The selected optimal liposomal formulation was successfully incorporated into a thermogel without loss of thermoresponsive properties, being suitable for administration as a subcutaneous injection. In the ex vivo permeation studies with fresh rodent skin, the thermogel with liposomes loaded with 0.5% LBPV (T-gel formulation 3) showed higher permeation rates compared to the starting formulation, thermogel with 0.5% LBPV (T-Gel 1), which will probably translate into better therapeutic benefits for treatment of postoperative analgesia, especially with regard to the number of doses applied.
Chronic myelogenous leukemia (CML) is rare in children, requiring extrapolation from treatment of adults. In this review, we explore similarities and differences between adult and pediatric CML with a focus on therapeutic advances and emerging clinical questions.
Pediatric CML is effectively treated with long-term targeted therapy using tyrosine kinase inhibitors (TKIs). Newly diagnosed pediatric patients in chronic phase can now be treated with imatinib, dasatinib, or nilotinib without allogeneic hematopoietic stem cell transplantation. While treatment-free remission is possible in adults in chronic phase with optimal response to therapy, data are currently insufficient to support stopping TKI in pediatrics outside of a clinical trial. Knowledge gaps remain regarding long-term and late effects of TKIs in pediatric CML. Targeted therapy has markedly improved outcomes for pediatric CML, while raising a number of clinical questions, including the possibility of treatment-free remission and long-term health implications of prolonged TKI exposure at a young age.

Nevertheless, in such studies, differences in socio-economic-educational status have not always been accounted for, with idiomatic translation of emotional concepts being a limitation, and the underlying psychophysiological mechanisms still un-researched. We set out to investigate whether native residents from Guinea-Bissau (West African culture) and Portugal (Western European culture)-matched for socio-economic-educational status, sex and language-varied in behavioural and autonomic system response during emotion recognition of nonverbal vocalizations from Portuguese individuals. Overall, Guinea-Bissauans (as out-group) responded significantly less accurately (corrected p less then .05), slower, and showed a trend for higher concomitant skin conductance, compared to Portuguese (as in-group)-findings which may indicate a higher cognitive effort stemming from higher difficulty in discerning emotions from another culture. Specifically, accuracy differences were particularly found for pleasure, amusement, and anger, rather than for sadness, relief or fear. Nevertheless, both cultures recognized all emotions above-chance level. The perceived authenticity, measured for the first time in nonverbal cross-cultural research, in the same vocalizations, retrieved no difference between cultures in accuracy, but still a slower response from the out-group. Lastly, we provide-to our knowledge-a first account of how skin conductance response varies between nonverbally vocalized emotions, with significant differences (p less then .05). In sum, we provide behavioural and psychophysiological data, demographically and language-matched, that supports cultural and emotion effects on vocal emotion recognition and perceived authenticity, as well as the universality hypothesis.Complement C9, as a member of terminal complement component (TCC) protein, plays important roles in innate immunity. However, some complement components appear to show difference and evolutionary complexity between higher and lower vertebrates. Hence, it is essential to carry on a study of evolutionary origin and systematic function of C9 in fish and non-fish vertebrates. This study aims to explore the complement gene evolution and potential function in fish based on molecular and structural biology. The optimal codons of C9 sequences tended to be closer to the genomes of lower vertebrates compared to higher vertebrates. Further, conserved amino acids in the C9 TMH1 region were identified, implying their potential functional association with MAC growth and pore formation. Transposons and simple repeats, as gene elements, exhibited a differential distribution in the genomic regions in different animal groups but were sparsely scattered around the sixth exon (TMH1 region). Notably, this demonstrated the regulatory complexity of the C9 gene in higher vertebrates. The negative selection pressures on fish and non-fish groups improved both the sequence conservation and similarity. Through gene/protein regulatory network and pathway analyses, the systematic function of C9 protein was showcased; thus, we could reveal the divergence of the systematic function of C9 across species from different evolutionary positions. In addition, more complicated functions of C9 in higher vertebrates could established by the altered spatial conformation of the protein. Collectively, the present study illustrates the C9 gene evolutionary process and the difference in its systematic function across multiple species. Such advances provide new insights for understanding the evolutionary and potential functions of complement C9.
