CONCLUSION A fall in blood pressure occurred more commonly in patients starting HD with lower overhydration as measured by bioimpedance, and those with smaller cardiac chamber sizes. Patients with the lowest ECW/TBW and smallest cardiac chamber sizes had the greater falls in SBP. All rights reserved.Recent advances in diffusion magnetic resonance imaging (dMRI) analysis techniques have improved our understanding of fibre-specific variations in white matter microstructure. Increasingly, studies are adopting multi-shell dMRI acquisitions to improve the robustness of dMRI-based inferences. However, the impact of b-value choice on the estimation of dMRI measures such as apparent fibre density (AFD) derived from spherical deconvolution is not known. Here, we investigate the impact of b-value sampling scheme on estimates of AFD. First, we performed simulations to assess the correspondence between AFD and simulated intra-axonal signal fraction across multiple b-value sampling schemes. We then studied the impact of sampling scheme on the relationship between AFD and age in a developmental population (n = 78) aged 8-18 (mean = 12.4, SD = 2.9 years) using hierarchical clustering and whole brain fixel-based analyses. Multi-shell dMRI data were collected at 3.0T using ultra-strong gradients (300 mT/m), using 6 diffusion-weighted shells ranging from b = 0 to 6,000 s/mm2 . Simulations revealed that the correspondence between estimated AFD and simulated intra-axonal signal fraction was improved with high b-value shells due to increased suppression of the extra-axonal signal. These results were supported by in vivo data, as sensitivity to developmental age-relationships was improved with increasing b-value (b = 6,000 s/mm2 , median R2 = .34; b = 4,000 s/mm2 , median R2 = .29; b = 2,400 s/mm2 , median R2 = .21; b = 1,200 s/mm2 , median R2 = .17) in a tract-specific fashion. Overall, estimates of AFD and age-related microstructural development were better characterised at high diffusion-weightings due to improved correspondence with intra-axonal properties. © 2020 The Authors. Here, we review recent works on the cytoskeleton using cryo-electron tomography, demonstrating the power of in situ studies. All rights reserved. © 2020 The Protein Society.We report the preparation of lithium salt free KDA (potassium diisopropylamide; 0.6 m in hexane) complexed with TMEDA (N,N,N',N'-tetramethylethylenediamine) and its use for the flow-metalation of (hetero)arenes between -78 °C and 25 °C with reaction times between 0.2 s and 24 s and a combined flow rate of 10 mL/min using a commercial flow set-up. The resulting potassium organometallics react instantaneously with various electrophiles, such as ketones, aldehydes, alkyl and allylic halides, disulfides, Weinreb amides and Me3SiCl, affording functionalized (hetero)arenes in high yields. This flow procedure is successfully extended to the lateral metalation of methyl-substituted arenes and heteroaromatics resulting in the formation of various benzylic potassium organometallics. KGaA, Weinheim.INTRODUCTION Kinsenoside is a characteristic component of Anoectochilus roxburghii and accounts for this herb's medicinal and edible values. No international certified standard method is available for kinsenoside analysis as well as extraction and preservation. OBJECTIVE To develop a more accurate analytical method of kinsenoside. The effects of extraction and drying methods of A. roxburghii on kinsenoside efficiency were investigated for the first time, as well as to examine the kinsenoside stability. MATERIAL AND METHODS The amino (NH2 ) and AQ-C18 columns for detecting kinsenoside extract was systematically compared by high-performance liquid chromatography evaporative light-scattering detector (HPLC-ELSD) and HPLC-diode-array detector (DAD), respectively. Kinsenoside, its epimer goodyeroside A and the degradation product during preservation were identified through HPLC-electrospray ionization mass spectrometry (ESI-MS). RESULTS An accurate method of kinsenoside detection by HPLC-ELSD with dual columns of NH2 and AQ-C18 was established. The ratio of Cgoodyeroside A to Ckinsenoside (Y) was determined using the AQ-C18 column method. The concentration detected by the NH2 column was multiplied by 1/(1 + Y) as the corrected result. Using this novel method, the average deviations were reduced by 7.64%. Moreover, the efficiency of kinsenoside extraction with water was almost twice that of extraction with ethanol. Freeze drying also led to a higher extraction efficiency (38.47% increase) than hot-air drying did. Furthermore, the degradation of kinsenoside extract exceeded 70% when stored at 37 °C for 3 months. CONCLUSION This study provides a reliable experimental method and theoretical basis for the quality control of kinsenoside from A. roxburghii, as well as other glycosides. © 2020 John Wiley & Sons, Ltd.Genome-wide expression quantitative trait loci (eQTLs) mapping explores the relationship between gene expression and DNA variants, such as single-nucleotide polymorphism (SNPs), to understand genetic basis of human diseases. Due to the large number of genes and SNPs that need to be assessed, current methods for eQTL mapping often suffer from low detection power, especially for identifying trans-eQTLs. In this paper, we propose the idea of performing SNP ranking based on the higher criticism statistic, a summary statistic developed in large-scale signal detection. We illustrate how the HC-based SNP ranking can effectively prioritize eQTL signals over noise, greatly reduce the burden of joint modeling, and improve the power for eQTL mapping. Numerical results in simulation studies demonstrate the superior performance of our method compared to existing methods. The proposed method is also evaluated in HapMap eQTL data analysis and the results are compared to a database of known eQTLs. © 2020 Wiley Periodicals, Inc.We aimed to determine the effect on choroidal thickness of daily 5 mg tadalafil use in patients with erectile dysfunction (ED) and to evaluate agreement between short form of International Index of Erectile Function (IIEF 1-5, 15) scores and choroidal thickness during follow-up. Enhanced depth imaging spectral domain-optical coherence tomography (EDI-OCT) was used to measure full choroidal thickness in the subfoveal area, choroidal thickness (CT) and small-choroidal-vessel-layer (SCVL) thickness. Thirty of the 45 patients included complete their follow-ups and exhibited medication continuity. Patients were divided into two groups, five (16.7%) with ED at any level at third-month follow-up, and 25 (83.3%) with no ED, and subgroup analysis was then performed. Median changes in SCVL thickness at first- and sixth-month follow-ups were 20.5 µm versus 9.0 μm (p = .001) and 23.5 µm versus 12.5 µm (p = .005) in patients without and with ED respectively. The SCVL thickness increased by 20 μm compared with the baseline level, indicating an improvement in the patient's complaints of erectile dysfunction. Explorative studies have shown CG motifs from the prokaryotic origin as activators of TLR9 culminating in the expression of NFκB. These CG rich short stranded DNA sequences have been further delineated into different classes based on their structural specificities and immunomodulatory properties. Here we discuss the progress of how activation of TLR9 can be utilized with novel parasitic CpG islands to function as potential adjuvants specifically against protozoan parasitic diseases primarily visceral leishmaniasis caused by Leishmania donovani. © 2020 Wiley Periodicals, Inc.Swine acute diarrhea syndrome coronavirus (SADS-CoV) is a novel coronavirus that is involved in severe diarrhea disease in piglets, causing considerable agricultural and economic loss in China. The emergence of this new coronavirus increases the importance of understanding SADS-CoV as well as antivirals. Coronaviral proteases, including main proteases and papain-like proteases (PLP), are attractive antiviral targets because of their essential roles in polyprotein processing and thus viral maturation. Here, we describe the biochemical and structural identification of recombinant SADS papain-like protease 2 (PLP2) domain of nsp3. The SADS-CoV PLP2 was shown to cleave nsp1 proteins and also peptides mimicking the nsp2|nsp3 cleavage site and also had deubiquitinating and deISGynating activity by in vitro assays. The crystal structure adopts an architecture resembling that of PLPs from other coronaviruses. We characterize both conserved and unique structural features likely directing the interaction of PLP2 with the substrates, including the tentative mapping of active site and other essential residues. These results provide a foundation for understanding the molecular basis of coronaviral PLPs' catalytic mechanism and for the screening and design of therapeutics to combat infection by SADS coronavirus. © 2020 The Protein Society.Organocatalytic enantioselective 1,3-dipolar [6+4] cycloadditions of pyrylium ion intermediates with fulvenes promoted by a chiral primary amine catalyst have been developed to proceed in moderate to good yields and high enantioselectivities. The resultant chiral bicyclo[6.3.0]undecane scaffold containing a transannular bridging ether is densely functionalised providing a rigid scaffold for further manipulations. Computational studies give important insights into the role of the primary amine catalyst. Analysis of the reaction shows that the catalytic reaction proceeds in a step-wise manner and rationalises the stereochemical outcome of the reaction. Several stereoselective complexity-generating transformations, facilitated by the diverse functional groups and transannular bridge, are presented, highlighting the versatility of the core towards a number of the cyclooctanoid natural products. © 2020 WILEY-VCH Verlag GmbH & Co.
