The research results are expected to provide a basis for the application of CSOV beams in beam reshaping, optical trapping, and rotating microparticle manipulation, particularly in separating cells.Airway smooth muscle hyperresponsiveness associated with chronic airway inflammation leads to the typical symptoms of asthma including bronchoconstriction and wheezing. Asthma severity is associated with airway inflammation; therefore reducing airway inflammation is an important therapeutic target. Gelsolin is an actin capping and severing protein that has been reported to be involved in modulation of the inflammatory response. Using mice genetically lacking gelsolin, we evaluated the role of gelsolin in the establishment of house dust mite (HDM) antigen-induced allergic lung inflammation. The genetic absence of gelsolin was found to be protective against HDM sensitization, resulting in reduced lung inflammation, inflammatory cytokines and Muc5AC protein in bronchoalveolar lavage (BAL) fluid. The number of eosinophils, lymphocytes and interstitial macrophages in the BAL were increased after HDM sensitization in wild type mice, but were attenuated in gelsolin null mice. The observed attenuation of inflammation may be partly due to delayed migration of immune cells, because the reduced eosinophils in the BALs from gelsolin null mice compared to controls occurred despite similar amounts of the chemoattractant eotaxin. Splenic T cells demonstrated similar proliferation rates, but ex vivo alveolar macrophage migration was delayed in gelsolin null mice. In vivo, the reduced lung inflammation after HDM sensitization in gelsolin null mice was associated with significantly diminished airway resistance to inhaled methacholine compared with HDM-treated wild type mice. Our results suggest that modulation of gelsolin expression or function in selective inflammatory cell types that modulate allergic lung inflammation could be a therapeutic approach for asthma.
In this study, we aimed to investigate the effect of DNA methyltransferase 1 gene (DNMT1) expression on leukemia cell proliferation.

Following stimulation with interferon-α (IFN-α) or methylation inhibitor for three days, we evaluated changes in the DNMT1 expression levels, cell proliferation activity, and Bcl-2-Associated X Protein (BAX) expression levels in the chronic myelogenous leukemia cell line K562 and the acute monocytic leukemia cell line THP-1.

DNMT1 expression levels and cell proliferation activity decreased in K562 and THP-1 cells, whereas BAX expression levels increased.

These results suggest that the enzymatic activity of DNMT1 promotes the proliferation of tumor cells and that tumor cell proliferation can be suppressed by inhibiting DNMT1 enzymatic activity. Furthermore, because DNA methylation is associated with apoptosis, a process critical to cell growth and injury in leukemia, assessing DNMT1expression levels might help in treatment decisions for leukemia patients.

Treatment for chronic immune thrombocytopenia (cITP) in children is largely limited to immunosuppressive agents. Thrombopoietin receptor agonists (TRAs) have been used to treat cITP in adults for over a decade. The objective of this integrated analysis was to examine the safety and efficacy of the TRA romiplostim in children with ITP.

We examined efficacy and safety in children with ITP across five romiplostim trials final data from two double-blind placebo-controlled trials and two open-label extensions, and interim data from an ongoing single-arm trial.

Patients (n=24 initially placebo; n=262 initially romiplostim) had a median age of 10.0years (Q1 6.0, Q3 13.0), ITP duration of 1.9years (Q1 1.0, Q3 4.0), and baseline platelet count of 14.3×10
/L (Q1 7.5, Q3 23.0). Among 282 patients receiving romiplostim, median treatment duration was 65weeks (range 8-471weeks) and median weekly dose was 6.6μg/kg (range 0.1-9.7μg/kg). Overall, 89% of romiplostim-treated patients had platelet responses. Nineteen patients (7%) maintained treatment-free responses for ≥6months while withholding all ITP therapy.
