However, a similar linkage between histone and mRNA modifications has not yet been investigated in plants. Here, we highlight the highly influential discovery in mammals, and also show that H3K36me2 is a possible determinant of m6 A deposition in Arabidopsis thaliana. Based on an extensive bioinformatics analysis of the global distributions of m6 A and various histone modifications in Arabidopsis, we identified a significant co-occurrence of m6 A and H3K36me2. Meta-gene analysis showed that m6 A deposition in mRNAs largely depends on the presence of the H3K36me2 histone mark in the genome. Additionally, molecular factors involved in H3K36me2 and m6 A metabolism form protein-protein interaction networks. These results provide insights into the conservation of the intimate connections between chromatin and mRNA modifications across the animal and plant systems. This article is protected by copyright. However, the roles of these genes in the contributions of in aphids'adaptation and whether these genes could be used as RNAi-based pest control targets are not yet clear. Thus, in this study we used parental RNAi to analyze the potential function of a carotenoid desaturase gene (CdeB) by combined molecular and chemical approaches in the pea aphid (Acyrthosiphon pisum). RESULTS Transcriptional analyses showed that CdeB was significantly more highly expressed in the red morphs compared to the green ones and was associated with the production of red carotenoid. Co-transferring of pET28a-CdeB (the CdeB gene was cloned into pET28a) and pACCRT-EIB (produced lycopene) showed a deep red color in the bacterial precipitate and produced more of a red pigment, lycopene, in vitro. Parental gene-silencing of CdeB resulted in a lower body color intensity in the treated aphids and following generations in vivo. Interestingly, the dsCdeB treatment also reduced aphid performance as reflected by a delay in nymphal developmental duration, lower weight, smaller number, and altered age structure of the population. CONCLUSION Our results demonstrate that CdeB is involved in red color formation and the silencing of this gene by parental RNAi reduced fitness in the pea aphid. The results enhance our understanding of the biosynthesis of carotenoid in aphids and provide insights into the potential ecological significance of carotenoids in the adaptation of the aphid's biology to the environment and developing environmentally friendly control strategies for this pest. © 2020 Society of Chemical Industry.AIM Approximately 15 years ago, four youth access clinics (YACs) were established in the Gippsland region to improve mental and general healthcare for young people living in the region. The aim of the study was to examine the barriers and enablers for young people accessing the YACs in Gippsland. METHODS Using qualitative methodology, nine female YAC staff members and seven youth YAC consumers were interviewed. Thematic analysis was utilized to identify important and consistent themes in the data derived through the consultation process. RESULTS Barriers under four themes were identified environmental (eg, limited transport); service (ie, limited opening hours); client (eg, parent permission) and staff (ie, retention of staff). Enablers were identified as environmental (ie, high social proximity), service (eg, funding), client (ie, awareness of service) and staff (eg, champion staff). CONCLUSION The success of new rural service models will likely depend on learning from what has already worked in some of the many small communities in this region. Beyond learning, further success and uptake of new service offerings will be enhanced through understanding community needs, obtaining community support and enhancing high social proximity. © 2020 John Wiley & Sons Australia, Ltd.BACKGROUND The beet armyworm, Spodoptera exigua, is a serious agricultural pest that is primarily controlled using chemical insecticides. Recently, resistance to the insecticide spinosad has been described in S. exigua field populations. To date, there has been no functional evidence proving the involvement of the nicotinic acetylcholine receptor (nAChR) α6 mutation in spinosad resistance in S. exigua. RESULTS In this study, using the CRISPR/Cas9 genome-editing system, a homozygous strain (Seα6-KO) with approximately 1760-bp deletion within Seα6 in S. exigua causing a premature truncation of Seα6 was successfully constructed. Insecticide bioassays showed that Seα6-KO exhibited 373-fold higher resistance to spinosad and 850-fold higher resistance to spinetoram compared to WH-S strain with the same genetic background but showed no significant change in susceptibility to emamectin benzoate and chlorantraniliprole. Genetic analysis revealed that Seα6-KO is inherited as an incompletely recessive trait. CONCLUSION The results clearly demonstrated the functional role of Seα6 in resistance to spinosyn insecticides and provide an example of using genome editing to verify a target premature truncation associated with resistance. © 2020 Society of Chemical Industry.OBJECTIVE To determine the influence of sampling methods on culture results and selection of antimicrobials for treating infected wounds in dogs. DESIGN Prospective study from January to July 2016. SETTING Three private multispecialty referral centers. ANIMALS Fifty-two client-owned dogs with infected wounds. INTERVENTIONS Each wound was sampled for culture by 3 methods swabbing prior to preparation (dirty swabs), swabbing after debridement and sterile lavage (clean swabs), and biopsy. Bacterial species and phenotypes were compared. Three clinicians unaware of patient, wound, and sampling information selected antimicrobial drugs based on culture and sensitivity reports. Antimicrobials were divided into class I, II, or III based on established guidelines. The number, highest class of antimicrobial chosen, and inter-investigator agreement were examined. MEASUREMENTS AND MAIN RESULTS Identical populations of bacteria were isolated for all 3 sampling techniques in only 31% of wounds. Significantly fewer bacterialr to sampling by swabbing did not alter the number or type of bacteria isolated, nor the number or the highest class of antimicrobial selected by clinicians. © Veterinary Emergency and Critical Care Society 2020.BACKGROUND Chronic musculoskeletal pain is a major health concern. The biopsychosocial approach is an evidence-based approach recommended for managing chronic musculoskeletal pain. However, the evidence for this approach is largely reported from high-income countries; therefore, it is important to ascertain how biopsychosocial approaches are operationalised in low- and middle-income countries to inform practice. AIM To examine the evidence for the operationalization of biopsychosocial interventions in managing patients with chronic musculoskeletal pain in low-and middle-income countries. METHODS The search included studies published in English from 2008 to September 2019 in Cochrane library, OVID, CINAHL, Scopus, PUBMED, Web of Science, and SportDiscus. Randomised and non-randomised trials using a biopsychosocial intervention were considered. The review team developed a search strategy; two independent reviewers screened and assessed results for quality. RESULTS Sixteen studies were included (n = 996) with mainly low back pain populations (n = 11 studies). Others were osteoarthritis (n = 1) and other musculoskeletal pain (n = 4). The majority (n = 12) of studies attained fair to poor quality, three had good quality, one scored excellent quality. MATERIALS AND METHODS The study was conducted in a private hair transplant clinic after obtaining informed consent. Only those patients were included who had a previous strip harvest surgery (FUT) and opted for Follicular Unit Excision/Extraction (FUE) procedure. In all the patients, the donor hairs were trimmed to about 1-2 mm length. The width of the scar, the hair density above and below the scar, and hair angles above and below the scar were measured. All the patients underwent FUE procedure using 0.9 mm serrated punch. Twenty attempts were made above and below the scar (about 0.5 cm), and the total number of hair and transections was measured. All the data were analyzed statistically. RESULTS Ten patients were included. The average width of the scar was 2.47 mm. The mean hair density was found to be 155.4 hairs per cm2 above the scar and 130.8 per cm2 below the scar. The mean hair angle was found to be 28.21 degrees above the scar and 39.19 degrees below the scar in midline occipital above the scar (P less then .05). The average hair count was found to be 49.8 hairs above the scar and 47.2 hairs above the scar (P less then .05). The transection of hair was doubled below the scar (P less then .05) CONCLUSION The Follicular Unit Transplantation (FUT) procedure does leave some impacts on the donor area other than the scar only. The change in angle results in higher transection of hair above the scar. © 2020 Wiley Periodicals, Inc.BACKGROUND Hair transplant procedure is a surgical procedure which affects the sensitivity of the scalp. OBJECTIVES To compare scalp two-point discrimination (2PD) values in healthy controls and patients who had hair transplant surgery. PATIENTS/METHODS The comparative study was conducted in 25 healthy individuals and 25 patient who underwent hair transplant surgery (either strip surgery or FUE). An area was marked o scalp about 10 cm from the nasion. A caliper with 15 g weight was used. Both the prongs were ensured to be placed perpendicular to the skin. The contact time was only 2.0 seconds. An interval of 10 seconds was given before increasing the 2PD distance. In each measurement, the distance was increased by 1 mm. All the data were recorded and analyzed statistically using ANOVA test. RESULTS The mean age of the control group was 29.72 and 31.7 years in patients' group. The average 2PD score in controls was 12.76 and 17.80 mm in patients. The average 2PD score was found to be 17.7 in patients who had surgery less than 18 months ago as compared to the score of 17.
