These findings provide a better understanding of the structural determinants of NMDA receptor antagonism, while also supporting the potential clinical repurposing of these drugs as neuroprotectors for glaucoma and other neurodegenerative diseases.Photodynamic therapy (PDT) is noninvasive, low toxicity, and photo-selective, but may be resisted by malignant cells. A previous study found chlorin e6 (Ce6) mediated PDT showed drug resistance in lung cancer cells (LLC), which may be associated with PDT-induced DNA damage response (DDR). DDR may up-regulate glutathione peroxidase 4 (GPX4), which in turn degrade ROS induced by PDT. However, dihydroartemisinin (DHA) was found to down-regulate GPX4. Accordingly, the DHA was hypothesized to improve the resistance to PDT. The present work explores the mechanism of Ce6 mediated drug resistance and reveals whether DHA can enhance the efficacy of PDT by suppressing GPX4. The in vitro experiments found Ce6 treatment did not inhibit the viability of LLC within 6 h without inducing significant apoptosis, suggesting LLC were resistant to PDT. Further investigation demonstrated PDT could damage DNA and up-regulate GPX4, thus degrading the generated ROS. DHA effectively inhibited the viability of LLC and induced apoptosis. Importantly, DHA displayed a prominent inhibitory effect on the GPX4 expression and thereby triggered ferroptosis. Combining DHA with Ce6 for treatment of LLC resulted in the suppressed GPX4 and elevated ROS. Finally, the findings showed DHA combined with Ce6 exhibited superb anti-lung cancer efficacy. In summary, Ce6 PDT damages DNA, up-regulates GPX4 to degrade ROS, thereby inducing drug resistance. Down-regulation of GPX4 by DHA-triggered ferroptosis significantly enhances the efficacy of PDT. This study provides an outstanding theoretical basis for the regulation of the intratumoral redox system and improving PDT efficacy against lung cancer by herbal monomer DHA.Multiple sclerosis (MS) is an autoimmune disease characterized by inflammatory infiltration in association with demyelination in the central nervous system. Among the factors involved in the immunological mechanisms of MS, Th1, Th17, and Th22 cells play a critical role. In the present study, we investigated the role of CA-074, a potent Cathepsin B inhibitor, in MS progression, using the SJL/J mouse model of experimental autoimmune encephalomyelitis (EAE). Following induction of EAE, mice were administered CA-074 (10 mg/kg) intraperitoneally each day, beginning on day 14 and continuing until day 28, and were evaluated for clinical signs. We further investigated the effect of CA-074 on Th1 (T-bet/STAT4), Th17 (IL-17A/RORγT), Th22 (TNF-α/IL-22), and regulatory T (Treg/Foxp3) cells in the spleen, using flow cytometry. We also analyzed the effect of CA-074 on T-bet, IL-17A, RORγT, IL-22, and mRNA and protein levels using RT-PCR and western blot analysis for brain tissues. Cathepsin B expression were also assessed by western blot in the brain tissues. The severity of clinical scores decreased significantly in CA-074-treated mice compared with that in EAE control mice. Moreover, the percentage of CD4+T-bet+, CXCR5+T-bet+, CD4+STAT4+, CD4+IL-17A+, CXCR5+IL-17A+, CD4+RORγT+, CCR6+RORγT+, CD4+TNF-α+, CD4+IL-22+, and CCR6+IL-22+ cells decreased while CD25+Foxp3+ increased in CA-074-treated EAE mice as compared to vehicle-treated EAE mice. Further, CA-074-treated EAE mice had downregulated Cathepsin B protein expression which was associated with decreased T-bet, IL-17A, RORγT, and IL-22 mRNA/protein expression. These results suggest that Cathepsin B could be a novel therapeutic candidate against for the treatment of MS.Lithium, in addition to its effect on acute and long-term bipolar disorder, is involved in neuroprotection after ischemic stroke. Therefore, we initially analyzed the dose-dependent effects of lithium on neurological deficits, infarct volume, brain edema and blood-brain barrier integrity, along with neuronal injury and survival in mice subjected to focal cerebral ischemia. Thereafter, we investigated the involvement of the PI3K/Akt and MEK signal transduction pathways and their components. Our observations revealed that 2 mmol/kg lithium significantly improved post-ischemic brain tissue survival. Although, 2 mmol/kg lithium had no negative effect on brain microcirculation, 5 and 20 mmol/kg lithium reduced brain perfusion. Furthermore, supratherapeutic dose of lithium in 20 mmol/kg lead to animal death. In addition, improvement of brain perfusion with L-arginine, did not change the effect of 5 mmol/kg lithium on brain injury. Additionally, post-stroke blood-brain barrier leakage, hemodynamic impairment and apoptosis have been reversed by lithium treatment. Interestingly, lithium-induced neuroprotection was associated with increased phosphorylation of Akt at Thr308 and suppressed GSK-3β phosphorylation at Ser9 residue. Lithium upregulated Erk-2 and downregulated JNK-2 phosphorylation. To distinguish whether neuroprotective effects of lithium are modulated by PI3K/Akt or MEK, we sequentially blocked these pathways and demonstrated that the neuroprotective activity of lithium persisted during MEK/ERK inhibition, whereas PI3K/Akt inhibition abolished neuroprotection. Collectively, we demonstrated lithium exerts its post-stroke neuroprotective activity via the PI3K/Akt pathway, specifically via Akt phosphorylation at Thr308, but not via MEK/ERK.Gravity Recovery and Climate Experiment (GRACE) and GRACE Follow-On (GRACE-FO), two successive satellite-based missions starting in 2002, have provided an unprecedented way of measuring global terrestrial water storage anomalies (TWSA). However, a temporal gap exists between GRACE and GRACE-FO products from July 2017 to May 2018, which introduces bias and uncertainties in TWSA calculations and modeling. Previous studies have incorporated hydroclimatic factors as predictors for filling the gap, but most of them utilized artificial intelligence or pure statistical models that generally de-trended TWSA and had no physical foundation. Thus, a physically-based reconstruction is required for increasing robustness. In this study, we bridge the temporal gap by developing an empirical hydrological model. The "abcd" model, a T-based snow component, and linear correction are utilized to represent runoff generation, snow dynamics, and long-term trends. The testing results indicate that our hydrological model can successfully reconstruct TWSA in tropical, temperature, and continental climates, although further improvement is needed for arid climates. Our reconstruction for the gap achieves high accuracy and robustness as shown by the evaluations against sea-level budget and GLDAS-derived TWSA. Compared to previous studies using artificial intelligence or statistical techniques, our hydrological model performs similarly in the gap filling but does not involve de-trended or de-seasonalized transformations, which will facilitate the combination of GRACE and GRACE-FO products and improve the physical understanding of global TWSA.Land cover has been demonstrated to have substantial impacts on climate and dry-wet environment, but potential influence of landscape pattern dynamics accompanying land cover change on drought remains unclear. In this study, response of dry-wet dynamics to landscape pattern in China was examined. Results suggest that landscape pattern in China's nine agricultural districts had transformed to varying extents and showed spatiotemporal heterogeneity from 1980 to 2018. For forest landscape, the highest annual average Percentage of Landscape (PLAND) was recorded in SC, reaching 62.26%; and the highest Largest Patch Index (LPI) was presented in YGP, followed by SC, with annual values of 53.79% and 46.26% respectively. The QTP has the most prominent forest connectivity in spite of its lower abundance. For grass landscape, the highest abundance and dominance were recorded in QTP, with annual PLAND fluctuation range of 49.66%-63.52% and annual LPI variation range of 34.10%-58.46%, which is associated with its climate should be taken into account.Volcanoes are a significant component of the Earth system, influencing the interaction between oceans and the atmosphere over large spatial and temporal scales. Being a volcanically dynamic region, the Tropical Western Pacific (TWP) can significantly impact variations in global climate. However, high-resolution continuous records of volcanic activity in this region are lacking, resulting in significant uncertainties regarding the coupling between the deep earth, climate changes, and atmospheric CO2 in the TWP. To address this issue, mercury (Hg) levels, isotopic compositions, and Hg/total organic carbon (Hg/TOC) ratios were determined at site U1486 to track volcanic activity throughout the mid-Pleistocene transition (MPT) from 1.3 Myr to 0.6 Myr. Our results of anomalously high Hg concentrations and Hg/TOC ratios provide evidence of time-varying volcanism throughout the MPT. Mercury isotopes in the Hg-enriched sediments were characterized by near-zero Δ199Hg values, which is consistent with volcanism acting as the primary source of Hg to the sediments. Spectral analysis of the Hg/TOC ratio showed significant periodicity at ~100 kyr and ~ 23 kyr as well as a weaker signal at ~41 kyr consistent with Milankovitch cycles. A cross spectral analysis of Hg/TOC and the LR04 δ18O stack record suggests that the peak in volcanism lags the temperature minimum by ~6 kyr, and occurs prior to the δ18O minimum known as the glacial termination by ~14 ± 2 kyr. The records of volcanic activity in this site are also consistent with a prominent rise in atmospheric CO2 and negative excursion of benthic carbon isotopes throughout the MPT. This study provides direct sedimentary evidence in the TWP of the feedback between volcanic activity, climate change and atmospheric CO2.The concept of groundwater vulnerability was first introduced in the 1970s in France to recognize sensitive areas in which surface pollution could affect groundwater, and to enable others to develop management methods for groundwater protection against surface pollutants. Since this time, numerous methods have been developed for groundwater vulnerability assessment (GVA). These can be categorized into four groups (i) overlay and index-based methods, (ii) process-based simulation models, (iii) statistical methods, and (iv) hybrid methods. This work provides a comprehensive review of modern GVA methods, which in contrast to previous reviews, examines the last two categories in detail. First, the concept of groundwater vulnerability is defined, then the major GVA methods are introduced and classified. This includes detailed accounts of statistical methods, which can be subdivided into orthodox statistical, data-driven and Bayesian methods, and their advantages and disadvantages, as well as modern hybrid methods.
