The composition of [Ru(III)-FBIMMT] complex has been established from Job's continuous variation method, mole ratio method, and log-log plot method. The specificity towards ruthenium(III) is well studied and appropriate masking agents are applied wherever required to boost it. The intra-day and inter-day precision values are found to be brilliant with % relative standard deviation of 0.52 and 0.68 respectively with % accuracy within the range of 99.00-100. The method is effectively used for determination of ruthenium(III) from water samples, binary and ternary synthetic mixtures, fissium alloy samples and catalyst materials. A scheme for sequential group separation of ruthenium(III), palladium(II) and osmium(VIII) has also been developed. The reproducible results of the present method confirm that the method has a good potential for quantitative determination of ruthenium(III) from various matrices.This work was attempted to evaluate the feasibility of a constructed on-line NIR platform coupled with efficient algorithms for rapid and robust quantification of quality parameter in cherry tomato. Specifically, a system was developed based on shortwave NIR spectroscopy for on-line quality inspection of cherry tomatoes. The spectra were recorded in diffuse reflectance mode from 900 to 1700 nm, and the conveyor belt speed was fixed to five samples per second. Three novel methods, namely variable combination population analysis (VCPA), uninformative variable elimination (UVE) and competitive adaptive reweighed sampling algorithm (CARS) were coupled with partial least square (PLS) for selecting optimal dataset, and modeling. The obtained results showed that under the optimal tuning parameters (N = 100, k = 500, ω = 14, σ = 10%), a total of 512 original variables, only 9 variables (1.75%) were extracted by VCPA. Subsequently, VCPA-PLS yielded outstanding performance in predicting soluble solid content in cherry tomatoes, with a higher correlation coefficient (RP = 0.9053), and lower root mean square errors (RMSEP = 0.382) in prediction set. This methodology demonstrated the versatile potential of the proposed installation coupled with VCPA methods for on-line detection of total soluble solids in cherry tomatoes.
Evidence is accruing to suggest that microbiota-gut-brain signalling plays a regulatory role in cardiorespiratory physiology. Chronic intermittent hypoxia (CIH), modelling human sleep apnoea, affects gut microbiota composition and elicits cardiorespiratory morbidity. We investigated if treatment with prebiotics ameliorates cardiorespiratory dysfunction in CIH-exposed rats.
Adult male rats were exposed to CIH (96 cycles/day, 6.0% O
at nadir) for 14 consecutive days with and without prebiotic supplementation (fructo- and galacto-oligosaccharides) beginning two weeks prior to gas exposures.
CIH increased apnoea index and caused hypertension. CIH exposure had modest effects on the gut microbiota, decreasing the relative abundance of Lactobacilli species, but had no effect on microbial functional characteristics. Faecal short-chain fatty acid (SCFA) concentrations, plasma and brainstem pro-inflammatory cytokine concentrations and brainstem neurochemistry were unaffected by exposure to CIH. The co-receptor lymphocyte-activation gene-3 (LAG3, LAG-3, CD223) is a potential target for immune checkpoint inhibition immunotherapies. However, little is known about the biological and clinical significance of LAG3 DNA methylation in melanoma and its microenvironment.
We evaluated LAG3 promoter and gene body methylation in a cohort of N=470 melanoma patients obtained from The Cancer Genome Atlas (TCGA cohort), an independent cohort of N=120 patients from the University Hospital Bonn, and in subsets of peripheral blood leukocytes, melanocytes, and melanoma cell lines. #link# We validated the association of LAG3 methylation with mRNA expression in vitro in the melanoma cell line A375 treated with the hypomethylating agent 5-azacytidine and stimulated with interferon-γ. Finally, we investigated correlations between LAG3 methylation and progression-free survival in patients treated with immune checkpoint blockade (ICB cohort, N=118).
Depending on the analysed locus (promoter, gene body) we found region-dependentive biomarker for response to anti-LAG3 immune checkpoint blockage.

Recently, we reported that patients with mild cognitive impairment (MCI) harbor specific signature of bacteria in their gut and that a modified Mediterranean ketogenic diet (MMKD) improves Alzheimer's disease (AD) markers in cerebrospinal fluid (CSF) and the signatures of gut bacteria. However, other microbial population such as gut fungi (mycobiome) in relation to MCI/AD pathology, gut bacteria and diet remain unknown.
We measure gut mycobiome by sequencing of the fungal rRNA ITS1 gene in 17 older adults (11 MCI; 6 cognitively normal [CN]) in a single-center, randomized, double-blind, crossover pilot study, before and after 6 weeks intervention of MMKD and American Heart Association Diet (AHAD), and determine its correlation with AD markers in CSF and gut bacteria.
Compared to CN counterparts, patients with MCI have higher proportion of families Sclerotiniaceae, Phaffomyceteceae, Trichocomaceae, Cystofilobasidiaceae, Togniniaceae and genera Botrytis, Kazachstania, Phaeoacremonium and Cladosporium and lower abundance of Meyerozyma.
