Single case studies continue to play an important role in neuropsychological research. However, the range of statistical tools specifically designed for single cases is still limited. The current gold standard is the Crawford's t-test, but it is crucial to note that this is limited to simple designs and it is not possible to make inferences relevant to support for the null hypothesis with it. The Bayesian Multilevel Single Case models (BMSC) provide a novel tool that grants the flexibility of linear mixed model designs. BMSC is also able to support both null and alternative hypotheses in complex experimental designs using the Bayesian framework. We compared the BMSC and Crawford's t-test in a simulation study involving a case of no-dissociation and a case of simple dissociation between a single case patient and a series of control groups of different sizes (N = 5, 15, or 30). We then showed how BMSC is useful in complex designs by means of an example using real data. The BMSC proved to be more reliable than the Crawford's test, in terms of first-type errors and more precise estimating the parameters. Notably, the BMSC model provides a comprehensive vision of the whole experimental design, interpolating a single model. It follows the recent trend which involves a shift in attention from p-values to other inferential indices and estimates.MYC-rearranged large B-cell lymphoma with BCL2 and/or BCL6 rearrangement, double-hit (DH) or triple-hit (TH) lymphoma, is associated with poor survival after standard treatment. To investigate the clinical impact of single-hit (SH) MYC rearrangement, we analyzed 241 cases of diffuse large B-cell lymphoma (DLBCL) for MYC, BCL2, and BCL6 rearrangement by fluorescence in situ hybridization. Fifty-five of 241 (22.8%) cases showed MYC rearrangements. Twenty-three cases were diagnosed as DLBCL; 18 as high-grade B-cell lymphoma (HGBCL)-DH; 3 as HGBCL-TH; and 11 as HGBCL, not otherwise specified. Both DH and TH lymphomas showed high-grade morphology (P = 0.002), higher stage (P = 0.022), and more frequent germinal center B-cell-like phenotype (P = 0.008). SH lymphomas displayed high-grade morphology (P = 0.002) but were not different from MYC-negative lymphomas in cell of origin, clinical stage, international prognostic index (IPI), or extranodal involvement. Patients with DH/TH lymphomas had worse overall survival (OS) (P = 0.016) and progression-free survival (PFS) (P less then 0.001), while OS and PFS of SH lymphomas were not different from those of MYC-negative lymphomas. There was no survival difference between cases of BCL2 and BCL6 rearrangements. Poorer prognostic factors included higher ECOG class, higher IPI, and DH or TH translocation for OS, and higher IPI and DH or TH translocation for PFS. Higher IPI was an independent prognostic factor for OS and PFS. In conclusion, large B-cell lymphomas with single MYC rearrangement showed high-grade morphology but were otherwise not different from MYC-negative lymphomas.In this work the effect of (-)-epicatechin on the development of amebic liver abscess in hamsters was evaluated. (-)-epicatechin is a flavonoid present in plants that possesses various biological properties, including its activity against some protozoal parasites; however its antiamebic activity in a living model had not been evaluated. Syrian golden hamsters were intrahepatically inoculated with 1x106E. histolytica trophozoites, three days after inoculation they received nine intraperitoneal doses of (-)-epicatechin (10 mg/100 g) every 48 h. Animals without treatments and treated with metronidazole were included as controls. Macroscopic characteristics of the hepatic abscess, histopathological analysis of the tissue and the levels of inflammatory cytokines were determined. Doramapimod (-)-epicatechin produced a decrease in liver abscess progression being observed only 9.49% of damage compared to 84% shown by untreated animals. During treatment with (-)-epicatechin hepatic tissue showed signs of liver repair and absence of amoebae. Additionally, (-)-epicatechin produced a modulating effect on inflammatory cytokines TNF-α, IL-1β and IL-10. All these events observed in animals treated with (-)-epicatechin could contribute to the elimination of trophozoites and liver healing.One major challenge in animal research on spatial learning and memory pertains to designing methods to dissociate spatial strategies (allocentric vs. egocentric). This is crucial for understanding the underlying cognitive processes and neural circuits that are recruited in navigational tasks. Taking the egocentric reference frames as a starting point, this review argues that in many extensively used spatial paradigms, multiple spatial reference frames are often available to the animals but remain unaccounted for. We discuss the implications this has for the inferences that can be made and propose a decision-algorithm to construct spatial learning paradigms that can reduce the influence of these confounding variables. Furthermore, with these considerations in mind, we review the role of the hippocampus in egocentric navigation forms, i.e. in response learning, egocentric sequential learning and path integration. This choice is based on the controversy surrounding the role of hippocampus in these spatial paradigms. We discuss the possible methodological confounders that may explain the inconclusive results.The COVID-19 pandemic has resulted in societal-level changes to sleep and other behavioral patterns. Objective data would allow for a greater understanding of sleep-related changes at the population level. About 163,524 active Fitbit users from 6 major US cities contributed data, representing areas particularly hard-hit by the pandemic (Chicago, Houston, Los Angeles, New York, San Francisco, and Miami). Sleep variables extracted include nightly and weekly mean sleep duration and bedtime, and variability (standard deviation) of sleep duration and bedtime. Deviation from similar timeframes in 2018 and 2019 were examined, as were changes in these sleep metrics during the pandemic, relationships to changes in resting heart rate, and changes during re-opening in May and June. Overall, compared to 2019, mean sleep duration in 2020 was higher among nearly all groups, mean sleep phase shifted later for nearly all groups, and mean sleep duration and bedtime variability decreased for nearly all groups (owing to decreased weekday-weekend differences).
