High-resolution solar observations show the complex structure of the magnetohydrodynamic (MHD) wave motion. We apply the techniques of proper orthogonal decomposition (POD) and dynamic mode decomposition (DMD) to identify the dominant MHD wave modes in a sunspot using the intensity time series. Here, we present a novel method that employs machine learning techniques to identify the underlying components present within observed spectral lines, before subsequently constraining the constituent profiles through single or multiple Voigt fits. Our method allows active and quiescent components present in spectra to be identified and isolated for subsequent study. Lastly, we employ a Ca ɪɪ 8542 Å spectral imaging dataset as a proof-of-concept study to benchmark the suitability of our code for extracting two-component atmospheric profiles that are commonly present in sunspot chromospheres. Here, we employ a simulation featuring propagating MHD waves within a flux tube with a known driver and atmospheric parameters. We invert the Stokes profiles for the 6301 Å and 6302 Å line pair emergent from the simulations using the well-known Stokes Inversions from Response functions code to see if the atmospheric parameters can be returned for typical spatial resolutions at ground-based observatories. The inversions return synthetic spectra comparable to the original input spectra, even with asymmetries introduced in the spectra from wave propagation in the atmosphere. The output models from the inversions match closely to the simulations in temperature, line-of-sight magnetic field and line-of-sight velocity within typical formation heights of the inverted lines. Deviations from the simulations are seen away from these height regions. The inversions results are less accurate during passage of the waves within the line formation region. The original wave period could be recovered from the atmosphere output by the inversions, with empirical mode decomposition performing better than the wavelet approach in this task. TRPV1 signaling has been implicated in bladder pain and is a potential analgesic target. Resiniferatoxin is the most potent known agonist of TRPV1. Acute exposure of the rat bladder to resiniferatoxin has been demonstrated to result in pain-related freezing and licking behaviors that are alleviated by virally encoded IL-4. The interleukin-4-inducing principle of Schistosoma mansoni eggs (IPSE) is a powerful inducer of IL-4 secretion, and is also known to alter host cell transcription through a nuclear localization sequence-based mechanism. We previously reported that IPSE ameliorates ifosfamide-induced bladder pain in an IL-4- and nuclear localization sequence-dependent manner. We hypothesized that pre-administration of IPSE to resiniferatoxin-challenged mice would dampen pain-related behaviors. IPSE indeed lessened resiniferatoxin-triggered freezing behaviors in mice. This was a nuclear localization sequence-dependent phenomenon, since administration of a nuclear localization sequence mutant version of IPSE abrogated IPSE's analgesic effect. In contrast, IPSE's analgesic effect did not seem IL-4-dependent, since use of anti-IL-4 antibody in mice given both IPSE and resiniferatoxin did not significantly affect freezing behaviors. RNA-Seq analysis of resiniferatoxin- and IPSE-exposed bladders revealed differential expression of TNF/NF-κb-related signaling pathway genes. In vitro testing of IPSE uptake by urothelial cells and TRPV1-expressing neuronal cells showed uptake by both cell types. Noninvasive clinical measures within the scopes of practice of those professions sensitive to cognitive impairment are needed. Purpose This study examined the differences of probable mild cognitive impairment (MCI) on latency and mean amplitude of the P3b auditory event-related potential. Method Fifty-four participants comprised two groups according to cognitive status (cognitively normal older adults [CNOA], n = 25; probable MCI, n = 29). P3b was recorded using an oddball paradigm for speech (/ba/, /da/) and non-speech (1000, 2000 Hz) stimuli. Amplitudes and latencies were compared from six electrodes (FPz, Fz, FCz, Cz, CPz, Pz) between groups across stimulus probability and type. Results CNOA participants had larger P3b mean amplitudes for deviant stimuli than those with probable MCI. Group effects of latency were isolated to deviant stimuli at FCz only when those with unclear P3bs were included.
