Fucosylated oligosaccharides are the major components of human milk oligosaccharide (HMO) which confer various beneficial effects including prebiotic effect and protection from pathogenic infection on the host.

A novel prebiotics was developed using bifidobacterial β-galactosidase and fucose and lactose as substrates. Structure analysis revealed it as β-D-galactopyranosyl-(1 → 3)-O-L-fucopyranose named as β-galactosyl fucose α1-4 linkages. Among the four Lactobacillus strains examined, all but L. delbrueckii subsp. bilgaricus KCTC 3635 grew better on gal-fuc than on β-GOS. Among the 11 bifidobacterial species examined, all except for B. bifium used gal-fuc as much as GOS.

Moreover, the gal-fuc was noticeably better used by Bifidobacterium infantis, the major intestinal bacteria of breast fed infant. Among 15 non-probiotic bacteria, only 4 strains used gal-fuc better than β-GOS. In conclusion, a novel gal-fuc is expected to contribute to beneficial changes of gut microbiota. Graphical abstract A novel form of β-galactosyl fucose with High-Fructose Corn-Syrup-Sweetened Beverage Intake Increases 5-Hour Breast Milk Fructose Concentrations in Lactating Women.This study determined the effects of consuming a high-fructose corn syrup concentrations in lactating women. At six weeks postpartum, lactating mothers (n = 41) were randomized to a crossover study to consume a commercially available HFCS-sweetened beverage or artificially sweetened control beverage. At each session, mothers pumped a complete breast milk expression every hour for six consecutive hours.

The baseline fasting concentrations of breast milk fructose, glucose, and lactose were 5 ± 1 µgmL, ± mgmL, and 6 ± 1 gdL, respectively. The changes over time in breast milk sugars were significant only for fructose (treatment × time, p 1). Post hoc comparisons showed the HFCS-sweetened beverage vs. control beverage increased breast milk fructose at 1 min (8 ± 2 vs. 5 ± 1 µgmL), 1 min (9 ± 1 vs. 5 ± 2 µgmL), 2 min (7 ± 1 vs. 5 ± 1 µgmL), and 0 min (6 ± 1 vs.

4 ± 1 µgmL) (all p 5). The mean incremental area under the curve for breast milk fructose was also different between treatments (14 ± 1 vs. -2. ± 1 µgmL × 3 min, p 1). There was no treatment × time interaction for breast milk glucose or lactose. Our data suggest that the consumption of an HFCS-sweetened beverage increased breast milk fructose concentrations, which remained elevated up to five hours post-consumption.Conflict of interest statement The authors declare no conflict of interest.

In this study, human milk oligosaccharide (HMO) 2'-fucosyllactose (α-L-Fuc-(1,2)-D-Galβ-1,4-Glu, 2'FL) and its isomer 3'-fucosyllactose (α-L-Fuc-(1,3)-D-Galβ-1,4-Glu, 3'FL) with potential prebiotic effect were synthesized efficiently by a novel recombinant α-L-fucosidase.

An α-L-fucosidase gene (PbFuc) from Pedobacter sp. CAU9 was successfully cloned and expressed in Escherichia coli (E. coli). The deduced amino acid sequence shared the highest identity of 36% with the amino sequences of other reported α-L-fucosidases. The purified α-L-fucosidase (PbFuc) had a molecular mass of kDa. The enzyme exhibited specific activity and 2'FL (3 Umg). It showed the highest activity at pH 5 and 35 °C, respectively.
