Here we combined extensive catchment sampling, molecular detection tools and genomic signatures to i) assess the prevalence of the rosette agent Sphaerothecum destruens in invasive and native fish populations in contrasting french regions, and ii) characterize the genetic diversity and population structure of its co-invasive and asymptomatic carrier Pseudorasbora parva. Although S. destruens was not detected in all the fish collected its presence in contrasting freshwater ecosystems suggests that the disease may already be widespread in France. Furthermore, our results show that the detection of S. destruens DNA in its asymptomatic carrier P. parva is still limited. Finally, we found that P. parva populations show a homogeneous genetic and geographical structuring, which raises the possibility of the occurrence of successive introduction events in France from their native and invasive range.Anammox is sensitive to temperature, which can limit its practical application in wastewater treatment. In this study, a step-feed anoxic-oxic (A/O) process coupled with PD/A was operated steadily from 26.8 °C to 13.1 °C for wastewater treatment for 200 days. The effluent total inorganic nitrogen (TIN) and phosphorus concentrations were 10.2 mg/L and 0.29 mg/L at C/N ratio of 4.6 and 15.0 °C even with increasing nitrogen loading rate (NLR). The anammox activity was 5.60 mg NH4+-N/gMLSS/d even at 14 °C, moreover, anammox abundance on the biocarriers increased with decreasing temperature. It was observed that the effect of partial denitrification (PD) was enhanced under low temperature, thus the contribution of anammox for nitrogen removal was improved. The pathway of anammox for nitrogen removal accounted for 48% and the effect of effluent did not deteriorate under low temperature. This study states that PD/A has advantages under low temperature operation, which is suitable for treatment of wastewater with low C/N ratio.The growing contamination of arsenic and plastics has severely effects on the soil fauna health, including shifts of gut microbiota community. A few studies have focused on effects of microplastics and metal(loid) in soil and fauna gut microbiome. However, the environmental effect of nanoplastics and arsenic on the earthworm gut microbiota, especially on arsenic biotransformation in the gut, remain largely unknown. Here, a microcosm study was performed to explore the effects of nanoplastics and arsenic on the microbiota characteristics in earthworm Metaphire vulgaris gut using Illumina high throughput sequencing, and to investigate changes in the gut microbiota-mediated arsenic biotransformation genes (ABGs) by using high-throughput quantitative PCR. Our results demonstrated that the concentration of arsenic in the earthworm body tissues after exposure to arsenic and nanoplastics was significantly lower from that with arsenic alone exposure. Moreover, the clearly different bacterial community was observed in the soil compared with the earthworm gut, which was dominated by Proteobacteria, Actinobacteria, and Firmicutes at phylum level. Arsenic exposure significantly disturbed bacterial community structure in the earthworm gut, but exposure to nanoplastics did not induce gut microbiota changes. More interestingly, nanoplastics can relieve adverse effect of arsenic on the gut microbiota possibly by adsorbing arsenic. In addition, a total of 16 ABGs were detected, and predominant genes involved in arsenic reduction and transport process were observed in the earthworm guts. In short, this study provides a new picture of the effects of nanoplastics and arsenic on the gut microbiota and arsenic biotransformation in soil fauna gut.The management of sugarcane ripening is essential to ensuring the supply of high-quality raw material for the sugar-alcohol industry; chemical ripeners are frequently used to accelerate sucrose accumulation in the stalks during harvesting. The potential ripening effect of a low dose of glyphosate was evaluated in sugarcane, along with its impact on productivity and sprouting in the next crop cycle. A field experiment was conducted in 2015 and 2016 using a randomized block design with eight replicates in a split-plot scheme, with the following treatments (1) control with only water application, (2) glyphosate at a low dose of 1.8 g a.e. ha-1 (corresponding to 0.005 L ha-1 of the commercial product (cp)), and (3) glyphosate at the commercially recommended dose for a ripener at 180 g a.e. ha-1 (corresponding to 0.50 L ha-1 of the cp) applied at 60, 45, 30, and 15 days before harvest (DBH). The harvest was performed on May 25, 2016 (0 DBH), and a total of five periods were evaluated. This study showed that the application of a hormetic dose of glyphosate to stimulate sugarcane ripening is promising, despite the limited duration of the effect. The application of the hormetic dose (1.8 g a.e. ha-1) at 30 DBH improved the technological quality of sugarcane in terms of Brix% juice, pol% cane, purity% juice, moisture% cane, reducing sugars, total reducing sugars, and total recoverable sugar. Additionally, it increased pol productivity, and did not affect ratoon sprouting in the subsequent cycle. Thus, this study provides a strategy for ripening management with a low environmental impact for sugarcane producers through a low (hormetic) dose of glyphosate.Serological tests play an important role in the fight against Coronavirus Disease 2019 (COVID-19), including monitoring the dynamic immune response after vaccination, identifying past infection and determining community infection rate. Conventional methods for serological testing, such as enzyme-linked immunosorbent assays and chemiluminescence immunoassays, provide reliable and sensitive antibody detection but require sophisticated laboratory infrastructure and/or lengthy assay time. Conversely, lateral flow immunoassays are suitable for rapid point-of-care tests but have limited sensitivity. Here, we describe the development of a rapid and sensitive magnetofluidic immuno-PCR platform that can address the current gap in point-of-care serological testing for COVID-19. Our magnetofluidic immuno-PCR platform automates a magnetic bead-based, single-binding, and one-wash immuno-PCR assay in a palm-sized magnetofluidic device and delivers results in ∼30 min. AZD9291 In the device, a programmable magnetic arm attracts and transports magnetically-captured antibodies through assay reagents pre-loaded in a companion plastic cartridge, and a miniaturized thermocycler and a fluorescence detector perform immuno-PCR to detect the antibodies. We evaluated our magnetofluidic immuno-PCR with 108 clinical serum/plasma samples and achieved 93.8% (45/48) sensitivity and 98.3% (59/60) specificity, demonstrating its potential as a rapid and sensitive point-of-care serological test for COVID-19.A small sample tilt away from a main zone axis orientation results in an elongation of the atomic columns in ADF STEM images. An often posed research question is therefore whether the ADF STEM image intensities of tilted nanomaterials should be quantified using a parametric imaging model consisting of elliptical rather than the currently used symmetrical peaks. To this purpose, simulated ADF STEM images corresponding to different amounts of sample tilt are studied using a parametric imaging model that consists of superimposed 2D elliptical Gaussian peaks on the one hand and symmetrical Gaussian peaks on the other hand. We investigate the quantification of structural parameters such as atomic column positions and scattering cross sections using both parametric imaging models. In this manner, we quantitatively study what can be gained from this elliptical model for quantitative ADF STEM, despite the increased parameter space and computational effort. Although a qualitative improvement can be achieved, no significant quantitative improvement in the estimated structure parameters is achieved by the elliptical model as compared to the symmetrical model. The decrease in scattering cross sections with increasing sample tilt is even identical for both types of parametric imaging models. This impedes direct comparison with zone axis image simulations. Nonetheless, we demonstrate how reliable atom-counting can still be achieved in the presence of small sample tilt.Membrane proteins can be regulated by alterations in material properties intrinsic to the hosting lipid bilayer. Here, we investigated whether the reversible photoisomerization of bilayer-embedded diacylglycerols (OptoDArG) with two azobenzene-containing acyl chains may trigger such regulatory events. We observed an augmented open probability of the mechanosensitive model channel gramicidin A (gA) upon photoisomerizing OptoDArG's acyl chains from trans to cis integral planar bilayer conductance brought forth by hundreds of simultaneously conducting gA dimers increased by typically >50% - in good agreement with the observed increase in single-channel lifetime. Further, (i) increments in the electrical capacitance of planar lipid bilayers and protrusion length of aspirated giant unilamellar vesicles into suction pipettes, as well as (ii) changes of small-angle X-ray scattering of multilamellar vesicles indicated that spontaneous curvature, hydrophobic thickness, and bending elasticity decreased upon switching from trans- to cis-OptoDArG. Our bilayer elasticity model for gA supports the causal relationship between changes in gA activity and bilayer material properties upon photoisomerization. Thus, we conclude that photolipids are deployable for converting bilayers of potentially diverse origins into light-gated actuators for mechanosensitive proteins.Because contaminated livestock trailers are a significant risk for transmitting viruses between herds, various methods of washing, disinfecting, and thermo-assisted drying and decontamination (TADD) have been evaluated for their effectiveness in inactivating porcine reproductive and respiratory syndrome virus (PRRSV) on contaminated surfaces. Information on when to expect negative qRT-PCR results after adequate trailer sanitation is lacking. The objective of this study was to evaluate whether there are conditions associated with washing-disinfectant-TADD procedures that will consistently produce a negative qRT-PCR result for the purpose of monitoring compliance with trailer sanitation and decontamination protocols for PRRSV on metal surfaces. 144 diamond plate aluminum coupons were spiked with PRRSV or phosphate-buffered saline (PBS) and treated with a designated disinfectant protocol. Disinfectants evaluated included multiple accelerated® hydrogen peroxide (AHP) disinfectants and a quaternary ammonium and glutaraldehyde combination disinfectant.
