METHODS Thoracic aortas were isolated from Wistar rats, and were suspended in tissue organ chambers for vascular tension measurement. The effect of cumulative osthole (10-?, 10-?, 10-?, 10-?, and 10-? mol/L) on endothelium-intact and endothelium-denuded thoracic aortic rings pre-contracted with phenylephrine (PE, 10-? mol/L) or KCl (6 × 10-? mol/L) was recorded. The effects of different osthole concentrations on endothelium-intact aortic rings, which were pre-inhibited with the non-selective nitric oxide synthase inhibitor L-Arg(NO2)-OMe·HCl (3 × 10-4 mol/L), endothelium-derived nitric oxide synthase inhibitor Nω-nitro-L-arginine (3 × 10-4 mol/L), guanylate cyclase inhibitor 1H-[1,2,4] oxadiazolo [4,3-α] quinoxaline-1-one (10-5 mol/L), cyclooxygenase inhibitor indometacin (10-5 mol/L), and the Ca2+ and control (4.21% ± 1.33%) groups. However, after cumulative CaCl2 (0.5, 1, 1.5, 2, 2.5, and 3 mmol/L), the Ca2+-induced contraction was significantly inhibited in the osthole and phentolamine groups compared with controls (P less then 0.05). After cumulative CaCl2 was added to Ca2+-free KHS (high K+ concentration), the contraction rate was significantly higher than both of the control and the osthole groups (P less then 0.05). The contraction rate in the osthole group was higher than the verapamil group (P less then 0.05). CONCLUSION Osthole has a vasorelaxant effect on isolated rat thoracic aortic rings, via inhibition of both receptor-operated and voltage-dependent Ca2+ channels in arterial smooth muscle, leading to decreased Ca2+ influx, and via inhibition of nitric oxide release on arterial endothelial cells.OBJECTIVE To evaluate biological potential of methanol and n-hexane extracts of aerial parts in seven species of family Poaceae. METHODS Qualitative phytochemical tests were done by using standard protocols. In vitro antioxidant activity was performed via different assays and antimicrobial potential was observed via disc diffusion method. Cytotoxic activity was carried out using brine shrimps' assay. RESULTS Phytochemical studies revealed the presence of alkaloids, flavonoids, glycosides, phenols, steroids, saponins, tannins, anthocyanins and coumarins in most of the plant extracts. Maximum antioxidant and antimicrobial potential were observed in Cymbopogon citratus methanol extract and Cymbopogon citratus n-hexane extract along with significant number of total flavonoids and phenols contents. However, Polypogon monspeliensis methanol extract and Polypogon monspeliensis n-hexane extract showed minimum antioxidant as well as antimicrobial potential. Moreover, methanol extracts showed a cytotoxic effect with their effectiveness ranked Polypogon monspeliensis methanol extract > Cymbopogon citratus methanol extract > Phalaris minor n-hexane extract > Aristida funiculata methanol extract > Stipagrostis plumosa methanol extract > Cenchrus ciliaris methanol extract > Panicum antidotale methanol extract. Similarly, n-hexane plant extracts revealed cytotoxic activity in decreasing order Cenchrus ciliaris n-hexane extract > Stipagrostis plumosa n-hexane extract > Phalaris minor n-hexane extract > Aristida funiculata n-hexane extract > Polypogon monspeliensis n-hexane extract > Panicum antidotale n-hexane extract > Cymbopogon citratus n-hexane extract respectively. CONCLUSION Methanol extracts exhibit significant antioxidant and antimicrobial potential which can be correlated to their medicinal values. The observed brine shrimp's lethality of the plant extracts revealed the presence of potent cytotoxic components in these plants.OBJECTIVE To focus a new chemoprevention approach that uses nanotechnology to deliver luteolin to human breast cancer cells (MCF-7), and its underlying mechanism. METHODS Water-soluble copolymer-encapsulated nanoparticle-luteolin (CENL) was formulated using the hydrophobic drug luteolin. The ability to load and release he anticancer drug into/from the synthesized hyperbranched polyester was evaluated by high-performance liquid chromatography. The successful synthesis of CENL was supported by analytical techniques including fourier transform infrared spectroscopy, nuclear magnetic resonance spectroscopy, gel permeation chromatography, and dynamic light scattering. Cell viability was measured using the 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide color method. Reactive oxygen species (ROS) were measured using a dichlorofluorescein probe and intracellular calcium (Cai2+) was evaluated with a flu3-AM probe. RESULTS The results showed that the drug delivery system is stable and that the loading capacity is high. Treatment with nanoparticles containing luteolin and free luteolin resulted in cell death in breast cancer cells at high concentrations [IC50 (33 ± 4) and (48 ± 6) μM, respectively]. At high concentrations, CENL reduced cell viability and increased ROS and Cai2+ production. CONCLUSION Our results demonstrate that CENL has potential for human breast cancer treatment.OBJECTIVE To review the role of vegetables to prevent and treat hemorrhoids in Persian Medicine (PM). METHODS We search main Persian Medicine manuscripts, including the books of Liber Continens, Canon of Medicine, Great Elixir, Akbarie's Medicine, Storehouse of Medicaments and Present for the faithful. Also, it was considered by searching in reference books and published papers with the help of PubMed, Scopus, Google scholar databases. RESULTS Twelve vegetables, relating to 8 plant families, have been found in PM that their effectiveness involved in laxative, anti- inflammation, antimicrobial, analgesic and wound healing. CONCLUSION Our findings suggest that 12 Persian Medicine vegetables can be used to prevent and treat hemorrhoids.
