Seed-growth synthesis is a common strategy to prepare silver nanoplates, whose peculiar plasmonic features can be exploited for surface enhanced Raman scattering (SERS) applications. Here we describe the fabrication and characterization of SERS chips using a peculiar in situ seed growth method, yielding a dense layer of nano-objects directly on a glass slide. In this way, geometric features (i.e. shape and dimensions) of the nano-objects can be tuned by controlling the growth time, obtaining a high concentration of hot spots on the surface. In particular, the SERS response of four kinds of chips were investigated to define the best SERS configuration in terms of size of the silver nano-objects, excitation wavelength and homogeneity of the SERS response. Silver nano-plates with a seeded growth time of 60 min demonstrated remarkable results both in terms of plasmonic enhancement, with an enhancement factor (EF) of 2 × 105 using a 532 nm laser excitation, and good homogeneity of the SERS response with intra- and inter-maps RSD of 10% and 5%, respectively. In order to demonstrate application of these chips for real sample analysis, an analytical procedure for the detection of a model pesticide, i.e. thiram fungicide, was developed and applied to its detection on green apples peels. SERS measurements on 60 min seeded growth silver nano-plates chip coupled with a multivariate PLS approach demonstrated high accuracy and repeatability for thiram detection in food matrix within the European law limits.Cyanotoxins are associated with harmful cyanobacterial blooms, but also exist in biological soil crusts and soils irrigated with cyanobacteria-contaminated water. To achieve an accurate analysis of cyanotoxins in soil, effective extraction, purification and determination methods are imperative. The most challenging aspect is extracting cyanotoxins from soil, due to their tendency to bind strongly to the soil matrix. We used a methanol-ammonium acetate solution to efficiently extract 17 cyanotoxins (microcystins, cylindrospermopsin, anatoxins, anabaenopeptins and cyanopeptolin) from soil. The extract was purified by on-line solid-phase extraction coupled with ultra-high-performance liquid chromatography tandem mass spectrometry. The optimized procedure involved two ultrasonication cycles of 15 min with 4 mL of methanol + 200 mM ammonium acetate, which recovered 60% to >90% of the added cyanotoxins from five soils with diverse organic matter, pH and texture. The method improved extraction by up to 10 times compared to a methanol/water solution. Linearity, accuracy and precision were validated on matrix-mixed soil with surrogate microcystin and cylindrospermopsin internal standards. Limits of detection were 0.001-0.3 ng g-1, depending on the cyanotoxins. The method was used to analyze cyanotoxins in 25 field-collected soils from Quebec, Canada. Out of the 25 soil samples, 11 soils had at least one cyanotoxin, and up to 8 different cyanotoxins were detected in one soil. The sum of all microcystins congeners was from 0.02 to 31 ng microcystins g-1 soil. We also detected anabaenopeptin, the first reported occurrence of this cyanotoxin in soil.Background There is renewed focus on the complement system in the pathogenesis of schizophrenia. In addition to providing aetiological insights, consistently dysregulated complement proteins in serum or plasma may have clinical utility as biomarkers. Vactosertib Methods We performed a systematic literature review searching PubMed, Embase and PsycINFO for studies measuring complement system activity or complement protein concentrations in serum or plasma from patients with schizophrenia compared to controls. Random-effects meta-analyses were performed to calculate pooled effect estimates (Hedges' g standardised mean difference [SMD]) for complement proteins whose concentrations were measured in three or more studies. The review was pre-registered on the PROSPERO database (CRD42018109012). Results Database searching identified 1146 records. Activity of the classical pathway did not differ between cases and controls in four of six studies, and conflicting results were noted in two studies of alternative pathway activity. Twenty studies quantified complement protein concentrations of which complement components 3 (C3) and 4 (C4) were measured in more than three studies. Meta-analyses showed no evidence of significant differences between cases and controls for 11 studies of C3 (SMD 0.04, 95% confidence interval [CI] -0.29-0.36) and 10 studies of C4 (SMD 0.10, 95% CI -0.21-0.41). Conclusions Serological studies provide mixed evidence regarding dysregulation of the complement system in schizophrenia. This was a prospective cohort study that investigated mode of delivery as an independent risk factor for postpartum weight retention at 1 year postpartum. Methods Data from 2500 first-time mothers, aged 18-35 with singleton pregnancies were included in this analysis. Postpartum weight retention was measured at 1 year after delivery and was defined as weight at 1 year compared to weight just before becoming pregnant. Logistic regression models assessed the association between mode of first delivery and subsequent weight retention of 10 pounds or more, adjusting for key confounders including pre-pregnancy body mass index, gestational weight gain, age, education, poverty status, smoking, race/ethnicity, gestational age, pregnancy complications, breastfeeding and exercise habits during pregnancy and in the first year after delivery. Results At 12 months after delivery the women who had delivered by cesarean were more likely to have a postpartum weight retention of 10 pounds or more (27.9%), than those who had delivered vaginally (22.2%), unadjusted OR 1.35 (95% CI 1.11-1.65, p=.003). Even after controlling for confounding variables, mode of delivery remained significantly associated with postpartum weight retention of 10 pounds or more, adjusted OR 1.30 (95% CI 1.04-1.62, p=.02). Conclusions Cesarean delivery at first childbirth is associated with increased risk of weight retention of 10 pounds or more at 1 year postpartum.Introduction The procedural sedation scale of the Niño Jesús Hospital (Madrid) (SSPNJH) has not been validated. Patients and methods A prospective analytical study was conducted in 2 hospitals on patients ≥ 6 months undergoing invasive procedures using sedation-analgesia with propofol or midazolam and fentanyl. All were monitored using the bispectral index (BIS). Videos were made of each procedure, which were then edited and randomised. A total of 150 videos were rated by four observers using the SSPNJH, the sedation scale of the University of Michigan (UMSS), and the Ramsay Scale (SR). These observers were blinded to the BIS, and at the time of drug administration. To assess test-retest reliability, 50 of the initial 150 randomly selected videos were re-assessed. Results The study included a total of 65 patients. The within-observer agreement was high (ρ = 0.793). The SSPNJH gave a good interobserver reliability when compared with the UMSS (ICC = 0.88) and the SR (ICC = 0.86), and there was none with the BIS. Internal consistency was moderate (α = 0.68). Construct validity was demonstrated by changes in scores after administering sedatives (p less then 0.0001). The SSPNJH had a very low correlation with the BIS (r = -0.166), and a moderate correlation with the UMSS (r = 0.497) and the SR (r = 0.405). As regards the applicability, this scale has been used in two hospitals in five different areas by four professionals of different categories. Conclusions The SSPNJH is valid, reliable and applicable for sedation monitoring in invasive procedures under deep sedation-analgesia in paediatric patients. Here we describe a severe case of Bothrops atrox snakebite that, owing to delayed medical assistance, presented with renal and respiratory failure, compartment syndrome, and tissue necrosis. After several fasciotomy surgeries, the patient survived; however, he showed significant functional disability. Few studies have compared the healing and hearing outcomes between gelatin sponge patching and ofloxacin otic solution. Objectives To compare the healing outcomes of large traumatic tympanic membrane perforations treated with gelatin sponge, ofloxacin otic solution, and spontaneous healing. Methods Traumatic tympanic membrane perforations >50% of the entire eardrum were randomly divided into three groups ofloxacin otic solution, gelatin sponge patch and spontaneous healing groups. The healing outcome and hearing gain were compared between the three groups at 6 months. Results A total of 136 patients with large traumatic tympanic membrane perforations were included in analyses. The closure rates were 97.6% (40/41), 87.2% (41/47), and 79.2% (38/48) in the ofloxacin otic solution, gelatin sponge patch, and spontaneous healing groups, respectively (p=0.041). The mean times to closure were 13.12±4.61, 16.47±6.24, and 49.51±18.22 days in these groups, respectively (p less then 0.001). Conclusions Gelatin sponge patch and ofloxacin otic solution may serve as effective and inexpensive treatment strategies for traumatic large tympanic membrane perforations. However, ofloxacin otic solution must be self-applied daily to keep the perforation edge moist, while gelatin sponge patching requires periodic removal and re-patching.
