Tuberculosis (TB) caused by Mycobacterium tuberculosis (MTB) is one of the leading causes of mortality worldwide, with an estimated 1.5 million deaths annually. Majority of infection cases are reported from the Southeast Asian region including India. After the discovery of Streptomycin in 1943 and its anti-tubercular activity in 1945, drug discovery efforts identified Isoniazid, Ethambutol, and Rifampin as TB-actives. However, over the years these drugs have been rendered ineffective due to genetic mutations in mycobacterial strains. This has shifted drug discovery efforts towards identifying new targets and drugs for drug-resistant forms of bacteria. TNO155 ATP synthase was identified as one of the key targets of MDR-TB. This review provides key insights of the ATP synthase target, structure activity relationship studies (SAR) of diarylquinoline class of inhibitors and their clinical relevance for treating MDR-TB.Polyphenol is an intricate bioactive molecule abundant in human/animal's diet particularly plant foods, and has been evidenced in numerous reports with health-promoting functions, owing to its possessed free radical scavenging properties and a broad spectrum of bioactivities. However, the beneficial functions are linked and restricted to bioavailability, which is dictated by the diversity of the gut microbiota. The human/animal's gut harbours a complex community of trillions of microbial species and their symbiotic relationship goes beyond mere aiding the host's digestive system, with important functions such as host nutrition and health by encouraging nutrient metabolism and absorption, regulation of the gastrointestinal development, protection against pathogens, maintenance of the gut mucosal barrier functions and host immune system. The disruption of the gut community (i.e., dysbiosis) is suggested to reflect several pathological processes, such as diabetes, obesity, and other metabolic-related comorbidities. Recent improvements in deepsequencing technologies and bioinformatics have enabled a more complex understanding of the reciprocal interactions of dietary polyphenols and gut microbiota, as well as their metabolic impact. Hence this review seeks to discuss the two-way synergistic interactions of dietary polyphenols and dietary constituents on the gut microbial composition with an updated and pivotal finding from literature suggesting whether these interactions depict a positive, negative, or neutralizing effect in the prevention of metabolic diseases.Cancer is amongst the leading public health problems globally with continuously increasing prevalence rate that demands for extensive and expensive treatment. Despite availability of number of potential cancer therapies, inadequate success has been achieved due to complexity and heterogeneity of tumors. Moreover, late/ terminal stage cancer leads to multidrug resistance, excruciating side effects, recurrence, etc. This is because of low penetrability and deleterious effects of drug on non-target cells/ tissues. This requires for cost effective, efficacious, alternative/ adjunct, complementary medicines with targeted drug delivery approach. A potential strategy to resolve this difficulty is to use theranostics i.e., formulations having both a therapeutic element and an imaging agent. Phytotherapeutics have been extensively used since times immemorial, having wide acceptability, easy availability, minimal side effects and comparatively inexpensive. These herbal formulations are mostly orally administered and thategy for cancer.
Active principles from natural sources, in the form of extracts and natural compounds, provide an infinite number of bioactive compounds with consummate disposal of chemical diversity. These compounds and active principles are of utmost importance in the discovery of drugs of biological origin particularly, from plants.

Development of resourceful technology for the isolation and extraction of bioactive compounds of medicinal importance is considered as an important task for researchers. There are a number of extraction, isolation, and characterization techniques currently utilized; however, most are laborious and use toxic chemicals and huge quantities of raw materials with a very low output. There are a number of abiotic and biotic factors that affects the quality and the quantity of plants bioactive compounds. Considering this the objectives of the current review is to discuss the various extraction and characterization techniques used to isolate the essential bioactive compounds from three plant speciefiting products with special reference to three industrially important plants Leonotis leonurus (L.) R.Br. (Lamiaceae) and Santalum album L. (Santalaceae) and Aloe vera (L.) Burm. f. (Aloaceae or Asphodelaceae).Exploration of new drugs targeting anxiety treatment is a major concern worldwide. Medicinal plants are being used as a potential source of novel drugs for anxiety disorders. The objective of this review is to provide information about the healing outcomes of anxiety treatment with natural products. Valeriana officinalis, Citrus aurantium, Commelina benghalensis, Achyranthes aspera, Mimosa pudica, Achillea millefolium, Nymphaea alba, Leonurus cardiac, Camellia sinensis, Turnera aphrodisiaca, Crataegus oxyacantha and Piper methysticum showed promising effects on anxiety in animal models. In clinical studies, passion flower, kava, valerian, St John's wort, and ashwagandha showed the most positive results. More studies are needed for the exploration of the anti-anxiety of medicinal plants. In drugs derived from natural sources have explored many components that are playing an essential role in curing anxiety disorders and associated complications.Changes in human lifestyles and environmental deterioration globally causes emergence of new viruses posing research challenges. Recent outburst of disease COVID-19 (nCoV19) is a recent example wherein effective management of virus using conventional medication or adopting preventive and effective diagnostic measures is a challenge. While many ongoing strategies from vaccine development to drug repurposing, are currently being investigated, a targeted approach with nanotechnology can be helpful to meet the demand for preventive and diagnostic measures. The significant results of nanotechnology in resolving better efficacy in pharmaceutical drugs is expected to be helpful in combating nCoV19 by using nanotechnology-based solutions preventive, treatment, and diagnosis. As vaccine development involves long clinical trial procedures, preventive measures such as masks, disinfectants, sanitizers, and personal protective equipment's are gaining popularity for effective management. The present write-up addresses the dire need of the nanotechnology-based solutions in present pandemic and studies of the ongoing innovation and existing patents for developing better solutions.
