Network pharmacology is a new discipline that integrates pharmacology, bioinformatics, and systems biology. It can be used to analyze the mechanism of action of drugs in the body from a holistic perspective by constructing a "disease-gene-drug" network, providing a new method to explore compounding mechanisms of Chinese medicine. Based on network pharmacology, this study explored the mechanism of the reduction of cardiotoxicity of anthracyclines by Qishen Huanwu Capsule.

The active ingredients of Qishen Huanwu Capsule and their targets were screened based on the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform and Chemistry Database. The target genes of ACT were screened through the PharmGkb, GeneCards, Online Mendelian Inheritance in Man (OMI, forkhead box class O (FoxO) and other signaling pathways by regulating targets such as RAC-alpha serine/threonine protein kinase (Akt1), mitogen-activated protein kinase 1 (MAPK1), and mitogen-activated protein kinase 8 (MAPK8) and thereby inhibits oxidative stress and regulates apoptosis and autophagy to reduce the cardiotoxicity of anthracyclines.

Thalidomide has been used to treat ankylosing spondylitis (AS) patients, but the efficacy and safety of thalidomide on psychological symptoms and sleep disturbances in the patient with refractory AS has not been evaluated.

In this 6-month open study, 35 cases of patients with refractory AS were recruited, using thalidomide at a dose of 150 mg/d before sleep. Bath Ankylosing Spondylitis Disease Activity Index (BASDAI), Bath Ankylosing Spondylitis Functional Index (BASFI), Bath Ankylosing Spondylitis Metrology Index (BASMI), fingertip-to-floor distance, nocturnal pain, total back pain, psychological symptoms, and sleep were assessed at baseline, 3 months and 6 months respectively.

Clinical parameters, included BASDAI, BASFI, fingertip-to-floor distance, nocturnal pain, total back pain, ESR, CRP, SAS, SDS and PSQI lower levels of month 3 were found when compared to initial levels (P<0.05). In the 6th month, the levels of BASDAI, fingertip-to-floor, nocturnal pain, total back pain, SDS were further lower than those in the 3rd month (P<0.05). The levels of BASMI were lower than this at baseline (P<0.05). The most tolerable common adverse reactions were drowsiness (10/35), dry mouth (8/35), constipation (8/35), dandruff (7/35), dizziness (4/35).

PubMed, EMBASE, Cochrane Library and Web of Science were searched to obtain the eligible studies following the "population, interventions, comparisons, outcomes, study design" (PICOS) criteria. The pooled analysis of objective response rate (ORR), disease controlled rate (DCR), progressionfree survival (PFS), overall survival (OS) and adverse events (AEs) were performed using STATA12.0 and RevMan5.0.

A total of 1,050 patients were included in the meta-analysis. The combined osimertinib ORR was 0.64 (95% CI, 0.60-0.69), the ORR of central nervous system (CNS) was 0.54 (95% CI, 0.37-0.71), DCR was 0.89 (95% CI, 0.86-0.92), PFS at six months (PFS-6m) rate was 0.69 (95% CI, 0.58-0.79), PFS at one year (PFS-1y) rate was 0.33 (95% CI, 0.20-0.46), OS at one year (OS-1y) rate was 0.69 (95% CI, 0.55-0.84). The pooled incidence rate of the AEs of grade ≥ III was 0.25 (95% CI, 0.09-0.40). The results from Begg's and Egger's tests presented no publication bias in the included studies.
