In the present study, we aimed to investigate the effects of pH and sodium chloride (NaCl) concentration on the structure of β-casein (β-CN) purified from buffalo milk using circular dichroism (CD), intrinsic tryptophan, and anilino-8-naphthalene sulfonate (ANS) fluorescence spectroscopy. We found that NaCl concentration played a critical role in the stability of the secondary structure of β-CN. The CD negative peak had a redshift as the NaCl concentration was increased and accompanied by a decrease of β-sheet content and an increase of α-helix content. ANS fluorescence spectroscopy also indicated that higher NaCl concentration and lower pH significantly affected the tertiary structure of β-CN. Dynamic light scattering (DLS) results showed that the particle size of buffalo β-CN had a blueshift, and then a redshift within the pH range of 5.0-7.5, and it showed a redshift when the NaCl concentration was increased.Frequent outbreaks of food quality and safety problems have seriously damaged the interests of consumers and reduced their confidence in China's food safety. In this study based on market supply and demand, we design a differential game model between food supplier and food retailer by considering different decision-making situations. We also analyze the optimal revenue of the food supplier and food retailer on food quality efforts, the overall return of the supply chain, the level of food quality and safety, and their evolutionary characteristics. Results of the analysis indicate the following. (a) From the situation of decentralized decision-making to the situation of decision-making under the incentive strategy, a Pareto improvement occurs in the food quality and safety strategy of food supplier, food retailer, and even the entire food supply chain. (b) The optimal revenues of the supplier and retailer, overall supply chain revenue, and efforts of the supplier and retailer are all affected by changes in market supply and demand, resulting in drastic fluctuations. On the whole, food quality tends to improve over time and will fluctuate slightly due to changes in market supply and demand. (c) If the market supply is stable when supply exceeds demand and the market demand turns from a downward trend to an upward trend, then food quality safety risk will be higher than in other periods.Paeonia ludlowii, a plant of the Paeoniaceae family, has abundant genetic diversity in different populations, and the seed oil can be used in a diverse number of activities. However, its neuroprotective effect is not clear. We investigated the memory-improving effects and associated mechanisms of Paeonia ludlowii seed oil (PLSO) on amyloid beta (Aβ)25-35-induced Alzheimer's disease (AD) in rats. The Morris water maze test was undertaken, and subsequently, the content of malondialdehyde (MDA), superoxide dismutase (SOD), glutathione (GSH), and acetylcholinesterase (ACHE) in the hippocampus was detected by biochemical analyses. To further study PLSO, we examined the pathologic structure and apoptosis of hippocampal tissue by staining. Immunohistochemical analysis was used to detect expression of IBA-1 and GFAP in the hippocampus. Detection of proinflammatory factors was achieved by reverse transcription-quantitative polymerase chain reaction and Western blotting. High-dose PLSO inhibited expression of GFAP and IBA-1. We demonstrated that high-dose PLSO can regulate activation of glial cells and mediate apoptosis of hippocampal cells, and significantly improve learning and memory deficits in AD rats. PLSO could be developed as a nutritional supplement and sold as a drug for AD prevention and/or treatment.Efficient light-induced floral stimulation plays a key role in energy conservation and maintaining stable productivity during off-season periods of dragon fruit plants. In this study, we first reported on results of a survey on dragon fruit farmers regarding use of lamps in performing artificially induced flowering process in Vietnam. It was found that the use of incandescent lamp was prevalent in dragon fruit cultivation practices, resulting in heavy electricity consumption, and that low-power compact fluorescent light (CFL) bulbs were not extensively utilized, possibly due to low floral induction performance of domestic CFL bulbs. Arguing that emission spectra of currently used lamps were not consistent with adsorption spectra of phytochromes, whose transformation is responsible for flowering process of dragon fruit, we then proposed three improved CFL lamps (power capacity of 20 W) having emission spectra focused on red and far-red regions. New lamp prototypes were tested in 7 field experiments in three different provinces in Vietnam. One improved CFL bulb (treatment 2) performed relatively well in comparison with the incandescent control lamp (60 W) in six out of seven experiments with regard to some growth indicators (e.g., number of floral stems, number of bubs, number of fruits per plant) and fruit yield. Recent success on commercialization of the improved CFL lamp demonstrates the potential of CFL lamps in floral stimulating irradiation of other crops and plants and in alleviating electricity burden in dragon fruit growing areas.Intralipids are widely used to provide energy and necessary fatty acids for the patients. The structure of lipids may affect their function. We developed a bio-catalyzed route to prepare various intralipids and investigated the protective effect of intralipids against α-naphthylisothiocyanate (ANIT) induced liver injury rats, further discussing the structure-function relationship. The middle-long-middle (MLM) structural intralipid was synthesized through alcoholysis-esterification, and the influence factors were investigated. ANIT treatment caused liver injury, further making hepatocyte damage, and increasing related biochemical indexes, like aspartate aminotransferase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), and total bilirubin (TBIL). Especially, MLM-based and structoglyceride (STG) intralipids worked better in the early stage, to reduce the AST, ALT, and TBIL (p less then .05). MLM showed a comparative advantage over other intralipids to accelerate the reduction of ALT (1st day) and AST (3rd day). MLM intralipid might be a promising next-generation intralipid than the current STG intralipid liver-injury patients. The biological catalysis MLM-based intralipids can make the maximum utilization of fatty acids for the liver regeneration, where middle-chain fatty acid (MCFA) in sn-1,3 position can be metabolized directly to provide energy and long-chain fatty acid (LCFA) in sn-2 position can be delivered effectively for cell membrane repairing.Main objectives of this work were to investigate the influences of ultrasound pretreatment (UP) and microwave pretreatment (MP) on extraction kinetics, chemical composition, and antioxidant activity of Kumquat peel essential oil (EO) obtained by hydrodistillation extraction (HDE). The effects of ultrasound power and processing time, and microwave power and processing time were evaluated. As compared with HDE individually, UP and MP decreased the extraction time, increased the yield and DPPH radical-scavenging activity but did not noticeably affect chemical composition of the EO. For UP and MP, the highest EO yield was obtained when the ultrasonic power and processing time, and microwave power and processing time were 210 W and 30 min, 300 W and 6 min, respectively. In comparison with MP, UP gave a higher yield and DPPH radical-scavenging activity of the EO. Overall, UP and MP are promising techniques for HDE of EO from kumquat peel.To study the role of Allicin in regulating Treg/Th17 ratio in splenic lymphocyte by increasing the expression of MEKK2 protein in MAPK signaling pathway, and to explore the mechanism of immune response in mice with collagen-induced arthritis (CIA). Flow cytometry detected Treg/Th17 ratio in splenic lymphocytes. Furthermore, RT-PCR and WB were used to detect the mRNA and protein expression of related transcription factors and inflammatory factors Foxp3, ROR-γt, and IL-17A, as well as MEK2 protein expression in splenic lymphocytes. The results showed that Allicin treatment could reduce the severity of arthritis and the proliferation of collagen fibers on the surface of cartilage and bone joints in CIA mice. Compared with NC group, Treg decreased and Th17 increased in spleen lymphocyte of Model group (p less then .01); after Allicin treatment, Treg increased while Th17 decreased significantly (p less then .01). Meanwhile, MEKK2 protein expression in spleen lymphocyte of Model group decreased compared to that in NC group (p less then .01), and MEK2 protein expression increased significantly after Allicin treatment (p less then .01). To sum up, the present study suggests that MEKK2 protein plays an important role in the pathogenesis of CIA model. In terms of mechanism, Allicin may play a therapeutic role in rheumatoid arthritis (RA) by increasing the expression of MEKK2 protein and affecting Treg/Th17 ratio.The health benefits and promising medical treatment potential of total flavonoids from Eupatorium lindleyanum DC. (TFELDC) have been recognized. The process parameters of extracting total flavonoids from Eupatorium lindleyanum DC. by ultrasonic-microwave synergistic extraction (UMSE) were optimized, and they were purified by AB-8 macroporous resin in the current study. In addition, the antioxidant and enzyme inhibitory activities of the purified TFELDC (PTFELDC) were evaluated. The results showed that the optimal parameters of UMSE were as follows ethanol volume fraction 71.5%, L/S ratio 12.2 ml/g, microwave power 318 W, and extraction time 143 s. After TFELDC were purified by AB-8 macroporous resin, the total flavonoid contents of PTFELDC increased from 208.18 ± 1.60 to 511.19 ± 3.21 mg RE/g FDS. Compared with TFELDC, the content of total flavonoids in PTFELDC was increased by 2.46 times. The antioxidant activities of PTFELDC were assessed using DPPH radical, superoxide anion radical, reducing power, and ferric reducing antioxidant power assays, and the IC50 values were found to be 37.13, 19.62, 81.22, and 24.72 μg/ml, respectively. The enzyme inhibitory activities of PTFELDC were measured using lipase, α-amylase, α-glucosidase, and acetylcholinesterase assays with the IC50 values 1.38, 2.08, 1.63, and 0.58 mg/ml, respectively. By comparing with their positive controls, it was found that PTFELDC had good antioxidant activities, and lipase, α-amylase, and α-glucosidase inhibitory activities, However, the acetylcholinesterase inhibitory activity was relatively weaker. These results suggested that PTFELDC have a promising potential as natural antioxidant, antilipidemic, and hypoglycemic drugs used in functional foods or pharmaceuticals.
Embedded implementation research (IR) can play a critical role in health systems strengthening by tackling systems and implementation bottlenecks of a program. To achieve this aim, with the financial support of GAVI, the Vaccine Alliance, in 2016, the Government of Pakistan, UNICEF and the Alliance for Health Policy and Systems Research (AHPSR) launched an Embedded IR for Immunisation Initiative (the Initiative) to explore health systems and implementation bottlenecks, and potential strategies to tackle such bottlenecks in the Expanded Programme on Immunisation (EPI) in Pakistan. In total, 10 research teams were involved in the Initiative, which was the first of its kind in the country. In this paper, we provided a brief overview of the Initiative's approach as well as the key learnings including challenges and successes of the research teams which could inform future embedded IR Initiatives.

Data were collected from members of the IR teams through an online survey. In addition, in-depth interviews were conducted via phone and in-person from IR team members to explore further the challenges they faced while conducting IR in Pakistan and recommendations for future IR initiatives.
