Why we use Method Controls- to verify the media, method, equipment and technique used in the testing of samples. Ensures the accuracy and reliability of results reported by the lab.
-Three Ways of Accuracy:
-Culture Collection, makes sure the culture is still able to be used.
-Accuracy Monitoring, proving the assay is within desired frame each time testing occurs.
-Estimation of Uncertainty

Method Controls must always be done inside the Positive Control Lab.
-Must be placed within a secondary container, must be bleached prior to leaving.
-Can be incubated with client samples, but must be placed upon a lower shelf to prevent contamination.
-Area must be bleached down before leaving.

PPE-Disposable equipment, coat, shoe covering, gloves. Cannot weigh out client samples or make media for the rest of the day. Equipment must be disposed of in Posco. Must wash hands in sink in Posco.

Method Controls are set up if that particular test was ordered that day. (Always APC, always Y/M, always Salm with Lac, always List with Bleb, UVM, DMFR)

Plating counts must be kept at a level of 25-250 CFU per plate, and 0.3 to 110 for MPN methods.
