In the present work, the determination of omeprazole (OME) enantiomers in oral fluid and plasma samples was carried out utilizing microextraction by packed sorbent (MEPS) and liquid chromatography-tandem mass spectrometry. A chiral column with cellulose-SB phase was used for the first time for enantiomeric separation of OME with an isocratic elution system using 0.2% ammonium hydroxide in hexane-ethanol mixture (70  30, v/v) as the mobile phase. OME enantiomers were determined utilizing a triple quadrupole tandem mass spectrometer in positive ion mode (ESI+) monitoring mass transitions m/z 346.3 ⟶ 198.0 for OME and m/z 369.98 ⟶ 252.0 for internal standard. The limits of detection and quantification of the present method for both enantiomers were 0.1 and 0.4 ng/mL, respectively. The method validation provided good accuracy and precision. The matrix effect factor was less than 5%, and no interfering peaks were observed. The interday precision values ranged from 2.2 to 7.5 (%RSD), and the accuracy of determinations varied from -9.9% to 8.3%. In addition, the pharmacokinetics (PK) of omeprazole enantiomers in healthy subjects after a single oral dose was investigated. (S)-Enantiomers showed higher levels than (R)-enantiomers throughout 24 h. It was found that the mean maximum concentrations of (R)- and (S)-omeprazole in plasma samples were about two times higher than in oral fluid.Peracetic acid (PAA) is widely used as a sterilizing/disinfecting agent, and, in endodontics, it has been introduced as a promising irrigant in root canal treatment. It has been used at different concentrations to achieve various functions. However, endodontic instruments in contact with PAA of a certain concentration may affect their fatigue resistance. Therefore, the aim of this study was to investigate the impact of PAA on the cyclic fatigue resistance of three commercial heat-treated nickel-titanium (NiTi) rotary files. Three types of heat-treated NiTi rotary files were selected One Curve (OC), ProTaper Gold (PTG), and Wave One Gold (WOG). Each type was divided into three subgroups (n = 6 for each file type) (1) untreated instruments; (2) files immersed in 0.002% PAA; and (3) files immersed in 0.35% PAA. The performance of each file type was tested in a simulated canal. Blasticidin S mw The number of cycles to fracture (NCF) was determined to assess cyclic fatigue resistance of the files. Independent sample t-test was applied to compare each treated file within a subgroup with its respective control group, and one-way ANOVA was used for comparison among the main groups. All types of tested files revealed a significant decline in the cyclic fatigue resistance after exposure to 0.002% PAA except the PTG (P=0.209). After exposure of the files to a higher concentration (0.35% PAA), a dramatic reduction was demonstrated by all the groups. Before and after exposure of the files to PAA, PTG displayed the highest cyclic fatigue resistance, followed by the WOG, while the OC showed the lowest resistance. Exposure of heated-treated NiTi files to PAA in a relatively high or low concentration adversely affects the cyclic fatigue resistance. The PTG files demonstrated the best performance among the tested types and can be disinfected with 0.002% PAA for clinical purpose.Enamel demineralization and white-spot lesions (WSLs) around the orthodontic brackets are common clinical complications after orthodontic fixed appliance therapy. WSLs form mainly due to plaque deposition around the brackets during the orthodontic treatment period. This study was designed to compare and evaluate the efficacy of two different remineralization agents on WSLs, which are "Clinpro 5000 and Colgate Sensitive Pro-Relief". 27 caries-free human premolar teeth were collected after extraction for orthodontic purposes. The crowns were set in acrylic resin, and the entire surfaces were coated with nail varnish apart from an area of 4 × 4 mm on the buccal surface. The surface microhardness (SMH) was measured using the Vickers microhardness testing machine at baseline, after demineralization, and after treatment. Then, the different SMH values were statistically analyzed using mixed-effects linear regression. All samples were immersed in demineralizing solution for ten days to create WSLs, and then the teetolgate Sensitive Pro-Relief" have significantly improved enamel's SMH.
Paeoniflorin has been reported to exert antitumor effects on human cancers. However, the role of paeoniflorin in gastric cancer and the underlying molecular mechanism are unelucidated. Therefore, we determined whether paeoniflorin could exhibit anticancer activity in gastric cancer cells.

MTT was used to measure the viability of cells after paeoniflorin treatment. FACS was performed to examine cell apoptosis. Wound healing and transwell invasion assays were conducted to examine cell migratory and invasive activities. Western blotting was used to explore the mechanism by which paeoniflorin exerted tumor suppressive effects.

We found that paeoniflorin suppressed cell growth, enhanced apoptosis, and reduced cell invasion. Notably, we showed that paeoniflorin inhibited the expression of TAZ in gastric cancer cells. The overexpression of TAZ abrogated the antitumor activity of paeoniflorin in gastric cancer cells. In contrast, the downregulation of TAZ promoted the tumor suppressive effects of paeoniflorin treatment.

Hence, targeting TAZ with paeoniflorin could be a novel approach for the treatment of human gastric cancer.

Previous studies have found that treatment with uncarboxylated osteocalcin can protect mice from high-fat diet-induced nonalcoholic fatty liver disease (NAFLD). However, the potential mechanisms remain unclear. Although the G protein-coupled receptor family C group 6 subtype A (GPRC6A) is the putative receptor of osteocalcin, there is no direct evidence showing that GPRC6A mediates the effects of uncarboxylated osteocalcin in alleviating NAFLD in mice. We aimed to figure out this using liver-specific GPRC6A knockout (GPRC6ALKO) mice. Consistent with previous studies, uncarboxylated osteocalcin significantly protected high-fat diet-fed wild-type mice from obesity and NAFLD, while it did not protect high-fat diet-fed GPRC6ALKO mice from NAFLD. Differential mRNA expression of lipogenesis and lipolysis between GPRC6ALKO mice and control mice revealed that GPRC6A mediated the effects of osteocalcin in alleviating NAFLD through inhibiting lipid synthesis and promoting lipolysis.
