We assessed whether objectively measured low- and high-intensity physical activity (LPA and HPA) and sedentary time (ST) were associated with white matter connectivity, both throughout the whole brain and in brain regions involved in motor function. In the large population-based Maastricht Study (n = 1715, age 59.6 ± 8.1 (mean ± standard deviation) years, and 48% women), the amounts of LPA, HPA, and ST were objectively measured during 7 days by an activPAL accelerometer. In addition, using 3T structural and diffusion MRI, we calculated whole brain node degree and node degree of the basal ganglia and primary motor cortex. Multivariable linear regression analysis was performed, and we report standardized regression coefficients (stβ) adjusted for age, sex, education level, wake time, diabetes status, BMI, office systolic blood pressure, antihypertensive medication, total-cholesterol-to-HDL-cholesterol ratio, lipid-modifying medication, alcohol use, smoking status, and history of cardiovascular disease. Lower HPA was associated with lower whole brain node degree after full adjustment (stβ [95%CI] = - 0.062 [- 0.101, - 0.013]; p = 0.014), whereas lower LPA (stβ [95%CI] = - 0.013 [- 0.061, 0.034]; p = 0.580) and higher ST (stβ [95%CI] = - 0.030 [- 0.081, 0.021]; p = 0.250) was not. In addition, lower HPA was associated with lower node degree of the basal ganglia after full adjustment (stβ [95%CI] = - 0.070 [- 0.121, - 0.018]; p = 0.009). Objectively measured lower HPA, but not lower LPA and higher ST, was associated with lower whole brain node degree and node degree in specific brain regions highly specialized in motor function. Further research is needed to establish whether more HPA may preserve structural brain connectivity.
The CT of PET CT provides diagnostic information, anatomic localisation and attenuation correction (AC). When only AC is required, very lose dose CT is desirable. CT iterative reconstruction (IR) improves image quality with lower exposures however there is little data on very low dose IR CT for AC of PET. This work assesses the impact of CT exposure and reconstruction algorithm on PET voxel values.
An anthropomorphic torso phantom was filled with physiologically typical [18]F concentrations in heart, liver and background compartments. A 17-mm-diameter right lung "tumour" filled with [18]F was included (surrounding lung contained no 18[F]). PET was acquired followed by 24 CT acquisitions with varying CT exposures (15-50 mAs, 80-120 kVp, pitch 0.671 or 0.828). Each CT was reconstructed twice using filtered back projection (FBP) or IR and these used for AC of PET. The reference PET reconstruction (RR) used CT acquired at 50 mAs, 120 kVp, pitch 0.828, IR, all others were test PET reconstructions (TR). Regionsstruction-specific validation should be employed prior very low dose CT AC for PET.

Scanner- and reconstruction-specific validation should be employed prior very low dose CT AC for PET.In a captive environment, it is challenging to ensure the highest level of social and psychological well-being for species with naturally complex social organizations and structures. There is a growing need to meet the social requirements for individuals of these species, especially chimpanzees, housed in zoos, sanctuaries, rehabilitation centers, and laboratories. Complex social interactions and broader social structures can be aptly described via social network analysis. We expand on the literature regarding captive chimpanzee social networks, but uniquely consider their human caregivers as potential social partners. We observed the social interactions between one group of seven chimpanzees (Pan troglodytes) and their caregivers in six behavioral contexts (nearest neighbor, play, aggression, grooming, grooming solicitation, and social vigilance) at Chimpanzee Sanctuary Northwest (USA). By constructing multiple chimpanzee and chimpanzee-caregiver social networks, our results indicate that caregivers are integrated partners in this group's social structure. Additionally, we observed that the type and strength of chimpanzee-caregiver relationships varied between the chimpanzees. These results support the notion that caregivers offer additional opportunities (i.e., beyond those with conspecifics) for captive chimpanzees to construct and maintain meaningful social relationships. Our results show that considerations of captive primate social dynamics should include caregivers as potentially important social partners. Our results also highlight the importance of evaluating individual chimpanzee characteristics when developing philosophies of care and adopting husbandry practices that offer fulfilling social niches. Our findings bear influence on contemporary discussions of interspecies social relationships, captive welfare, health, translocation, and husbandry protocols for captive chimpanzees and other nonhuman primates.
Carcinoembryonic antigen is overexpressed in colorectal cancer (CRC), making it an optimal target for fluorescence imaging. A phase I/II study was designed to determine the optimal imaging dose of SGM-101 for intraoperative fluorescence imaging of primary and recurrent CRC.
Patients were included and received a single dose of SGM-101 at least 24h before surgery. Patients who received routine anticancer therapy (i.e., radiotherapy or chemotherapy) also were eligible. A dedicated near-infrared imaging system was used for real-time fluorescence imaging during surgery. Thirty-seven patients with CRC were included in the analysis. Fluorescence was visible in all primary and recurrent tumors. In seven patients, no fluorescence was seen; all were confirmed as pathological complete responses after neoadjuvant therapy. Two tumors showed false-positive fluorescence. In the 37 patients, a total of 97 lesions were excised. The highest mean intraoperative tumor-to-background ratio (TBR) of 1.9 (p = 0.019) was seen in the 10-mg dose. This dose showed a sensitivity of 96%, specificity of 63%, and negative predictive value of 94%. Nine patients (24%) had a surgical plan alteration based on fluorescence, with additional malignant lesions detected in six patients.
The optimal imaging dose was established at 10mg 4days before surgery. The results accentuate the potential of SGM-101 and designated a promising base for the multinational phase III study, which enrolled the first patients in June 2019.
