Type 2 diabetes mellitus (T2D) is a common chronic disease, which lacks a specific diagnostic method. A substantial body of literature has demonstrated that the molecular constituents of extracellular vesicles (EVs) are promising as a novel biomarker for clinical diagnosis, especially exosomal miRNAs in biological fluids.

To find a diagnostic biomarker for T2D, we isolated exosomes from plasma and then quantitative PCR (qPCR) was used to detect the miRNA expression in plasma and exosomes from control subjects and T2D subjects. The ROC (receiver operating characteristic) curve and AUC (area under curve) were used to evaluate the diagnostic value of exosomal miRNAs.

We found the exosomal levels of miR-23a and miR-192 were both significantly higher in T2D subjects. The AUC of miR-23a and miR-192 were 0.828 and 0.717, respectively. Further bioinformatics analysis was performed to speculate possible mechanisms.

In conclusion, exosomal miR-23a and miR-192 have good potential as diagnostic biomarkers for type 2 diabetes.

The current study aims to investigate the expression and diagnostic value of ADAM12 in patients with cervical cancer before general anesthesia.

Seventy-eight cases of cervical cancer patients were included in the present study. RT-PCR and western blot were used to detect the expression of ADAM12 in cervical cancer tissues and adjacent tissues. Meanwhile, the expression of secretory ADAM12 in serum of cervical cancer patients and healthy people was detected by ELISA. The relationship between ADAM12 expression and prognosis of cervical cancer patients was analyzed. ROC analysis was carried out to explore the diagnostic value of ADAM12.

Our data showed that the expression of ADAM12 mRNA and protein in cervical cancer tissues was significantly up-regulated compared with the adjacent tissues. ELISA assay showed that the content of ADAM12 in serum of cervical cancer patients was significantly higher than that of healthy people. Furthermore, ADAM12 expression was closely related to tumor invasion, TNM stage, lymph node metastasis and tumor differentiation. Kaplan-Meier survival analysis showed that the overall survival rate of patients with high ADAM12 was significantly lower than that of patients with low ADAM12 expression. The AUC of ADAM12, CEA, CA125, and SCC for cervical cancer was 0.893, 0.510, 0.769 and 0.550, respectively, while the highest value of AUC was 0.946 by the combination of the four indexes.

To investigate the correlations of serum homocysteine (Hcy), α2-Heremans-Schmid glycoprotein (AHSG), and C-reactive protein (CRP) with insulin resistance (IR), 25-hydroxyvitamin D (25-OH-VD), and blood lipids in patients with gestational diabetes mellitus (GDM) by detecting their levels.

A total of 72 GDM patients (GDM group) and 72 healthy pregnant women (control group) delivered in our hospital from February 2017 to January 2019 were randomly selected. The basic data, somatological parameters [height, weight, body mass index (BMI), waist circumference, hip circumference, waist-to-hip ratio (WHR), blood pressure, and body fat content], and biochemical indexes (glucose metabolism indexes, lipid metabolism indexes, Hcy, AHSG, CRP, and 25-OH-VD) were compared between the two groups. Additionally, Pearson's correlation analysis was employed to analyze the correlations among indicators.

In comparison with the control group, the GDM group had a higher average rate of family history of DM (p < 0.05), largeted with HOMA-IR, TC, and AHSG (r = 0.485, 0.331, 0.226, p < 0.05), negatively associated with 25-OH-VD (r = -0.443, p < 0.05), and had no correlation to TG and Hcy (r = 0.019, 0.058, p > 0.05). AHSG displayed positive correlations with HOMA-IR, TC, TG, and CRP (r = 0.647, 0.321, 0.314, 0.226, p < 0.05) and no association with Hcy and 25-OH-VD (r = 0.058, -0.034, p > 0.05).
