There is considerable interest in the long-term brain health of retired contact and collision sport athletes; however little is known about possible underlying changes in functional brain connectivity in this group. We evaluated whole-brain functional connectivity patterns using multi-voxel pattern analysis (MVPA) to determine whether alterations in functional connectivity distinguish retired professional athletes from a matched group of healthy, community control subjects. Thirty-two retired athletes with a history of multiple self-reported sport-related concussions and thirty-six healthy community control subjects, who were similar in age and education, completed functional magnetic resonance imaging (fMRI). We identified brain regions with abnormal functional connectivity patterns using whole-brain multi-voxel pattern analysis (MVPA) as implemented in the Conn toolbox. First level MVPA was performed using 64 PCA components. Second level F test was performed using the first 3 MVPA components for retired athletes > controls group contrast. Post-hoc seed-to-voxel analyses using the MVPA cluster results as seeds were performed to characterize functional connectivity abnormalities from brain regions identified by MVPA. MVPA revealed one cluster of abnormal functional connectivity located in cerebellar lobule V. This region of lobule V corresponded to the ventral attention network. Post-hoc seed-to-voxel analysis using the cerebellar MVPA cluster as a seed revealed multiple areas of cerebral cortical hyperconnectivity and hypoconnectivity in retired athletes when compared to controls. This initial report suggests that cerebellar dysfunction might be present and clinically important in some retired athletes.Plant growth-promoting rhizobacteria (PGPR) are naturally occurring soil microorganisms that colonize roots and stimulate plant growth. Some PGPR strains can directly regulate plant growth in the absence of physical contact with the plant, via volatile organic compounds (VOCs) emissions. Recently, we have described that Arabidopsis thaliana respond differentially to diacetyl, a VOC from Bacillus amyloliquefaciens strain GB03 (GB03), through integral modulation of the immune system and the phosphate-starvation response (PSR) system, resulting in either mutualism or immunity. Under phosphate deficient conditions, diacetyl enhances salicylic acid- and jasmonic acid-mediated immunity and consequently causes plant hyper-sensitivity to phosphate deficiency. Here, we show that application of exogenous gibberellin (GA) partially alleviates the deleterious effect caused by either B. GI254023X concentration amyloliquefaciens GB03 VOCs or diacetyl in Arabidopsis under phosphate deficient conditions, while DELLA quadruple mutant exposed to GB03 VOCs exhibits a partial reduction on the stress symptoms. Moreover, diacetyl appears to enhance DELLA protein accumulation and increase the expression of several GA deactivation-related genes. These findings suggest that the DELLA-mediated GA signaling pathway is involved in the bi-faceted role of GB03 VOCs in regulating plant growth.Malignant mesothelioma is strongly associated with prior asbestos exposure. Recently there has been interest in the role of talc exposure in the pathogenesis of mesothelioma. We have analyzed lung tissue samples from a large series of malignant mesothelioma patients. Asbestos bodies were counted by light microscopy and mineral fiber concentrations for fibers 5 µm or greater in length were determined by scanning electron microscopy equipped with an energy dispersive spectrometer. The values were compared with 20 previously published controls. Among 609 patients with mesothelioma, talc fibers were detected in 375 (62%) and exceeded our control values in 65 (11%). Elevated talc levels were found in 48/524 men (9.2%) and 17/85 women (20%). Parietal pleural plaques were identified in 30/51 informative cases (59%) and asbestosis in 5/62 informative cases (8%). Commercial amphiboles (amosite and/or crocidolite) were elevated in 52/65 (80%) and noncommercial amphiboles (tremolite, actinolite or anthophyllite) in 41/65 (63%). Both were elevated in 34/65 (52%). Asbestos body counts by light microscopy were elevated in 53/64 informative cases (83%). A history of working in industries associated with asbestos exposure and increased mesothelioma risk was identified in 36/48 cases in men, and a history of exposure as household contacts of an occupationally exposed individual was identified in 12/17 cases in women. We conclude that among patients with mesothelioma, the vast majority have talc levels indistinguishable from background. Of the remaining 11% with elevated talc levels, the vast majority (80%) have elevated levels of commercial amphibole fibers.Dose selection is one of the most difficult and crucial decisions to make during drug development. As a consequence, the dose-finding trial is a major milestone in the drug development plan and should be properly designed. This article will review the most recent methodologies for optimizing the design of dose-finding studies all of them are based on the modeling of the dose-response curve, which is now the gold standard approach for analyzing dose-finding studies instead of the traditional ANOVA/multiple testing approach. We will address the optimization of both fixed and adaptive designs and briefly outline new methodologies currently under investigation, based on utility functions.Multiple autophagic processes are triggered in response to bacterial infection as the host attempts to eliminate intracellular invaders. However, it is still unclear how the mechanisms contributing to canonical macroautophagy/autophagy, including xenophagy, coordinate with the more recently described features that are characteristic of noncanonical autophagy. Recently, we revealed that infection with Streptococcus pneumoniae can trigger the formation of RB1CC1/FIP200-independent LC3-associated phagosome-like vacuoles (PcLVs) that contain the pneumococci at an early stage of infection. We also found that interactions of SQSTM1/p62 with the ATG16L1 WD domain are essential for PcLV formation. Intriguingly, PcLVs were required for the subsequent generation of bactericidal autophagic vacuoles (PcAVs). Furthermore, we also identified LC3-delocalized SQSTM1-positive PcLVs as intracellular intermediates that link PcLVs and PcAVs. These findings reveal a novel multi-step mechanism that contributes to xenophagy of the critical S.
