Given the increase in exposure to BPA alternatives, continuous biomonitoring, and further investigation of their health effects through prospective cohort studies are warranted.Nitrosamines, as ubiquitous environmental carcinogens with adverse impact on human health, were crucial inducers of esophageal cancer (EC). Esophageal inflammation (EI) was an important biological process and considered to be associated with the progression of EC. However, the underlying regulatory mechanism of EI process caused by nitrosamines exposure remained largely unclear. In this study, a metabolomics approach based on mass spectrometry was utilized to explore the effect of nitrosamines exposure to ICR mice. Also, the changes of pivotal metabolic enzyme levels, urinary nitrosamines and histopathological analysis were evaluated. The results showed that nitrosamines exposure was intimately interrelated with EI process in mice. Metabolomics profiling analysis indicated that nitrosamines caused significant alterations of metabolic pathway predominantly enriched in fatty acid metabolism. Targeted metabolomics analysis revealed that nitrosamines promoted decomposition of fatty acids and facilitated fatty acid β-oxidation (FAO) of mice. The significant increase of carnitine palmitoyltransferase 1 (CPT1) and downregulation of acetyl-CoA acyltransferase 2 (ACAA2) would promote FAO in EI process induced by nitrosamines. Additionally, the exposure levels of more than half of nitrosamines in urine were correlated with inflammatory fatty acid biomarkers. Overall, this study found that EI triggered by nitrosamines may be associated with the promotion of FAO, and provided novel insights for evaluating the underlying mechanism of environmental pollutant-caused toxicity based on metabolomics.Titanium dioxide nanoparticles (nTiO2) are widely used in numerous products, yet their role in the accumulation and transfer of other contaminants in the aquatic food webs is not well understood. The influence of nTiO2 on inorganic (IHg) and monomethyl mercury (MeHg) accumulation in invertebrate Daphnia magna through waterborne and dietary exposure was thus thoroughly investigated. The results showed that nTiO2 led to a substantial decrease of the total mercury body burden (THg) in D. magna in direct waterborne exposure to IHg/MeHg. However, exposure to nTiO2 pre-treated with IHg/MeHg resulted in an increase of the THg body burden in daphnids. The presence of nTiO2 led to a substantial decrease of the THg in D. magna when exposed to IHg/MeHg via algal food. These effects were more pronounced for IHg than that for MeHg due to the higher adsorption capabilities of nTiO2 for IHg. In addition, high concentrations of nTiO2 favored the trophic transfer of IHg/MeHg through feeding on nTiO2 pre-treated with Hg, however lessened it when D. magna were fed on alga pre-treated with IHg/MeHg. Comparable assimilation efficiency (AE), determined as Hg retained in daphnids after depuration, was observed in D. magna when exposed to IHg/MeHg via algal food regardless the absence or presence of 20 mgL-1 nTiO2. By contrast, an increase of the AE of MeHg through feeding on nTiO2 and alga was found in the presence of higher concentration of 200 mgL-1 nTiO2. The present results will help to better understand the role of nTiO2 on bioavailability and trophic transfer of global contaminants, such as mercury, known to bioaccumulate and biomagnify in the aquatic environment.Glycoprotein hormones are formed by the heterodimerization of alpha and beta subunits. In vertebrates, there are five glycoprotein hormones, four of which have a common alpha subunit (GPA1) bound to a specific beta subunit (GPB1, GPB2, GPB3, or GPB4), and the fifth, thyrostimulin, is formed by the dimerization of GPA2 and GPB5 subunits. These hormones mediate physiological events such as development, metabolism, and reproduction, although the functional role of thyrostimulin in vertebrates has not been fully elucidated. Recent reports in invertebrates, specifically in holometabolous insects, suggest that GPA2/GPB5 plays a critical role in development, diuresis, and reproduction. C-reactive protein (CRP) acts as early defence system in response to these infections. This study investigated role of CRP single-nucleotide polymorphism (SNP) genotypes and protein levels towards DENV/CHIKV mono and co-infection among eastern Indian patients. 128 DENV-CHIKV co-infected, 206 DENV and 167 CHIKV mono-infected patients were subjected to genotyping of two CRP SNPs by PCR-RFLP along with 102 healthy individuals. CRP levels were determined by immunoturbidimetry. Statistical correlation of CRP genotypes with CRP concentration, DENV-CHIKV mono/co-infection and viral load was performed. Patients with rs3093059-CT and rs3091244-TT were more susceptible to DENV-CHIKV co-infection, whereas, rs3091244-CT might have imparted protection against CHIKV mono-infection. DENV-HVL was more prevalent within rs3093059-TT and rs3091244-CT co-infected patients, whereas, CHIKV-HVL among rs3091244-CC. Acute phase co-infected patients had significantly higher CRP level compared to mono-infections. Both mono and co-infected patients with aches/pain exhibited 2-3-fold higher CRP levels compared to those without. rs3093059-CT and rs3091244-CT co-infected patients had higher CRP concentration compared to rs3093059-TT and rs3091244-CC, respectively. Co-infected patients with WHO-defined warning signs had higher anti-dengue IgG/IgM ratio and serum CRP level compared to those without warning signs. Thus, patient's CRP genotype might play significant role in determining serum-CRP concentration, viral load and DENV-CHIKV mono/co-infection.Bovine alphaherpesviruses 1 and 5 (BoHV-1 and BoHV-5) are closely related viruses that co-circulate in South America and recombine in the field. The complete genomes of three natural gB gene recombinant viruses between BoHV-1 and BoHV-5 were obtained by Illumina next-generation sequencing. Complete genome sequences of the three recombinant strains (RecA1, RecB2, and RecC2) have a similar size of approximately 138.3kb and a GC content of 75%. The genome structure corresponds to herpesvirus class D, with 69 open reading frames (ORFs) arranged in the same order as other bovine alphaherpesviruses related to BoHV-1. Their genomes were included in recombination network studies indicating statistically significant recombination evidence both based on the whole genome, as well as in the sub-regions. Our findings highlight the importance of studying whole genome diversity in the field and determining the role that homologous recombination plays in the structure of viral populations. A whole-genome recombinant characterization is a suitable tool to help understand the emergence of new viral forms with novel pathogenic features.
COVID-19 is a worldwide pandemic representing the most challenging global health crisis currently. Screening tests availability are a problematic task due to resource-limited abilities of some countries using RT-qPCR technique for SARS-COV-2 detection.

To cope with these health emergencies, in particular with this COVID-19 pandemic, states with low molecular diagnostic resources must optimize their capacity in molecular tests. We aimed to design a simple and effective strategy to improve inputs in the RT-qPCR tests as we attempted to check the financial advisability of using such an approach by calculating reduction rate of the test unit cost.

The used RNA was taken from suspected Covid-19 positive people. Nasopharyngeal swabs were collected at Pasteur Institute Diagnostic Center, Constantine, Algeria, 2020. We have optimized a screening strategy by grouping 16 individuals per pool, without reducing the sensitivity of RT-qPCR.

A 1/16 dilution of a positive sample was a practical limit that does not require the use of robotic systems or mathematical modeling to construct the pools. The financial analysis of our strategy has shown that the costs can be reduced to 90 %. The pooled testing strategy that was proven in this study could be recommended to help COVID-19 containment in countries with low potential screening infrastructures using RT-qPCR technique by reducing the number of tests required to identify all positive subjects.
