These promising results should be further evaluated in experimental models in vivo.Xanthomonas oryzae pv. oryzae (Xoo), the causative agent of bacterial blight, is one of the major threats to rice productivity. Yet, the molecular mechanism of rice-Xoo interaction is elusive. Here, we report comparative proteome profiles of Xoo susceptible (Dongjin) and resistant (Hwayeong) cultivars of rice in response to two-time points (3 and 6 days) of Xoo infection. Low-abundance proteins were enriched using a protamine sulfate (PS) precipitation method and isolated proteins were quantified by a label-free quantitative analysis, leading to the identification of 3846 proteins. Of these, 1128 proteins were significantly changed between mock and Xoo infected plants of Dongjin and Hwayeong cultivars. Based on the abundance pattern and functions of the identified proteins, a total of 23 candidate proteins were shortlisted that potentially participate in plant defense against Xoo in the resistant cultivar. Of these candidate proteins, a mitochondrial arginase-1 showed Hwayeong specific abundance and was significantly accumulated following Xoo inoculation. Overexpression of arginase 1 (OsArg 1) in susceptible rice cultivar (Dongjin) resulted in enhanced tolerance against Xoo as compared to the wild-type. In addition, expression analysis of defense-related genes encoding PR1, glucanase I, and chitinase II by qRT-PCR showed their enhanced expression in the overexpression lines as compared to wild-type. Taken together, our results uncover the proteome changes in the rice cultivars and highlight the functions of OsARG1 in plant defense against Xoo.
Pathogenic variants in SCN8A have been demonstrated with a wide spectrum of epilepsy phenotypes, ranging from benign infantile epilepsy (BIE), to early onset developmental and epileptic encephalopathy (DEE) with moderate to severe developmental delay. In order to provide further insight on the spectrum of SCN8A-related epilepsy, we aimed to explore the clinical and genetic phenotype in Chinese children.

A cohort of fifty Chinese patients with SCN8A-related disorders was included in the retrospective study. Genetic and clinical features and treatment effect of patients were further assessed based on phenotype parameters. The pathogenicity of variants was classified using the next-generation sequencing variation study.

We found 50 patients who presented with severe developmental and epileptic encephalopathy (DEE, 70%), benign infantile epilepsy (BIE, 12%), developmental encephalopathy with epilepsy (16%), and severe developmental delay without epilepsy (2%). The seizure onset age ranged from 1 day to 1 yevelopmental delay without epilepsy to severe DEE. Three new variants were associated with SCN8A-BIE. Sodium channel blockers were effective in treating seizures for some SCN8A-related disorders however may not be relevant to the mutant location.Bone morphogenetic protein 15 (BMP15) and follicle-stimulating hormone (FSH) both play important roles in mammalian ovary and follicular development. The aim of the present study is to investigate the effects of BMP15 and FSH in the prepubertal ovary of Rongchang pigs considering a possible signaling mechanism involving TβRII/ SMAD4 and FSHR in granulosa cells. For this purpose, we quantified expression levels of BMP15, SMAD2, SMAD3, SMAD4, SMAD7, TGF-β1, TGF-β2, TGF-β3, TGFβRI, TGFβRII, and FSHR via qRT-PCR at different ages in prepubertal ovaries and cultured biopsy of 90-day-old ovary in Rongchang pig. Additionally, the protein levels of BMP15, FSHR, SMAD2, SMAD4, TGFβRI, TGFβRII, TGF-β1, TGF-β2 were quantified via Western blot and the localizations of BMP15, FSHR and TGFβRII were observed via immunofluorescence confocal microscope. The results showed that expression levels of BMP15, TGF-β1, TGFβRII and FSHR increased significantly at day 60 as compared to day 30 and reached peak value at day 90 in prepubertal ovary of Rongchang pigs. We observed that BMP15, TGFβRII and FSHR was highly presented, which TGFβRII and FSHR displayed co-localization in the follicles of the prepubertal ovaries of 90-day-old Rongchang gilts. Treatment with TGFβRI/II inhibitor LY2109761 significantly decreased the expression of TGFβRI, TGFβRII and SMAD4 and TGFβRI inhibitor LY2157299 decreased TGFβRI, but increased the TGFβRII, SMAD4 and FSHR expression levels. Furthermore, the addition of rBMP15 and rFSH group significantly increased the expression of TGFβRII and FSHR proteins (P 0.05) was observed by Western blot. In conclusion, BMP15, TGFβRII and FSHR were increased significantly in the prepubertal ovarian follicles of Rongchang pigs and FSHR expression in GCs was regulated by BMP15 and FSH through the TGFβRII.The COVID-19 pandemic has placed unprecedented stress on US acute care hospitals, leading to overburdened ICUs. It remains unknown if increased COVID-19 ICU occupancy is crowding out non-COVID-related care and whether hospitals in vulnerable communities may be more susceptible to ICUs reaching capacity. Using facility-level hospitalization data, we conducted a retrospective observational cohort study of 1753 US acute care hospitals reporting to the US Department of Health and Human Services Protect database from September 4, 2020 to February 25, 2021. 63% of hospitals reached critical ICU capacity for at least two weeks during the study period, and the surge of COVID-19 cases appeared to be crowding out non-COVID-19-related intensive care needs. Hospitals in the South (OR = 3.31, 95% CI OR 2.31-4.78) and West (OR = 2.28, 95% CI OR 1.51-3.46) were more likely to reach critical capacity than those in the Northeast, and hospitals in areas with the highest social vulnerability were more than twice as likely to reach capacity as those in the least vulnerable areas (OR = 2.15, 95% CI OR 1.41-3.29). The association between social vulnerability and critical ICU capacity highlights underlying structural inequities in health care access and provides an opportunity for policymakers to take action to prevent strained ICU capacity from compounding COVID-19 inequities.One of the most important areas of medical science is oncology, which is responsible for both the diagnostics and treatment of cancer diseases. Simultaneously one of the main challenges of oncology is the development of modern drugs effective in the fight against cancer. Statins are a group of biologically active compounds with the activity of 3-hydroxy-3-methyl glutaryl-CoA reductase inhibitors, an enzyme catalyzing the reduction of 3-hydroxy-3-methyl-glutaryl-CoA (HMG-CoA) to mevalonic acid. By acting on this enzyme, statins inhibit the endogenous cholesterol synthesis which in turn causes the reduction of its systemic concentrations. However, in vitro and in vivo studies confirm also the cytostatic and cytotoxic effects of statins against various types of cancer cells including colon cancer. In the presented studies the influence of mevastatin on cancerous colon cells CaCo-2 by Raman spectroscopy and imaging is discussed and compared with biochemistry characteristic for normal colon cells CCD-18Co. Based on vibrational features of colon cells normal cells CCD-18Co, cancerous cells CaCo-2 and cancerous cells CaCo-2 treated by mevastatin in different concentrations and incubation times we have confirmed the influence of this statin on biochemistry composition of cancerous human colon cells. Moreover, the spectroscopic results for colon normal cells and cancerous cells based on data typical for nucleic acids, proteins, lipids have been compared. The cytotoxisity of mevastatin was determined by using XTT tests.Molecular tellurium (Te2) offers a wide range of potential applications, including frequency reference. Spectroscopic results about the 130Te2 spectrum were previously reported, but few lines lie around the PbF transition of A (2Σ+) (υ' = 0) ← X12Π1/2 (υ = 0) (∼444.4 nm) which is regarded as an eEDM (electron's Electric Dipole Moment) sensitive transition. Here electronic transition lines for 130Te2 were determined using the Saturated Absorption Spectroscopy method, where the origin of the transition band was investigated for the B1 (3Σu-)0u+ (υ' = 10) ← X1 (3Σg-)0g+ (υ = 5) transition. The Dunham model with high orders was utilized to assign these transition lines, while Dunham parameters of the excited state B1 (υ' = 10) were updated to a new level. The Toptica TA-SHG Pro laser was then locked to a single absorption line using a P-I servo. Our results here not only provide the assigned atlas of Te2 spectrum near 444.4 nm, but also contribute to a stable and sensitive spectroscopic detection of PbF molecules toward the eEDM measurement, which is significant in understanding the fundamental physics beyond the Standard Model.Nitric oxide (NO), an essential biological messenger molecule, participates in various physiological and pathological processes. The sensitive and specific detection of NO is of great significance for understanding the biological function of NO. Here, we synthesized a fluorescent probe (Rho-NO) for highly selective detection of NO both in vitro and in vivo. The high selectivity of Rho-NO is attributed to the fact that NO is easily replaced by electron donor amino group to form N-nitrosation products, causing rhodamine spiro ring open and fluorescence emit. Rho-NO showed a good linear response to NO (0-100 µM) with a low detection limit (0.06 µM). Importantly, it exhibited excellent specificity for NO detection in human serum and was also applied for imaging NO in living cells and inflammatory model of zebrafish. This work proves the potential of Rho-NO in pathological research and disease diagnosis.This study determined the ratio between glenohumeral and three-dimensional scapular motion during arm elevation and lowering in 91 individuals without shoulder pain. Scapular kinematics were assessed using an electromagnetic tracking device. Individuals performed 3 repetitions of elevation and lowering of the arm in the sagittal plane. Two-way ANOVAs (interval 30-60°, 60-90°, 90-120° x phase elevation and lowering) and paired t-tests were used for data analysis. For scapular internal/external rotation, lesser scapular internal rotation contribution was found during the 60-90° interval as compared to the 90-60° interval. Lesser scapular external rotation was identified in the 60-30° interval of arm lowering. The ratio was greater during arm elevation (1.89) compared to lowering (1.74) across the entire motion arc. For scapular upward rotation, greater upward rotation contribution was observed during arm elevation at the 30-60° interval, and less scapular downward rotation contribution in the final range of arm lowering. For scapular tilt, lesser scapular posterior tilt contribution during arm elevation was observed compared to arm lowering. The ratios between glenohumeral elevation/lowering and each individual scapulothoracic motion showed either differences between intervals and/or between elevation and lowering during specific intervals in healthy individuals.
