DNA extractions of semen samples commonly utilize dithiothreitol (DTT) to reduce and disrupt disulfide bonds. Although traditional extraction techniques remove DTT before downstream analyses, the forensic DNA community has recently explored Y-screening, direct amplification, and direct cell lysis assays that omit purification but employ reducing agents to lyse spermatozoa. This study examined the impact of residual DTT on downstream processes involving fluorescent dyes. Quantification using Investigator® Quantiplex HYres revealed a significant increase in the male DNA yield (p = 0.00056) and a >150,000,000-fold increase in the malehuman DNA ratio when DTT remained in extracts versus when it was filtered out using a traditional purification method. When DTT was present with Quantifiler™ Trio, the true mean DNA yield for the large autosomal target significantly increased (p = 0.038) and the average reported DNA yields increased 1.1-fold, >9.5-fold, and 1.3-fold for the small autosomal, large autosomal, and male targets, respectively. check details DTT-spiked DNA standards from both kits were impacted similarly to samples with residual DTT, demonstrating that observed effects were related to DTT and not the extraction method. This study corroborates other reports that DTT adversely affects multiple dyes (e.g., Cy5, Quasar 670, SYBR Green I, TMR, and Mustang Purple® ). Overall, DTT causes inaccurate quantities and, consequently, inaccurate calculated malefemale ratios when used in conjunction with these kits. Thus, implementation of newer direct-to-PCR assays incorporating DTT should either be avoided or used only with carefully evaluated, compatible dyes.Changes in the abundance of protein and RNA molecules can impair the formation of complexes in the cell leading to toxicity and death. Here we exploit the information contained in protein, RNA and DNA interaction networks to provide a comprehensive view of the regulation layers controlling the concentration-dependent formation of assemblies in the cell. We present the emerging concept that RNAs can act as scaffolds to promote the formation ribonucleoprotein complexes and coordinate the post-transcriptional layer of gene regulation. We describe the structural and interaction network properties that characterize the ability of protein and RNA molecules to interact and phase separate in liquid-like compartments. Finally, we show that presence of structurally disordered regions in proteins correlate with the propensity to undergo liquid-to-solid phase transitions and cause human diseases. Many of these repeat expansion disorders are neurodegenerative movement disorders. Radical cures for these disorders have yet to be established. Although conventional treatments for repeat expansion disorders have mainly targeted the abnormal mRNA and proteins encoded by the affected genes, therapeutic approaches targeting repeat DNA, the root cause of repeat disorders, is also being explored in current research. In particular, a small molecule has been found that binds to abnormally expanded CAG repeats, the cause of Huntington's disease, and shortens them. Such small molecules targeting nucleic acids are expected to be developed into groundbreaking therapeutic drugs capable of ameliorating the symptoms of repeat expansion disorders and preventing their onset. © 2020 International Parkinson and Movement Disorder Society.
The obligatory sunflower root parasite Orobanche cumana Wallr. deprives its host of essential nutrients, resulting in a dramatic reduction in yield and biomass. A post-emergence application with an imidazolinone herbicide on an imidazolinone-tolerant sunflower is highly effective against O. cumana. link2 The herbicide inhibits the enzyme acetohydroxy acid synthase and consequently, growth of the parasite is inhibited, although the sunflower survives the treatment through mutations in the target enzyme. Interestingly, field studies have shown that a combined application of an imidazolinone herbicide with prohexadione resulted in reduced emergence of O. cumana compared with the sole application of the herbicide. The aim of this study was to investigate whether prohexadione is herbicidal to O. cumana.

Prohexadione was rapidly distributed within the sunflower, reaching the roots, the site of O. cumana attack, as early as 6 h after application (HAA) on sunflower leaves. A direct impact of prohexadione on O. cumana girect impact on O. link3 cumana germination. © 2020 Society of Chemical Industry.
Azadirachtin has the potential to be used for pest control. Nevertheless, few studies have investigated the effects of azadirachtin on the cognitive behavior of pests. In this study, expression of the cAMP response element-binding protein (CREB) and its gene were studied via a series of experiments in Spodoptera litura larvae treated with azadirachtin.

RNA-Seq analysis of S. litura larvae treated with azadirachtin was undertaken. According to Kyoto Encyclopedia of Genes and Genomes analysis, the top 20 enriched pathways included neuroactive ligand-receptor interaction pathways, with seven significantly differentially expressed genes including CREB. Quantitative real time polymerase chain reaction (qRT-PCR) results indicated that the CREB gene was expressed during all developmental stages of S. litura, but relative expression of the CREB gene was significantly downregulated after treatment with azadirachtin. Grayscale statistical analysis also showed that expression levels of protein kinase A (PKA), extracellular signal-regulated kinase (ERK) and CREB proteins were significantly downregulated after treatment with azadirachtin. Moreover, RNA interference results showed that the effect of azadirachtin on the cognitive behavior of S. litura was consistent with that seen after interfering with CREB. In addition, larval selectivity to addictive odor sources was reduced, and the initial reaction time was increased.

This study clarified that azadirachtin can affect the cognitive behavior of S. litura and treatment with azadirachtin resulted in a downregulation of gene and protein expression of CREB and its pathway proteins. © 2020 Society of Chemical Industry.

Whether anemic cats and dogs with increased left heart dimensions are at higher risk of transfusion-associated circulatory overload, and the effect of blood transfusion on left heart dimensions in naturally occurring anemia is unknown.

To evaluate the effect of blood transfusion on left heart dimensions in clinically relevant anemia.

Twenty dogs and 20 cats presenting to a university veterinary teaching hospital.

In this prospective observational study, anemic dogs and cats requiring blood transfusion were included. Packed cell volume (PCV), total solids, and echocardiography were performed before and within 24 hours of blood transfusion. Signalment, bodyweight, disease process, transfusion duration and volume, and prior treatments were recorded. Nonparametric statistics were reported as median [range]. Post hoc Bonferroni correction set significance at P < .006.

After transfusion, PCV increased in cats (12% [6-16] to 18% [10-33], P = .001) and dogs (14% [7-24] to 25% [9-37], P = .001), heart rate decreased in dogs (104 bpm [86-166] to 87 bpm [56-138], P < .001), and fractional shortening decreased in cats (57.1% [36.0-84.7] to 41.0% [28.1-69.6], P = .002) and dogs (33.79% [19.33-62.79] to 31.89% [19.06-51.47], P = .006). Left ventricular internal diameter in systole increased in cats (6.5 mm [2.7-9.8] to 7.9 mm [5.3-11.1], P = .001). Normalized left ventricular internal diameter in diastole (1.48 [1.25-1.79] to 1.57 [1.33-2.00], P = .001) and systole (0.87 [0.58-1.19] to 1.00 [0.74-1.36], P = .001) increased in dogs. Incidence of volume overload did not differ before (14/20 cats, 70%; 9/20 dogs, 45%) or after (12/20 cats, 60%; 11/20 dogs, 55%) transfusion (P = .64).

Blood transfusion is well tolerated when signs of volume overload are present before transfusion.

Long-term placement of lumen apposing metal stents (LAMS) with high lumen apposing force may result in adverse events. The aim of the current study was to assess the long-term efficacy and safety of a self-approximating LAMS with lower lumen apposing force for endoscopic ultrasound-guided choledochoduodenostomy (EUS-CDS) and -gallbladder drainage (EUS-GBD).

Five Asian institutions participated in this study. Consecutive patients suffering from obstructive jaundice with failed ERCP or acute cholecystitis that were at high risk for cholecystectomy were recruited. We evaluated the technical and clinical success rates, adverse events rates, types of interventions through the stent and the patency profile.

From June 2017 to Oct 2018, a total of 53 patients received EUS-CDS (26) and EUS-GBD (27). The technical and clinical success rates were similar between the two groups (88.5% vs 88.9%, P=1 and 88.5% vs 88.9%, P=1 respectively). The differences in 30-day mortality rates [2 (7.7%) vs 2 (7.7%), P=1] and adverse events [3 (11.5%) vs 3 (11.5%), P=1] did not reach significance. Regarding long-term outcomes, two patients in each group suffered from adverse events (P=1). One patient in the EUS-GBD group who was on direct oral anticoagulant suffered from stent induced bleeding.

The self-approximating LAMS with lower lumen apposing force was effective and safe with a low risk of buried stent syndrome and bleeding in the longer term. The ClinicalTrials.gov Identifier was NCT03002051.

To explore the relations between asthma, allergies and mental health problems in preschool children, aged 3-5years.

In this cross-sectional Swedish study, we used data on 4649 children in Uppsala municipality whose parents and preschool teachers had responded to questions measuring asthma and allergies, and the Strengths and Difficulties Questionnaire (SDQ) for assessment of mental health problems. Logistic regression models were used to explore the relations between asthma and allergies and mental health problems.

Children with asthma (8.5%) had elevated odds of having emotional symptoms as rated by parents (OR 1.34; 1.02-1.76) and teachers (OR 1.44; 1.09-1.91). According to parents' ratings, these children also had elevated odds of showing mental health problems in general according to the SDQ total score (OR 1.42; 1.05-1.94). Children with food allergies or intolerance (4.4%) only had elevated odds of having emotional symptoms (OR 1.64; 1.16-2.33), as reported by parents. These results remained significant after adjustment for parental background factors.
