Type 2 inflammation is characterized by enhanced activity of interleukin (IL)-4, -5 and -13, and treatments targeting these pathways are available for treatment of severe asthma. At present, the pattern of pathway activity and the implications overlapping of pathway activity are unknown.
We hypothesized that clustering of airway mRNA expression would identify distinct molecular subtypes of severe asthma and thereby uncover the prevalence and overlap of pathway activity.
Sputum mRNA expression of genes related to expression of IL-5(CLC, CPA3 and DNASE1L3), IL-13(IL13Ra1, TNFSF14 and SERPINB2), T1/Th17 activity(IL1B, ALPL and CXCR2) and in vitro response to corticosteroids (FKBP512) and mepolizumab (ARAP3) was analysed in patients (n=109) with severe asthma and healthy controls (n=22). A cluster analysis of gene expression was performed. The response to a short course of OCS was assessed in a subset of patients (n=29).
Five molecular clusters were identified. Three had abundant T2gene expression of whicn of IL-5- and IL-13-related genes at moderate levels was present in almost half of patients, while marked elevated expression of both was rare. In contrast to IL-5, clusters with isolated IL-13- and Th1/Th17-related gene expression were not identified.Is workers' health more sensitive to losses than gains in job security? I address this question using the 1999 rise in the French Delalande tax as a quasi-natural experiment. The tax design allows to separately identify the causal impact of exogenous gains and losses in job security on workers' health. Difference-in-differences estimation results show that a greater job insecurity reduces significantly self-reported health. At the same time, more job security does not translate into a higher level of self-reported health.As native ranges are often geographically structured, invasive species originating from a single source population only carry a fraction of the genetic diversity present in their native range. The invasion process is thus often associated with a drastic loss of genetic diversity resulting from a founder event. However, the fraction of diversity brought to the invasive range may vary under different invasion histories, increasing with the size of the propagule, the number of reintroduction events, and/or the total genetic diversity represented by the various source populations in a multiple-introduction scenario. In this study, we generated a SNP data set for the invasive termite Reticulitermes flavipes from 23 native populations in the eastern United States and six introduced populations throughout the world. Using population genetic analyses and approximate Bayesian computation random forest, we investigated its worldwide invasion history. We found a complex invasion pathway with multiple events out of the native range and bridgehead introductions from the introduced population in France. Our data suggest that extensive long-distance jump dispersal appears common in both the native and introduced ranges of this species, probably through human transportation. Overall, our results show that similar to multiple introduction events into the invasive range, admixture in the native range prior to invasion can potentially favour invasion success by increasing the genetic diversity that is later transferred to the introduced range.Photo-electrochemical cathodic protection (CP) technology is considered to be a green metallic corrosion protection technology that uses solar energy to protect from corrosion and does not consume any anode materials. In this work, a CdIn2 S4 /WO3 nanocomposite photoelectrode was prepared, and its photo-electrochemical CP performance and mechanism were studied and analyzed. WO3 has a well band matching with CdIn2 S4 , leading to a significantly enhanced photo-electrochemical CP performance of the nanocomposite. Meanwhile, as confirmed in this work, the CdIn2 S4 /WO3 nanocomposite can store photoinduced electrons under light illumination through intercalation reactions and changing the valence state of tungsten. Moreover, it can discharge in the dark state to provide continuous CP for the coupled metals. This research will promote the practical application process of the photo-electrochemical CP technology.The objective of this study was to determine the effects of Nigella sativa oil extract on cardiometabolic risk factors in people with type 2 diabetes (T2D). A randomized, controlled, clinical trial was conducted on 43 patients with T2D (23 women and 20 men; aged 53.5 ± 7.4 years). The intervention group (N = 23) received two 500-mg per day soft gel capsules containing Nigella sativa oil extract and the control group (N = 20) received two identical placebo soft gel capsules containing sunflower oil per day for the same period, 8 weeks. Pre- and post-intervention cardiometabolic risk factors were measured. Compared with the placebo, the N. sativa oil significantly decreased FBS (p = .03(, HbA1c (p = .001), total cholesterol (p = .04), TG (p = .003), LDL-c (p = .001), BMI (p less then .001), waist circumference (p less then .001), SBP (p = .001), and DBP (p = .002). HOMA-IR (p = .51) and HDL-c (p = .91) did not change significantly following Nigella sativa supplementation. Nigella sativa oil exerted beneficial effects on glycemic control, serum lipid profile, blood pressure, and body weight among people with T2D. Further long-term trials in the future may help confirm the current therapeutic benefits of Nigella sativa in T2D.Herein, a new type of healable thermoplastic poly(urethane-urea) (PUU) elastomer with a unique dual dynamic network structure consisting of multi-strength hydrogen bonds and aromatic disulfide bonds, is designed and synthesized. The resultant PUU elastomer exhibits high tensile strength (41 MPa), great toughness (104 MJ m-3 ), and excellent self-healing capability (completely severes, heals at 60 ℃ for just 1 h, and recovers more than 80% of the original tensile strength). Although the PUU possesses high-density hydrogen bonds, it is completely homogeneous without micro-phase separation. MST-312 cost The unique dual dynamic network structure alleviates the adverse effects of strong molecular interactions on self-healing process, simultaneously endowing the polymer with outstanding mechanical properties and excellent self-healing capability at mild temperature. In addition, the underlying mechanism between performance and structure is revealed. The PUU has potential applications in soft robots and wearable electronics.
