To select the most promising protocol for creating highly immunogenic mRNA vaccines, we performed a comparative analysis of mRNA modifications aimed at increasing its translational activity and decreasing toxicity. We used mRNA encoding a green fluorescent protein (GFP) as a model. Eight mRNA-GFP variants with different modifications (M0-M7) were obtained using the classic cap(1), its chemical analog ARCA (anti-reverse cap analog), pseudouridine (Ψ), N6-methyladenosine (m6A), and 5-methylcytosine (m5C) in different ratios. Modifications M2, M6, and M7, which provided the most intensive fluorescence of transfected HEK293FT cells were used for template synthesis when mRNA encoded influenza immunogens AgH1, AgH3, and AgM2. Virus specific antibodies were registered in groups of animals immunized with a mix of mRNAs encoding AgH1, AgH3, and AgM2, which contained either ARCA (with inclusions of 100% Ψ and 20% m6A (M6)) or a classic cap(1) (with 100% substitution of U with Ψ (M7)). M6 modification was the least toxic when compared with other mRNA variants. M6 and M7 RNA modifications can therefore be considered as promising protocols for designing mRNA vaccines.A novel microwave annealing system and a specific processing condition are proposed for the pre-oxidation of carbon fiber. Rupture of silicone gel-filled breast implants usually is asymptomatic and is one of the more dangerous complications due to free silicone migration. The aim of our study was to evaluate the diagnostic value of ultrasound (US) in the evaluation of the integrity of silicone breast implants and identify the main sign of intact and ruptured breast implants. Patients and Methods In this retrospective study, the medical documentation of women who underwent breast implant surgery and US checkups at Tautrimas Aštrauskas Clinic in Kaunas, Lithuania, during 2015-2020 was analyzed. The patients were divided into two groups patients with intact and ruptured breast implants. The accuracy, sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV) as well as the signs of implant integrity of US examination were evaluated. Based on the findings, we recommend performing US examination after breast augmentation surgery with silicone gel-filled implants annually.As an additive to improve the performance of asphalt binder, tire pyrolysis carbon black is gradually being used in road engineering, but the effect of carbon black (CB) with different particle sizes on asphalt modification remains to be further studied. In this study, three kinds of particle sizes and three kinds of contents of CB were used to modify asphalt, and different tests were conducted to research the high temperature performance and fatigue resistance of carbon black modified asphalt binder. It is found that the addition of CB can enhance the rutting resistance and medium temperature fatigue performance of virgin asphalt binder in general. However, for CB of 270 μm and 2.6 μm, its addition under certain contents lead to the decrease of high temperature performance and fatigue performance of the asphalt binder. For aged asphalt, the addition of CB decreases the rutting resistance and improves the fatigue resistance. The recommended content and particle size of CB are 2% and 2.6 μm. This study refines the complex effects of CB on asphalt properties, providing a reference for determining the size and content of CB in asphalt modification.Multidrug resistant bacteria create a challenging situation for society to treat infections. Multidrug resistance (MDR) is the reason for biofilm bacteria to cause chronic infection. Plant-based nanoparticles could be an alternative solution as potential drug candidates against these MDR bacteria, as many plants are well known for their antimicrobial activity against pathogenic microorganisms. Spondias mombin is a traditional plant which has already been used for medicinal purposes as every part of this plant has been proven to have its own medicinal values. In this research, the S. mombin extract was used to synthesise AgNPs. The synthesized AgNPs were characterized and further tested for their antibacterial, reactive oxygen species and cytotoxicity properties. The characterization results showed the synthesized AgNPs to be between 8 to 50 nm with -11.52 of zeta potential value. The existence of the silver element in the AgNPs was confirmed with the peaks obtained in the EDX spectrometry. Significant antibacterial activity was observed against selected biofilm-forming pathogenic bacteria. The cytotoxicity study with A. salina revealed the LC50 of synthesized AgNPs was at 0.81 mg/mL. Based on the ROS quantification, it was suggested that the ROS production, due to the interaction of AgNP with different bacterial cells, causes structural changes of the cell. This proves that the synthesized AgNPs could be an effective drug against multidrug resistant bacteria.Hepatic resection has been widely accepted as the first choice for the treatment of colorectal metastases. Radiation-induced double-strand breaks (DSBs) are known to be toxic lesions if not repaired correctly. Non-homologous end joining (NHEJ) is the major DSB-repair pathway available throughout the cell cycle and, recently, has been hypothesised to consist of a fast and slow component in G0/G1. The slow component has been shown to be resection-dependent, requiring the nuclease Artemis to function. However, the pathway is not yet fully understood. This study compares two hypothesised models, simulating the action of individual repair proteins on DSB ends in a step-by-step manner, enabling the modelling of both wild-type and protein-deficient cell systems. Performance is benchmarked against experimental data from 21 cell lines and 18 radiation qualities. A model where resection-dependent and independent pathways are entirely separated can only reproduce experimental repair kinetics with additional restraints on end motion and protein recruitment. However, a model where the pathways are entwined was found to effectively fit without needing additional mechanisms. It has been shown that DaMaRiS is a useful tool when analysing the connections between resection-dependent and independent NHEJ repair pathways and robustly matches with experimental results from several sources.Colorectal cancer (CRC) is still one of the most frequent forms of cancer in the world in terms of incidence. Around 40% of CRC patients carry a mutation of the Kirsten rat sarcoma (KRAS) gene, while 10% have a mutation in the B-Raf proto-oncogene serine/threonine kinase (BRAF) gene. These mutations are responsible for dysregulation of the mitogen-associated protein kinase (MAPK) pathway, leading to the proliferation, differentiation, angiogenesis, and resistance to apoptosis of cells. Activation of the MAPK pathway results in adaptive therapeutic resistance, rendering EGFR inhibitors ineffective. This review aims to highlight the recent findings that have improved our understanding of KRAS and BRAF mutations in colorectal cancer and to describe new targeted therapies, used alone or in combination.Nanocomposites containing palladium nanoparticles were synthesized by in situ generation route from palladium acetate and a polyether block amide matrix with the aim to obtain materials with specific nanoparticle location and function properties. The chosen Pebax matrix was composed of a continuous soft phase containing dispersed semi-crystalline rigid domains. Nanocomposite films with Pd amount up to 30 wt% (corresponding to 3.5 vol%) were directly prepared from the palladium precursor and the copolymer matrix through a solvent cast process. The microstructure of the films was investigated by microcalorimetry, X-ray diffraction analyses and transmission electron microscopy. The nanocomposites' function properties in terms of electrical conductivity and interaction towards hydrogen were studied as a function of the palladium content. It was shown that the spherical crystalline Pd nanoparticles that were in situ formed were located in the continuous soft phase of the copolymer matrix. They did not induce modification of Pebax microstructure and chain mobility. The specific location of the metal nanoparticles within the copolymer matrix associated with their low size allowed obtaining conductive materials for Pd amount equal to 3.5 vol%. Moreover, the affinity towards hydrogen evidenced from hydrogen permeation experiments made this nanocomposite series promising for further development in sensing applications.
