce a new TRALI mechanism based on the specific binding of transfused, soluble antigens to activated endothelial cells in pre-immunized recipients. We suggest that further studies and clinical work-up of TRALI should also include antibody investigation of the recipient.Fibrinogen γ' accounts for 3% to 40% of plasma fibrinogen. Earlier studies indicated that fibrinogen γ' forms altered fibrin clots under static conditions, whereas clinically, altered plasma γ' levels are associated with arterial and venous thrombosis. However, the effects of static vs flow conditions on the role of γ' throughout the pathophysiological range is unknown. This study explores the effects of γ' levels on clot formation and structure in static and flow conditions. Coagulation of plasma samples with low (n = 41; 3%), normal (n = 45; 10%), or high (n = 33; 30%) γ' levels were compared with that of purified fibrinogen mixtures with increasing ratios of γ' (3%, 10%, 30%). Clots were analyzed by confocal microscopy, permeation, turbidity, and lysis techniques. In a novel 2-step flow-perfusion model, fibrinogen-deficient plasma repleted with increasing ratios of γ' (3%, 10%, 30%) or plasmas with low (n = 5, 3%) or high (n = 5, 30%) γ' were flowed over preformed platelet aggregates at arterial (500 s-1) and venous (150 s-1) shear rates. Increasing γ' percentages within the pathophysiological range (3%-30%) did not result in any change in clot-formation rates; however, it led to significantly higher clot density, thinner fibers, and slower lysis in static conditions. Under flow at arterial shear, high γ' (30%) led to faster (+44.1%-75.3%) and increased (+104%-123%) fibrin deposition, with clots exhibiting a larger volume (+253%-655%) and height (+130%-146%). These trends were magnified at venous shear. Overall, our findings demonstrate the significant impact of pathophysiological fibrinogen γ' levels on clot structure and provide new flow-dependent mechanisms to explain how γ' increases thrombosis risk.The Halo Effect is a widely studied phenomenon that interests multiple disciplines. Moreover, previous works reported that individuals' attachment styles affected their emotional responses to the pandemic. Individuals' attachment styles may influence the magnitude of change of the Halo. Here we investigate how priming (Social Distancing or Contact with others) affects the strength of the Halo Effect, with respect to individuals' attachment styles. Participants (N = 298) rated the Aesthetics and Perceived Trustworthiness of strangers' faces (N = 96) presented twice, before and after the presentation of a prime. Results revealed that individuals' attachment styles affect the strength of the Halo. However, we found no evidence supporting the fact that different attachment styles lead to differences in the magnitude of changes after priming. Results help shed light on how attachment styles influence individuals' impression formation.The COVID-19 pandemic disrupted health systems and economies throughout the world during 2020 and was particularly devastating for the United States, which experienced the highest numbers of reported cases and deaths during 20201-3. Many of the epidemiological features responsible for observed rates of morbidity and mortality have been reported4-8; however, the overall burden and characteristics of COVID-19 in the United States have not been comprehensively quantified. Here we use a data-driven model-inference approach to simulate the pandemic at county-scale in the United States during 2020 and estimate critical, time-varying epidemiological properties underpinning the dynamics of the virus. The pandemic in the US during 2020 was characterized by national ascertainment rates that increased from 11.3% (95% credible interval (CI)8.3 - 15.9%) during March to 24.5% (18.6 - 32.3%) during December. Population susceptibility at year's end was 69.0% (63.6 - 75.4%), indicating that roughly one third of the US population had been infected. Community infectious rates, the percentage of people harbouring a contagious infection, rose above 0.8% (0.6 - 1.0%) before the end of the year, and were as high as 2.4% in some major metropolitan areas. In contrast, the infection fatality rate fell to 0.3% by year's end.
To study perforating scleral vessels (PSV) in patients with high myopia with swept-source optical coherence tomography (SS-OCT) and to determine their relationship with myopic choroidal neovascularization (mCNV) and its activity.

Retrospective analysis of patients with high myopia (≥ -6 D or ≥ 26 mm of axial length) using multimodal imaging. The presence of PSVs and mCNV was assessed using SS-OCT images (TRITON, Topcon Corporation, Japan).

564 eyes from 297 highly myopic patients were studied. 155 eyes showed signs of mCNV (27.5 %) while PSVs were found in 500 eyes (88.6%). PSVs were found in 93.5% (145/155) of eyes with mCNV and they were under or in contact with the mCNV in 80.6 % (117/145). The mean number of intravitreal injections (IVI) received by mCNV patients was 4.06±4.17 along 66.9±4.1 months of follow-up. The number of injections per year was 1.32±1.56, the mean number of relapses was 1.11±1.83 and the mean number of relapses/year was 0.25±0.41.
