Alpha diversity between the sample groups was not significantly different (P>0.05) regardless of different STEC markers. Non-metric multidimensional scaling (nMDS) of slaughter samples showed that the bacterial composition in faecal and hide samples shared the least similarity with the communities in carcase and environmental samples. Surface samples from slaughter (carcase and environmental) and boning (carcase, beef trim and environmental) all appeared randomly plotted on nMDS. This indicated that the STEC presence also did not have a significant effect (P>0.05) on beta diversity. While presence of STEC appeared to correlate with changes in diversity of faecal and hide bacterial communities in previous studies, it did not appear to have the same effect on other samples throughout processing.Background Poor sleep may increase the likelihood of fatigue, and both are common in later life. However, prior studies of the sleep-fatigue relationship used subjective measures or were conducted in clinical populations, thus the nature of this association in healthier community-dwelling older adults remains unclear. We studied the association of actigraphic sleep parameters with perceived fatigability-fatigue in response to a standardized task-and with conventional fatigue symptoms of low energy or tiredness. Methods We studied 382 cognitively normal participants in the Baltimore Longitudinal Study of Aging (aged 73.1±10.3 years, 53.1% women) who completed 6.7±0.9 days of wrist actigraphy and a perceived fatigability assessment, including rating of perceived exertion (RPE) after a 5-minute treadmill walk or the Pittsburgh Fatigability Scale (PFS). Participants also reported non-standardized symptoms of fatigue. Results After adjustment for age, sex, race, height, weight, comorbidity index, and depressive symptoms, shorter total sleep time (TST; less then 6.3 hours versus intermediate TST ≥6.3 to 7.2 hours) was associated with high RPE fatigability (OR=2.56, 95% CI=1.29, 5.06, p=0.007), high PFS physical (OR=1.88 , 95% CI=1.04, 3.38, p=0.035) and high mental fatigability (OR=2.15, 95% CI=1.02, 4.50, p=0.044), whereas longer TST was also associated with high mental fatigability (OR=2.19, 95% CI=1.02, 4.71, p=0.043). Additionally, longer wake bout length was associated with high RPE fatigability (OR=1.53, 95% CI=1.14, 2.07, p=0.005), and greater wake after sleep onset was associated with high mental fatigability (OR=1.14, 95% CI=1.01, 1.28, p=0.036). Conclusion Among well-functioning older adults, abnormal sleep duration and sleep fragmentation are associated with greater perceived fatigability.Background Advanced liver disease due to HCV is a leading cause of HIV-related morbidity and mortality. There remains a need to develop noninvasive predictors of clinical outcomes in persons living with HIV/HCV co-infection. Methods We conducted a nested case-control study in 126 patients with HIV/HCV and utilized multiple quantitative metabolomic assays to identify a prognostic profile that predicts ESLD events including ascites, hepatic encephalopathy, hepatocellular carcinoma, esophageal variceal bleed, and spontaneous bacterial peritonitis. Each analyte class was included in predictive modeling and Area Under the Receiver Operator Characteristic Curves (AUC) and accuracy were determined. Results The baseline model including demographic and clinical data had an AUC of 0.79. Three models, baseline + amino acids, lipid metabolites, or all combined metabolites, had very good accuracy (AUC 0.84-0.89) in differentiating patients who are at risk of developing an ESLD complication up to 2 years in advance. The all combined metabolites model had a sensitivity of 0.70, specificity of 0.85, positive likelihood ratio of 4.78 and negative likelihood ratio of 0.35. Conclusions We report that quantification of a novel set of metabolites may allow earlier identification of patients with HIV/HCV who have the greatest risk of developing ESLD clinical events.Aims To investigate wheather incident Atrial fibrillation or flutter (AF) diagnosis increases the risk of psychiatric outcomes compared to the general population. Methods and results First-time AF patients and population controls naïve to psychiatric disease or filled presciptions for psychotropic drugs were identified in Danish nationwide registries during 2005-2014. AF patients were matched 12 with exposure density matching. Patients and controls were compared for 1-year cumulative incidences of depression, anxiety and stress disorders, and for filled drug prescriptions for antidepressant, anxiolytic, selected antipsychotics and hypnotics. Lastly, we examined 1-year cumulative incidences of a composite endpoint of the above mentioned diagnoses or drug redemptions. We included 146,377 AF patients and 292,754 matched controls, 55% men and median age 74 (25%-75% 65-82) years. AF patients had significantly higher cumulative incidences of composite endpoints. Furthermore, filled prescriptions for anxiolytics and hypnotics were significantly higher for AF patients compared to healthy population controls. The cumulative incidence of the composite endpoint was significantly higher in AF patients relative to controls 11.1% vs. 8.3%. Acetalax clinical trial For the composite endpoint, a significantly higher risk was apparent both in unadjusted (HR 2.76, 95% CI 2.67-2.85) and adjusted (HR 2.51, 95% CI 2.43-2.60) models for AF patients versus controls in the first three months after study inclusion. Conclusion First-time AF patients were significantly more likely to have psychiatric outpatient or hospital contacts and to fill presciptions for psychotropic drugs compared to healthy population controls. The risk was significantly elevated only during the first three months after AF diagnosis.Maternal nutrition can affect the susceptibility of the offspring to metabolic disease later in life, suggesting that this period is a window of opportunity for intervention to reduce the risk of metabolic disease. Resveratrol, a natural polyphenol, has a wide range of beneficial properties including anti-obesogenic, anti-atherosclerotic, and anti-diabetic effects. We previously reported that maternal resveratrol intake during pregnancy and lactation has early metabolic effects in the offspring with these effects at weaning depending on the type of diet ingested by the mother and the offspring's sex. Here we analyzed whether these metabolic changes are maintained in the adult offspring and if they remain sex and maternal diet dependent. Wistar rats received a low-fat (LFD; 10.2% Kcal from fat) or high fat (HFD; 61.6% Kcal from fat) diet during pregnancy and lactation. Half of each group received resveratrol in their drinking water (50 mg/L). Offspring were weaned onto standard chow on postnatal day 21. Maternal resveratrol reduced serum cholesterol levels in all adult offspring from HFD mothers and increased it in adult female offspring from LFD mothers.
