Lamina-associated polypeptide 1 (LAP1) is a nuclear envelope (NE) protein whose function remains poorly characterized. In a recent LAP1 protein interactome study, a putative regulatory role in the DNA damage response (DDR) has emerged and telomeric repeat-binding factor 2 (TRF2), a protein intimately associated with this signaling pathway, was among the list of LAP1 interactors. To gain insights into LAP1's physiological properties, the interaction with TRF2 in human cells exposed to DNA-damaging agents was investigated. The direct LAP1TRF2 binding was validated in vitro by blot overlay and in vivo by co-immunoprecipitation after hydrogen peroxide and bleomycin treatments. The regulation of this protein interaction by LAP1 phosphorylation was demonstrated by co-immunoprecipitation and mass spectrometry following okadaic acid exposure. The involvement of LAP1 and TRF2 in the DDR was confirmed by their increased nuclear protein levels after bleomycin treatment, evaluated by immunoblotting, as well as by their co-localization with DDR factors at the NE and within the nucleoplasm, assessed by immunocytochemistry. Effectively, we showed that the LAP1TRF2 complex is established during a cellular response against DNA damage. This work proposes a novel functional role for LAP1 in the DDR, revealing a potential biological mechanism that may be disrupted in LAP1-associated pathologies.In recent years more and more attention has been given to autogenous shrinkage due to the increasing use of high-performance concrete, which always contains supplementary materials. With the addition of supplementary materials-e.g., fly ash and blast furnace slag-internal relative humidity, chemical shrinkage and mechanical properties of cement paste will be affected. These properties significantly influence the autogenous shrinkage of cement paste. In this study, three supplementary materials-i.e., silica fume, fly ash and blast furnace slag-are investigated. Measurements of final setting time, internal relative humidity, chemical shrinkage, compressive strength and autogenous deformation of the cement pastes with and without supplementary materials are presented. Two water-binder ratios, 0.3 and 0.4, are considered. The effects of different supplementary materials on autogenous shrinkage of cement paste are discussed.Poultry meat is commonly marketed at refrigerated temperatures (2-5 °C). The major concern for retailers and consumers is the quality and safety of refrigerated poultry meat. During the chilling period, poultry meat undergoes too many undesirable changes due to microbial growth that leads to spoilage and economic loss. Therefore, this study was conducted to assess the effects of olive leaf extracts (OLE) used at three concentrations (0.25, 0.5, and 1%) on the sensory attributes, as well as the chemical and microbiological quality of raw poultry meat stored at 4 ± 1 °C for 15 days. The results revealed that the OLE addition reduced microbial growth successfully, and maintained the chemical quality and sensory attributes of poultry meat. Moreover, OLE extended the shelf-life of the poultry meat that held under proper refrigeration conditions up to 15 days compared to the control group, that was completely spoiled by the sixth day of storage. This study concludes that OLE could be used both as a natural antioxidant and an antimicrobial preservative for chilled poultry meat held at refrigerated temperature.Apoptosis, programmed cell death type I, is a critical part of neurodegeneration in cerebral ischemia, Parkinson's, and Alzheimer's disease. Ferritinophagy, a ferritin degradation pathway, depends on a selective autophagic cargo receptor (NCOA4). By screening various types of natural compounds, formosanin C (FC) was identified as a novel ferroptosis inducer, characterized by attenuations of FC-induced viability inhibition and lipid ROS formation in the presence of ferroptosis inhibitor. FC also induced autophagic flux, evidenced by preventing autophagic marker LC3-II degradation and increasing yellow LC3 puncta in tandem fluorescent-tagged LC3 (mRFP-GFP) reporter plasmid (ptfLC3) transfected cells when combined with autophagic flux inhibitor. It is noteworthy that FC-induced ferroptosis and autophagic flux were stronger in HepG2 cells expressing higher NCOA4 and lower ferritin heavy chain 1 (FTH1) levels, agreeing with the results of gene expression analysis using CTRP and PRISM, indicating that FTH1 expression level exhibited a significant negative correlation with the sensitivity of the cells to a ferroptosis inducer. Confocal and electron microscopy confirmed the pronounced involvement of ferritinophagy in FC-induced ferroptosis in the cells with elevated NCOA4. Since ferroptosis is a non-apoptotic form of cell death, our data suggest FC has chemotherapeutic potential against apoptosis-resistant HCC with a higher NCOA4 expression via ferritinophagy.Sensor networks in real-world environments, such as smart cities or ambient intelligent platforms, provide applications with large and heterogeneous sets of data streams. Outliers-observations that do not conform to an expected behavior-has then turned into a crucial task to establish and maintain secure and reliable databases in this kind of platforms. However, the procedures to obtain accurate models for erratic observations have to operate with low complexity in terms of storage and computational time, in order to attend the limited processing and storage capabilities of the sensor nodes in these environments. In this work, we analyze three binary classifiers based on three statistical prediction models-ARIMA (Auto-Regressive Integrated Moving Average), GAM (Generalized Additive Model), and LOESS (LOcal RegrESSion)-for outlier detection with low memory consumption and computational time rates. As a result, we provide (1) the best classifier and settings to detect outliers, based on the ARIMA model, and (2) two real-world classified datasets as ground truths for future research.
