In addition, the virulence of serotype 2b was well correlated with high disease severity.Water quality monitoring programs often collect large amounts of data with limited attention given to the assessment of the dominant drivers of spatial and temporal water quality variations at the catchment scale. This study uses a multi-model approach a) to identify the influential catchment characteristics affecting spatial variability in water quality; and b) to predict spatial variability in water quality more reliably and robustly. Tropical catchments in the Great Barrier Reef (GBR) area, Australia, were used as a case study. We developed statistical models using 58 catchment characteristics to predict the spatial variability in water quality in 32 GBR catchments. see more An exhaustive search method coupled with multi-model inference approaches were used to identify important catchment characteristics and predict the spatial variation in water quality across catchments. Bootstrapping and cross-validation approaches were used to assess the uncertainty in identified important factors and robustness of multi-model structure, respectively. The results indicate that water quality variables were generally most influenced by the natural characteristics of catchments (e.g., soil type and annual rainfall), while anthropogenic characteristics (i.e., land use) also showed significant influence on dissolved nutrient species (e.g., NOX, NH4 and FRP). The multi-model structures developed in this work were able to predict average event-mean concentration well, with Nash-Sutcliffe coefficient ranging from 0.68 to 0.96. This work provides data-driven evidence for catchment managers, which can help them develop effective water quality management strategies.

We present the case of a patient diagnosed with a stage IC1 yolk sac tumour of the ovary at 15 weeks gestation, who underwent surgical staging and adjuvant chemotherapy during pregnancy. Intrauterine growth restriction was diagnosed and the patient delivered by cesarean at 36 weeks gestation for obstructed labour.

The integrity of national health care systems has also been challenged, owing to shifts in guidance and misinformation. Although initial reports suggested that pregnant people were not at increased risk of severe COVID-19, current data arising from the "third wave" paint a much more concerning picture. In addition, pregnant and lactating people were excluded from vaccine trials, which has hindered the ability of health care professionals to provide evidence-based counselling regarding the safety and efficacy of the available vaccines in these populations. This commentary reviews the current data on the safety of COVID-19 vaccines in pregnancy. The evidence is clear that the risks of hospitalization and severe maternal and potentially fetal morbidity from COVID-19 in pregnancy far outweigh the very minimal risks of COVID-19 vaccination in pregnancy.The successes with immune checkpoint blockade (ICB) and chimeric antigen receptor (CAR)-T-cell therapy in treating multiple cancer types have established immunotherapy as a powerful curative option for patients with advanced cancers. Unfortunately, many patients do not derive benefit or long-term responses, highlighting a pressing need to perform complete investigation of the underlying mechanisms and the immunotherapy-induced tumor regression or rejection. In recent years, a large number of single-cell technologies have leveraged advances in characterizing immune system, profiling tumor microenvironment, and identifying cellular heterogeneity, which establish the foundations for lifting the veil on the comprehensive crosstalk between cancer and immune system during immunotherapies. In this review, we introduce the applications of the most widely used single-cell technologies in furthering our understanding of immunotherapies in terms of underlying mechanisms and their association with therapeutic outcomes. We also discuss how single-cell analyses help to deliver new insights into biomarker discovery to predict patient response rate, monitor acquired resistance, and support prophylactic strategy development for toxicity management. Finally, we provide an overview of applying cutting-edge single-cell spatial-omics to point out the heterogeneity of tumor-immune interactions at higher level that can ultimately guide to the rational design of next-generation immunotherapies.
To study the utility of clinical exome sequencing (CES) using next generation sequencing (NGS) in evaluating neonates with suspected genetic conditions.
This is an observational study conducted in a tertiary care neonatal unit. We included neonates with suspected genetic conditions, for whom CES were done either by direct sampling or from stored DNA. Data was collected from the Sri Ramachandra centre of excellence in perinatal health (SCOPE) case records of 2016-2019. Yield of CES, percentage of pathogenic, non-pathogenic and variant of uncertain significance (VUS) and associated disorders were studied.
CES was done in 36 neonates. Variants were detected in 78% (28/36). However, significant variants with clinical correlation were present in 20 (56%) babies. Test was carried out from the stored sample in 10 (28%) babies. Mean turn-around time was 39±7 days. Specialist was involved in 1 and treatment changes were done in 5 neonates. Five out of 8 VUS were clinically correlating. Inborn errors of metabolism were the commonest (60%).
