Domoic acid (DA) is a marine neurotoxin that accumulates in filtering shellfish during harmful algal blooms. A health protection limit of 20 ppm DA in razor clams (RC) has been set based principally upon an episode of acute DA toxicity in humans that included Amnesic Shellfish Poisoning among survivors. The objective of this study was to determine the dose-response relationship between estimated DA exposure through RC consumption and memory loss in Washington state Native Americans from 2005 to 2015. Results from total learning recall (TLR) memory scores were compared before and after the highest DA exposures. A decrease in TLR was related to DA dose (p less then 0.01) regardless whether the effect was assumed to be transient or lasting, and whether the dose was expressed as an average daily dose or an average dose per meal. Benchmark dose modeling identified BMDL10 values of 167 ng/kg-day and 2740 ng/kg-meal assuming a transient effect, and 196 ng/kg-day and 2980 ng/kg-meal assuming no recovery of function occurs. These DA dose thresholds for a measurable memory function reduction observed in this study of clam consumers are well below the safe acute dose underpinning the current regulatory DA limit of 20 ppm (ca. 60 μg/kg).We conducted a me-too validation study to confirm the reproducibility, reliability, and predictive capacity of KeraSkin™ skin irritation test (SIT) as a me-too method of OECD TG 439. With 20 reference chemicals, within-laboratory reproducibility (WLR) of KeraSkin™ SIT in the decision of irritant or non-irritant was 100%, 100%, and 95% while between-laboratory reproducibility (BLR) was 100%, which met the criteria of performance standard (PS, WLR≥90%, BLR≥80%). WLR and BLR were further confirmed with intra-class correlation (ICC, coefficients >0.950). WLR and BLR in raw data (viability) were also shown with a scatter plot and Bland-Altman plot. Comparison with existing VRMs with Bland-Altman plot, ICC and kappa statistics confirmed the compatibility of KeraSkin™ SIT with OECD TG 439. The predictive capacity of KeraSkin™ SIT was estimated with 20 reference chemicals (the sensitivity of 98.9%, the specificity of 70%, and the accuracy of 84.4%) and additional 46 chemicals (for 66 chemicals [20 + 46 chemicals, the sensitivity, specificity and accuracy 95.2%, 82.2% and 86.4%]). The receiver operating characteristic (ROC) analysis suggested a potential improvement of the predictive capacity, especially sensitivity, when changing cut-off (50% → 60-75%). Collectively, the me-too validation study demonstrated that KeraSkin™ SIT can be a new me-too method for OECD TG 439.4-Methylimidazole (4MEI) is a contaminant in food and consumer products. Pulmonary toxicity and carcinogenicity following chronic dietary exposures to 4MEI is a regulatory concern based on previous rodent studies. This study examined acute pulmonary toxicity in B6C3F1 mice from 6 h to 5 days after oral gavage with a single dose of 150 mg/kg 4MEI, a double dose delivered 6 h apart, or vehicle controls. Oral gavage of 150 mg/kg naphthalene, a prototypical Club cell toxicant, was used as a positive control. Intrapulmonary conducting airway cytotoxicity was assessed in fixed-pressure inflated lungs using qualitative histopathology scoring, quantitative morphometric measurement of vacuolated and exfoliating epithelial cells, and immunohistochemistry. 4MEI treatment did not change markers of cytotoxicity including the mass of vacuolated epithelium, the thickness of the epithelium, or the distributions of epithelial proteins secretoglobin 1A1, proliferating cell nuclear antigen, calcitonin gene-related peptide, and myeloperoxidase. 4MEI and vehicle controls caused slight cytotoxicity with rare vacuolization of the epithelium relative to the severe bronchiolar epithelial cell toxicity found in the naphthalene exposed mice at terminal bronchioles, intrapulmonary airways, or airway bifurcations. In summary, 4MEI caused minimal airway epithelial toxicity without characteristic Club Cell toxicity when compared to naphthalene, a canonical Club Cell toxicant.T cell responses to antigen are initiated by engagement of the T cell receptor (TCR)1, leading to activation of diverse signaling cascades, including an incompletely defined pathway that triggers rapid remodeling of the actin cytoskeleton. Defects in the control of actin dynamics and organization are associated with several human immunodeficiency diseases, emphasizing the importance of cytoskeletal remodeling in the functioning of the adaptive immune system. Here, we investigate the role of the adaptor protein Bcl102 in the control of actin dynamics. Although Bcl10 is primarily known as a component of the pathway connecting the TCR to activation of the NF-κB3 transcription factor, a few studies have implicated Bcl10 in antigen receptor-dependent control of actin polymerization and F-actin-dependent functional responses. However, the role of Bcl10 in the regulation of cytoskeletal dynamics remains largely undefined. To investigate the contribution of Bcl10 in the regulation of TCR-dependent cytoskeletal dynamics, we monitored actin dynamics at the immune synapse of primary murine CD8 effector T cells. Quantification of these dynamics reveals two distinct temporal phases distinguished by differences in speed and directionality. Our results indicate that effector CD8 T cells lacking Bcl10 display faster actin flows and more dynamic lamellipodia, compared to wild-type cells. These studies define a role for Bcl10 in TCR-dependent actin dynamics, emphasizing that Bcl10 has important cytoskeleton-directed functions that are likely independent of its role in transmission of NF-κB -activating signals.Rapid transmission and spread of infectious pathogens are enhanced by the agricultural fair environment, where large numbers of livestock and people from numerous backgrounds congregate for several days. The transmission of influenza A virus and zoonotic enteric pathogens to fairgoers is a considerable risk and has occurred several times over the past decade. In an effort to mitigate zoonotic disease transmission in these settings, public health guidelines and recommendations including hand sanitation stations have been implemented. While hand hygiene recommendations to prevent the spread of zoonotic disease are well communicated, it is hypothesized that the adoption of these recommendations by agricultural fairs and fairgoers is low. To test this hypothesis, hand hygiene data collected from 658 agricultural fairs between 2012 and 2019 was analyzed to determine frequency and function of hand sanitation stations at the fairs, as well as utilization of educational signage. Ferrate(VI) (FeVIO42-, FeVI) is a promising oxidant and the removal of pharmaceuticals from wastewater effluents has been investigated in this study. Firstly, FeVI oxidation of selected pharmaceuticals was examined by determining the apparent second-order rate constants (kapp) in buffer solutions as a function of pH (5.0-9.5). At pH 8.0, kapp of cimetidine, famotidine, nalidixic acid, ronidazole, dimetridazole, tinidazole, and caffeine are (1.6 ± 0.2)×103, (7.8 ± 0.3)×102, 2.6 ± 0.4, 1.7 ± 0.1, 0.9 ± 0.3, 0.2 ± 0.1, and less then 0.1 M-1 s-1, respectively. However, kapp could not be directly employed to predict the removal of pharmaceuticals in the effluents due to the inhibited or enhanced effects of effluent organic matters (EfOM). Therefore, an alternative approach of spectroscopic surrogates was investigated since fluorophore was co-degraded with pharmaceuticals in the wastewater effluents. Particularly, the humic-like fluorescent peak correlated well with the pharmaceutical attenuation. The relationship of the reduction of fluorescence and the removal of pharmaceuticals could be described through a universal equation [Formula see text] . The practical utility of the fluorescence surrogate was validated by applying to field samples. Monitoring the changes of the fluorescence surrogate provides a promising, rapid, and inexpensive method for estimating the degradation of pharmaceuticals during FeVI treatment of wastewater effluents.Rare earth elements (REEs) are emerging contaminants due to their large scale of exploitation worldwide for using in the high-technology sector. Diffusive gradients in thin films (DGT) are a good method to measure the bioaccessibility of inorganic substances. This study is the first to evaluate the combined toxicity of REEs in sediments to aquatic biota using the DGT technique. The intertidal zone of the Pearl River Estuary (PRE) was selected as a case study, as the upper reaches of the Pearl River is a major REE reserve and production region. The DGT-labile measured concentrations of REEs (∑REEs) were found to range from 7.02-16.06 μg/L in intertidal surface sediments of the PRE. selleck chemicals Assessment of single REE toxicity found that risk quotient (RQ) values for Y, Pr, Nd, Eu, Dy, Er, and Yb are significantly higher than 1, indicating that the adverse effects of these single REEs should be not ignored. This review provides an overview on the use of flame retardants in plastic manufacturing, the kinetics of their releases from microplastics, the factors affecting their releases, and the potential environmental and ecosystem risk of the released flame retardants. The releases of flame retardants from microplastics typically involve three major steps internal diffusion, mass transfer across the plastic-medium boundary layer, and diffusion in the environmental medium, while the overall mass transfer rate is commonly controlled by diffusion within the plastic matrix. The overall release rates of additive flame retardants from microplastics, which are dependent on the particle's geometry, can often be described by the Fick's Law. The physicochemical properties of flame retardant and plastic matately minimize their risk to the environment and ecosystem.Combining organometallics and biology has generated broad interest from scientists working on applications from in situ drug release to biocatalysis. Engineered enzymes and biohybrid catalysts (also referred to as artificial enzymes) have introduced a wide range of abiotic chemistry into biocatalysis. Predominantly, this work has concentrated on using these catalysts for single step in vitro reactions. However, the promise of using these hybrid catalysts in vivo and combining them with synthetic biology and metabolic engineering is vast. This report will briefly review recent advances in artificial metalloenzyme design, followed by summarising recent studies that have looked at the use of these hybrid catalysts in vivo and in enzymatic cascades, therefore exploring their potential for synthetic biology.
