Once cancers occupy bone, the prognosis is poor as disease recurrence and visceral spread is imminent. Understanding the pathways and cellular interactions, which regulate tumour cell seeding, dormancy and growth in bone, is pertinent to improving outcomes for patients with advanced cancers. Advances in imaging techniques have facilitated the development of the concept that the behavior of bone marrow resident cells dictates the fate of tumour cells upon arrival in bone. This review summarises recent findings achieved through intravital imaging. It highlights the importance of developing both longitudinal static and acute dynamic data to develop our understanding of tumour cell engraftment, dormancy, activation and the subsequent establishment of metastases. We also describe how imaging techniques have developed our knowledge of the elements that make up the complex bone microenvironment which tumour cells interact with to survive and grow. We also discuss how through combining these imaging insights with single cell RNA sequencing data, we are entering a new era of research which has the power to define the cell-cell interactions which control tumour cell growth in bone.
Fetal growth patterns and birth weight (BW) have been associated with bone mineral density (BMD) and content (BMC) throughout infancy and childhood up to early adulthood. We hypothesized that in small for gestational age (SGA) children, compensatory infant catch-up growth to normal height centiles counteracts the adverse consequences of low BW on bone accrual.

To evaluate BMD and BMC of SGA children born at term who experienced a normal catch-up growth as compared to children born appropriate for gestational age (AGA).

We recruited 53 SGA (26 females) and 60 AGA children (27 females), aged 6 to 18years, matched for sex and body mass index (BMI). Fat mass (FM); Free fat mass (FFM); Lumbar spine and Total body less head (TBLH) BMD; BMC and BMD standard deviation scores corrected for body size (BMAD, BMAD z-score and TBLH BMD/Height) and TBLH BMC for FFM (TBLHBMC/FFM) were derived from Dual Energy X-ray absorptiometry (DXA) scans.

SGA and AGA children did not differ in any auxological, body composition and bone parameters appropriately adjusted for height and FFM. BMI, FM and, remarkably, FFM were significantly correlated with bone mass parameters in both groups while no correlation was found between FM and FFM with the BW SDS in the univariate analysis.

Our preliminary data demonstrate that SGA children born at term who recover from their growth deficiency through catch-up growth achieve bone mass and body composition not different from children born AGA.

A novel bacterial consortium CTB-4 which stands for the molecularly identified species Burkholderia sp., Xanthomonas sp., Shewanella sp., and Pseudomonas mosselii was successfully developed. The CTB-4 consortium showed high efficiency in the removal of naphthalene and phenol. It also revealed reduction in lignin, hemicellulose, and cellulose by 19.4, 52.5, and 76.8%, respectively. The main metabolites after the CRO degradation were acetic acid, succinate, pyruvate, and acetaldehyde. Pretreatment of treated BSD mixed with CRO enhanced the total methane yield (162 L/kg VS) by 82.7% and biomass reduction by 54.7% compared to the untreated substrate. CRO showed a toxicity decrease of >90%, suggesting the efficiency of constructed bacterial consortia in bioremediation and biofuel production. The aromatic nature of lignin makes it a good source for the production of numerous platform chemicals. The valorization of lignin into valuable compounds depends upon the type of bonds and functionality present in lignin. PR-171 mw Here, we have studied the depolymerization of rice straw alkali lignin in N2 and O2 with acidic (ZSM-5), basic (MgO) catalyst and with their mixtures (11, 37 and 73). The effect of hydrogen peroxide on lignin depolymerization was also examined. Maximum yield of bio-oil (50 wt%) was obtained with pure ZSM-5 and 1 ml hydrogen peroxide in nitrogen atmosphere, while maximum conversion (60%) was observed in oxygen environment during the non-catalytic depolymerization of lignin. Bio-oil characterization through GC-MS showed maximum selectivity towards 2-methoxy-4-vinylphenol with 38.5 area% in the bio-oil of ZSM-5-N2. The bio-oils have also been characterized using 1H NMR, FT-IR and GC-MS.A novel biorefinery platform integrating thermal pretreatment and solid-liquid separation unit is here proposed to fully exploit food waste (FW) potential for production of valuable chemicals and energy through semi-continuous anaerobic bioconversion. The liquid fraction deriving from raw or pretreated FW, was fermented into volatile fatty acids (VFAs, from acetic to caproic acid) while the residual fraction was converted into biomethane. Thermal pretreatment effectively extracted a portion of the macromolecular organics, especially starch, to the liquid phase, promoting acidogenic fermentation and chain elongation pathways (0.43 gVFA g-1VSfed and 0.58 gVFA g-1VSfed with raw and pretreated extract, respectively). In parallel, anaerobic digestion of solid residue in 10 L reactors showed process stability and higher conversion rate for the pretreated residue (0.31 against 0.26 Nm3CH4 kg-1VSfed). The mass-transfer balance coupled with the economic assessment, calculated in terms of direct gross added value, indicated promising revenues by integrating the thermal upstream treatment.
