There were 362 (3%) ADR labels of anaphylaxis. Of those ADR containing a reaction description (n=11 868), 89% of reports entered as allergy had a reaction description that was consistent with intolerance and 8% of the entered intolerances had descriptions consistent with allergy when assessed using predefined criteria.

Harpacticoid copepods are among the organisms most sensitive to neonicotinoids. We exposed the brackish copepod Nitocra spinipes to 4 neonicotinoid insecticides (clothianidin, imidacloprid, thiacloprid, and thiamethoxam) to investigate acute toxicity on adults (96-h exposure) and effects on larval development (7-d exposure). We used these results in combination with publicly available ecotoxicity data to derive environmental quality standards (EQS). These EQS were ultimately used in a single-substance and mixture risk assessment for the Belgian part of the North Sea. Acute toxicity testing revealed that immobilization is a more sensitive endpoint than mortality, with 96-h median effect concentration (EC50) values of 6.9, 7.2, 25, and 120 µg L-1 for clothianidin, thiacloprid, imidacloprid, and thiamethoxam, respectively. However, the mechanisms translating it to reproductive dysfunction are not fully elucidated. Three experiments were performed on Wistar rats were designed to evaluate Sertoli cell function under stress. In Experiment I, rats were randomised into three groups saline baseline group given saline, ASEMA baseline group given aqueous extract of Massularia acuminata, zinc baseline group administered zinc orally. In Experiment II, exposure to psychological stress (for 1 hour per day) was layered on Experiment I while Experiment III substituted stress with administration of dexamethasone (DX). Six rats were sacrificed per group per experiment on days 7 and 14 and the right testis was excised and processed for PAS-haematoxylin staining and the left used for Zn determination. Results show significantly lower testicular Zn level as well more intensely immunoexpression of p53 in saline stress and saline DX groups compared with other groups. Also seen are the presence of residual bodies in the seminiferous tubular lumen of saline groups in Experiments II and III suggesting failure of residual bodies to be transported back towards the basement membrane. This study demonstrates that psychological stress impairs the ability of Sertoli cells to recycle residual bodies.Aquatic exposures to aromatic compounds (ACs) may be important contributors to biological effects of oil spills. The present study examined the acute toxicity of 11 ACs and 3 binary AC mixtures on stage 1 American lobster larvae using a passive dosing test design. The ACs investigated covered a range of classes and log octanol-water partition coefficient values (KOW ; 2.5-5.5). Silicone O-rings were used to partition ACs into seawater and maintain stable exposures. Exposed lobster larvae were assessed for mobility and survival at 3, 6, 12, 24, 36, and 48 h. Fluorometry and gas chromatography-mass spectrometry measurements confirmed well-defined substance exposures. Expressing lethality in terms of chemical activities yielded values between 0.01 and 0.1, consistent with a baseline mode of action. Analysis of time-dependent median lethal/effect concentration (L/EC50) values were used to determine incipient values. An expected linear relationship between the incipient log L/EC50 and log KOW was fit to the empirical toxicity data to derive critical target lipid body burdens for immobilization and lethality endpoints. These values indicate that American lobster larvae fall on the sensitive end of the acute species sensitivity distribution. We used AC toxicity data to successfully predict toxicity of binary mixtures assuming additive toxicity. The observed time-dependent toxicity was inversely related to log KOW and occurred more quickly than reported previously. Observations that established tumor regressions in response to immune checkpoint inhibitors (ICIs) often persist, despite the use of steroids to mitigate ICI-related autoimmune breakthrough, are not sufficiently reassuring, because these observations do not address the potential blunting of immune priming at the initiation of ICI therapy. With increasing indications for ICI in combination with chemotherapy, this issue merits reconsideration. Professional society guidance advises that dexamethasone should be used as first-line prophylaxis for nausea and vomiting in patients receiving ICI and highly emetogenic chemotherapy combination regimens. Here, we review the availability of data on this subject and propose an alternative approach focused on the adoption of steroid minimization or sparing for prophylaxis of nausea until the underlying immune biology is better understood.
