Haematological parameters were generally in the reference values and any significant changes except levels of lymphocytes and percent of lymphocytes which shown significant increase in the second collection period were found. Based on the result we can state that the experimental sample collection has no effect on blood parameters of the animals but we spectated a statistically insignificant decrease in the levels of cortisol which can suggest that the dark collection is possibly less stressful to the animals.Pyrazinamide (PZA) is a component of first-line drugs, active against latent Mycobacterium tuberculosis (MTB) isolates. The prodrug is activated into the active form, pyrazinoic acid (POA) via pncA gene-encoded pyrazinamidase (PZase). Mutations in pncA have been reported, most commonly responsible for PZA-resistance in more than 70% of the resistant cases. In our previous study, we detected many mutations in PZase among PZA-resistance MTB isolates including A46V, H71Y, and D129N. The current study was aimed to investigate the molecular mechanism of PZA-resistance behind mutants (MTs) A46V, H71Y, and D129N in comparison with the wild type (WT) through molecular dynamic (MD) simulation. MTB positive samples were subjected to PZA drug susceptibility testing (DST) against critical concentration (100ug/ml). The resistant samples were subjected to pncA sequencing. Thirty-six various mutations have been observed in the coding region of pncA of PZA-resistant isolates (GenBank accession No. MH461111) including A46V, H71Y, and D129N. The post-simulation analysis revealed a significant variation in MTs structural dynamics as compared to the WT. Root means square deviations (RMSD) and Root means square fluctuation (RMSF) has been found in variation between WT and MTs. Folding effect and pocket volume were altered in MTs when compared with WT. Geometric matching supports the effect of mutation A46V, H71Y, and D129N on PZase structure that may have an insight effect on PZase dynamics, making them vulnerable to convert pro-PZA into active form, POA. In conclusion, the current analyses will provide useful information behind PZA-resistance for better management of drug-resistant TB.

Compared with the model group, the serum TG and the ratio of liver to body weight of the model mice that suffered from acute alcoholic hepatic injury could be significantly decreased in the group that practiced aerobic exercise, the group administered with SF, and the group treated with the approach of SF integrated with aerobic exercise (

Activation of microglia often aggravates the pathological process of various neurological diseases. Natural chemicals have unique advantages in inhibiting microglia-mediated neuroinflammation and improving neuronal function. Here, we examined the effects of asperosaponin VI (ASA VI) on LPS-activated primary microglia. Microglia were isolated from mice and pretreated with different doses of ASA VI, following lipopolysaccharide (LPS) administration. Activation and inflammatory response of microglia cells were evaluated by real-time fluorescence quantitative polymerase chain reaction (q-PCR), immunohistochemistry and enzyme-linked immunosorbent assay (ELISA). Signaling pathways were detected by western blotting. We found that the ASA VI inhibited the morphological expansion of microglia cells, decreased the expression and release of proinflammatory cytokines, and promoted the expression of antiinflammatory cytokines in a dose-dependent manner. ASA VI also activated PPAR-γ signaling pathway in LPS-treated microglia. The anti-inflammatory effects of ASA VI in microglia were blocked by treating PPAR-γ antagonist (GW9662). These results showed that ASA VI promote the transition of microglia cells from proinflammatory to anti-inflammatory by regulating PPAR-γ pathway.Salt stress is known to be momentous abiotic stress which treats agricultural lands and crop production throughout the world and effects the system of food security. The current study aims to investigate the effect of foliar application of 10 mg/l of zinc oxide (ZnO) as a bulk or as a green synthesized nanoparticle (ZnO-NPs) which were hexagonal and spherical in shape and at size 16-35 nm to alleviate the impact of salinity concentrations (0, 10, 25, 50, 75 and 100% SW) on Okra (Abelmoschus esculentus L. Moench cv. Hasawi) species. The results demonstrated a gradual decrease in the photosynthetic pigments (i.e., chlorophyll a and b with total chlorophylls and carotenoids) with the growth of salinity conc. However, the sea water levels between 0 and 75% will led to increase in proline, total soluble sugar and activity of the antioxidant enzymes i.e., superoxide dismutase (SOD) and catalase (CAT) and then decrease at 100% SW. The addition of bulk ZnO or ZnO-NPs enhances the contents of the photosynthetic pigments, activity of both SOD and CAT and then lowers the accumulation of proline and total soluble sugar when equated with controls. Plants treated with ZnO-NPs showed the greatest results when compared with other treatments. The results of current study showed ZnO-NPs as an appropriate eco-friendly and low-cost application for plant growth under salinity which has an ability to moderate the salt stress effect of plants.
