Waterborne diseases cause millions of deaths worldwide, especially in developing communities. The monitoring and rapid detection of microbial pathogens in water is critical for public health protection. This study reports the development of a proof-of-concept portable pathogen analysis system (PPAS) that can detect bacteria in water with the potential application in a point-of-sample collection setting. A centrifugal microfluidic platform is adopted to integrate bacterial cell lysis in water samples, nucleic acid extraction, and reagent mixing with a droplet digital loop mediated isothermal amplification assay for bacteria quantification onto a single centrifugal disc (CD). Coupled with a portable "CD Driver" capable of automating the assay steps, the CD functions as a single step bacterial detection "lab" without the need to transfer samples from vial-to-vial as in a traditional laboratory. The prototype system can detect Enterococcus faecalis, a common fecal indicator bacterium, in water samples with a single touch of a start button within 1 h and having total hands-on-time being less than 5 min. An add-on bacterial concentration cup prefilled with absorbent polymer beads was designed to integrate with the pathogen CD to improve the downstream quantification sensitivity. All reagents and amplified products are contained within the single-use disc, reducing the opportunity of cross contamination of other samples by the amplification products. This proof-of-concept PPAS lays the foundation for field testing devices in areas needing more accessible water quality monitoring tools and are at higher risk for being exposed to contaminated waters.Due to their relatively large production and few restrictions on uses, novel substitutes for historically used per and poly-fluoroalkyl substances (PFAS) are being used and accumulating in the environment. However, due to a lack of information on their toxicological properties their hazards and risks are hard to estimate. Before fertilization, oocytes of two salmonid species, Arctic Char (Salvelinus alpinus) and Rainbow Trout (Oncorhynchus mykiss), were exposed to three PFAS substances used as substitutes for traditional PFAS, PFBA, PFBS or GenX or two archetypical, historically used, longer-chain PFAS, PFOA and PFOS. Exposed oocytes were subsequently fertilized, incubated and were sampled during several developmental stages, until swim-up. All five PFAS were accumulated into egg yolks with similar absorption rates, and their concentrations in egg yolks were less than respective concentrations in/on egg chorions. Rapid elimination of the five PFAS was observed during the first 3 days after fertilization. Thereafter, amounts of PFOS and PFOA were stable until swim-up, while PFBA, PFBS and GenX were further eliminated during development from one month after the fertilization to swim-up. In these two salmonid species, PFBA, PFBS and GenX were eliminated faster than were PFOS or PFOA.Tree species diversity promotes multiple ecosystem functions and services. However, little is known about how above- and belowground resource availability (light, nutrients, and water) and resource uptake capacity mediate tree species diversity effects on aboveground wood productivity and temporal stability of productivity in European forests and whether the effects differ between humid and arid regions. We used the data from six major European forest types along a latitudinal gradient to address those two questions. We found that neither leaf area index (a proxy for light uptake capacity), nor fine root biomass (a proxy for soil nutrient and water uptake capacity) was related to tree species richness. Leaf area index did, however, enhance productivity, but negatively affected stability. Productivity was further promoted by soil nutrient availability, while stability was enhanced by fine root biomass. We only found a positive effect of tree species richness on productivity in arid regions and a positive effect on stability in humid regions. This indicates a possible disconnection between productivity and stability regarding tree species richness effects. In other words, the mechanisms that drive the positive effects of tree species richness on productivity do not per se benefit stability simultaneously. Our findings therefore suggest that tree species richness effects are largely mediated by differences in climatic conditions rather than by differences in above- and belowground resource availability and uptake capacity at the regional scales.The present work aimed to study the effect of ozonation on the organic sum parameters linked to enhanced biodegradability. Laboratory experiments were conducted with the effluent of four Austrian urban wastewater treatment plants with low food to microorganism ratios and different matrix characteristics. Biochemical oxygen demand over 5 days (BOD5) was measured before ozonation and after application of different specific ozone doses (Dspec) (0.4, 0.6 and 0.8 g O3/g DOC). Other investigated organic parameters comprised chemical oxygen demand (COD), dissolved organic carbon (DOC), UV absorption at 254 nm (UV254), which are parameters that are applied in routine wastewater analysis. Carbamazepine and benzotriazole were measured as reference micropollutants. The results showed a dose-dependent increase in biological activity after ozonation; this increase was linked to the enhanced biodegradability of substances that are recalcitrant to biodegradation in conventional activated sludge treatment. The highest relatiement in addition to micropollutant abatement.A comprehensive emission inventory of the transport sector through fuel-based emission factors (EFs) was developed for the first time in Nepal. This study estimates air pollutants emission from diesel vehicles between the years 1989 and 2018 based on national statistical data, average vehicle kilometers travelled, fuel mileage, and measurement-based EFs for each vehicle category during idle and moving conditions. The consumption of diesel by vehicle category was also estimated and total consumption was compared with national sales data. The Monte Carlo was used to estimate uncertainties. Nationally, total diesel consumption was estimated as 892,770 kL (85-115%) in 2017/18, 13.4 times higher than 1989/90. Ratnoze1 and Microaeth were used to conduct the tail pipe emission measurements. The fuel-based EFs of CO2, CO, BC, and PM2.5 were calculated through the carbon mass balance method. Of all diesel vehicles measured (n = 29) during idling, the average EFs were estimated as CO2 2600 (99-101%), CO 33.3 (44-156%), BC 0.6 (25-101%), and PM2.5 5.2 (0-235%) in unit of g L-1. For moving conditions (n = 5), the average EFs were estimated to be CO2 2476 (90-110%), CO 97.3 (0-232%), BC 1.7 (46-110%), and PM2.5 20.7 (0-255%), all in g L-1. Multiplying fuel consumption by EFs, national air pollutant emissions were estimated as 2214 (90-110%) to 2781(85-115%) for CO2, 27.7 (42-158%) to 88.8 (0-232%) for CO, 0.51 (23-177%) to 3.55 (46-110%) for BC and 3.42 (0-236%) to 23.47 (0-255%) for PM2.5 in 2017/18 in unit of Gg. This paper recommends revising national vehicle mass emission standards based on the findings of this study and including and enhancing sustainable low-carbon transport through amendment of transport policy.The UV/Cl2 process (also known as chlorine photolysis, which is the combination of chlorine and simultaneous irradiation of UV light) is conventionally applied at acidic mediums for drinking water treatment and further treatment of wastewater effluents for secondary reuse. This is because the quantum yield of HO• from HOCl (ϕHO•, 254 = 1.4) is greater than the one from OCl- (ϕHO•, 254 = 0.278) by approximately 5 times. Moreover, chlorine photolysis in acidic mediums also tends to have lower radical quenching rates than that of their alkaline counterparts by up to 1000 times. The aim of this research is to investigate the applicability of the UV/Cl2 process by assessing its efficacy on the removal of trimethoprim (TMP) at not only acidic to neutral conditions (pH 6-7), but also alkaline mediums (pH 8-9). At alkaline pH, free chlorine exists as OCl- and since OCl- has a higher molar absorption coefficient as compared to HOCl at higher wavelengths, there would be higher reactive chlorine species (RCS) formation and contribution. TMP removal followed pseudo-first order kinetics and depicted that a maximum fluence based constant (kf' = 0.275 cm2/mJ) was obtained using 42.25 μM (3 mg/L) of chlorine at pH 9, with an irradiation of 275 nm. At alkaline conditions, chlorine photolysis performance followed the trend of UV (275)/Cl2 > UV (265)/Cl2 > UV (310)/Cl2 > UV (254)/Cl2. RCS like Cl•, Cl2-• and ClO• contributed to the degradation of TMP. When the pH was increased from 6 to 8, contribution from hydroxyl radicals (HO• ) was decreased whilst that of RCS was increased. Application of UV (310)/Cl2 had the highest HO• generation, contributing to TMP removals up to 13% to 48% as compared to 5% to 27% in UV (254, 265, 275)/Cl2 systems at pH 6-9. Artificial neural networks modelling was found to be able to verify and predict the contribution of HO• and RCS conventionally calculated via the general kinetic equations in the UV/Cl2 system at 254, 265, 275 and 310 nm.Water-, energy-, and food (WEF) related practices, such as low impact development (LID), residential solar panels, and rooftop urban agriculture, have been applied to improve urban sustainability and resilience under climate change and urbanization. However, most practices require space. This requirement may result in competition for land. In addition, not all newly built practices benefit the environment from the life cycle perspective. Therefore, this study aims to develop a systematic WEF-related practice planning method to improve urban sustainability and resilience in a limited space. The core method is a multi-objective optimization model that considers the performance and environmental impacts of the selected practices. The assessment was conducted in a densely populated area in Taipei, the capital city of Taiwan, to describe the planning processes and demonstrate the feasibility of the methods. In the Taipei case, five goals were defined the supply of WEF, the sponge city development target, and the greenhouse gas reduction target. The optimal results of the multi-objective optimization model indicated the closeness of the optimal implementation of WEF-related practices to achieving the goals. The results showed that the optimal arrangement of WEF-related practices could provide water supply benefits and was favorable for developing a sponge city. According to the sensitivities, to achieve urban sustainability and resilience, the priorities in order of importance are as follows establish a rainwater harvesting system for buildings, encourage the implementation of rooftop photovoltaic systems, and improve the materials and processes used solar panel and bioretention cell production. The systematic planning method provides a quantitative assessment and delivers practical cross-sectoral integrated strategies for decision-making.Social and environmental concern about the use of crumb rubber from end-of-life car tires in the construction of different sport and recreational facilities is increasing due to the presence of hazardous compounds. The aim of this research was the assessment of 42 organic chemicals, including polycyclic aromatic hydrocarbons (PAHs), phthalates, adipates, antioxidants and vulcanisation agents in a large number of infill samples (91) from synthetic turf football pitches of diverse characteristics and geographical origin. Samples were taken worldwide, in 17 countries on 4 continents, to show the global dimension of this problem. Ultrasound assisted extraction was employed to extract the target compounds, followed by gas chromatography coupled to tandem-mass spectrometry (UAE-GC-MS/MS). Seventy-eight crumb rubber samples as well as thirteen samples of alternatives materials, such as cork granulates, thermoplastic elastomers and coconut fibre, were analyzed. The results highlight the presence of all target PAH in most rubber samples at concentrations up to μg g-1, including the eight ECHA (European Chemicals Agency) PAHs considered as carcinogenic, and anthracene (ANC), pyrene (PYR) and benzo[ghi]perylene (B[ghi]P), catalogued as substances of very high concern (SVHC).
