In the current study, an optimum solvent was selected for the extraction of fibrinolytic active components. Furthermore, a strong fibrinolytic enzyme named WPI01 was purified from Whitmania pigra Whitman through various chromatographic steps. WPI01 has a molecular mass of 27044.297 Da, and the N-terminal 8 amino acid sequence was determined as VVGGVEAR. WPI01 was stable within the pH range of 6.0-10.0 and with maximum fibrinolytic activity at 40 °C and a pH of 8.0. At 500 U/mL, WPI01 induced 50.59% blood clot reduction in vitro within 6 h, which was higher than that induced by urokinase at 1000 U/mL. In an analysis of the plasminogen activator activity, WPI01 produced obvious halos on heated and unheated fibrin plates, suggesting that WPI01 may not only act as a plasminogen activator but also degrade fibrin clots directly, and more study is needed to support this. In conclusion, WPI01 is obviously different from known fibrinolytic enzymes in terms of substrate specificity and fibrinolytic mode of action, suggesting that it is a novel fibrinolytic enzyme with potential applications in the treatment and prevention of thrombosis.Objective To investigate the association between urinary vanadium concentrations and blood pressure (BP) changes in Chinese pregnant women. Methods This longitudinal study included 716 pregnant women who visited our hospital in Wuhan, China, from 2014 to 2016. Urinary sample collection and BP measurements were performed at 9-15, 21-27, and 34-40 weeks of gestation. Outcomes were repeated BP measurements (systolic blood pressure [SBP], diastolic blood pressure [DBP], pulse pressure [PP], and mean arterial pressure [MAP]) during pregnancy. Results The geometric mean of urinary vanadium concentrations was 0.65 μg/g creatinine. After adjusting for potential confounders, each threefold increase in urinary vanadium concentration was observed to be associated with a 1.06 mmHg decrease in SBP (95 % confidence interval [CI] -1.80, -0.32), a 0.68 mmHg decrease in DBP (95 % CI -1.32, -0.04), and a 0.81 mmHg decrease in MAP (95 % CI -1.42, -0.19). Cross-sectional analysis showed a negative association between urinary vanadium concentrations and BP in the third trimester. Specifically, each threefold increase in urinary vanadium concentration in the second trimester was associated with decreases in SBP, DBP, PP, and MAP by 2.72 mmHg (95 % CI -4.02, -1.41), 1.57 mmHg (95 % CI -2.72, -0.42), 1.14 mmHg (95 % CI -2.15, -0.14), and 1.95 mmHg (95 % CI -3.06, -0.85), respectively, in the third trimester. Conclusion To the best of our knowledge, this is the first longitudinal study that suggests the negative association between vanadium exposure and BP among pregnant women in China.Dengue diseases are a group of infectious diseases that have been widespread throughout the world for several decades. In the recombinant analysis, these strains were identified as recombinants derived from Chinese counterparts for both the major (DENV-1 genotype I) and the minor (DENV-1 genotype IV) parental strains. The recombination event occurred within the prM and E genes. This corresponded with the result of multiple alignments where several amino acid residues between the recombination breakpoints in the strains were identical to those in DENV-1 genotype IV. Several of the amino acid substitutions also have changed the amino acid properties, which might affect viral infection and antigenicity. These results provide insight into the genetic variation of DENV in this endemic area, which might have been involved in the dengue outbreak and high numbers of dengue haemorrhagic fever and dengue shock syndrome cases in Thailand in 2015.Acute myocarditis (AM), a recent-onset inflammation of the heart, has heterogeneous clinical presentations, varying from minor symptoms to high-risk cardiac conditions with severe heart failure, refractory arrhythmias, and cardiogenic shock. AM is moving from being a definitive diagnosis based on histological evidence of inflammatory infiltrates on cardiac tissue to a working diagnosis supported by high sensitivity troponin increase in association with specific cardiac magnetic resonance imaging (CMRI) findings. Though experts still diverge between those advocating for histological definition versus those supporting a mainly clinical definition of myocarditis, in the real-world practice the diagnosis of AM has undoubtedly shifted from being mainly biopsy-based to solely CMRI-based in most of clinical scenarios. It is thus important to clearly define selected settings where EMB is a must, as information derived from histology is essential for an optimal management. As in other medical conditions, a risk-based , based on available evidence and clinical experience.Neural stem cells (NSCs) in the adult hippocampus generate new neurons via a process referred to as neurogenesis, supporting cognitive functions. Since altered neurogenesis has been reportedly associated with several diseases such as epilepsy, the molecular basis of NSC activity is an important focus in the study of neurogenesis. Furthermore, facilitation of neurogenesis in the injured brain would be an ideal approach to replenish lost neurons for damage recovery. However, natural neurogenesis by endogenous NSCs in the adult brain is insufficient for complete recovery after severe injury. Recent advances in understanding forced neurogenesis from brain-resident non-neuronal cells by direct reprogramming and clearing hurdles to achieve it have improved the ability to replace damaged neurons in the brain. In this review, we describe molecular mechanisms underlying natural and forced neurogenesis, and discuss future directions for treatments of diseases in the central nervous system.
