Collectively, our findings suggest that A7 could serve as a promising lead for the development of more efficient microtubule polymerization inhibitors for cancer therapy.Glyphosate-based herbicides (GBH) are the main pesticides applied worldwide on maize production. Glyphosate-resistant weeds led to the repeated application of high doses of the pesticide. In addition to environmental conditions, the presence of GBH affects the development of Aspergillus species and aflatoxin B1 (AFB1) production under in vitro conditions. The aim of this work was to evaluate the influence of a commercial GBH on growth and AFB1 production by Aspergillus flavus and Aspergillus parasiticus strains under different water activity (aW) conditions. The following concentrations of active ingredient glyphosate were evaluated 20, 50, 200 and 500mM. The lag phase prior to growth and growth rate did not change at 20 and 50mM (that is, at field recommended doses) at 0.98 and 0.95 aW; however, at increasing GBH concentrations, between 200 and 500mM, the growth rate decreased at all aW conditions. In general, as the GBH concentration increased, AFB1 production decreased. However, a significant increase in toxin accumulation was found only at one of the aW conditions (0.95) at 21 days with 50mM of GBH in A. flavus and 20 and 50mM of GBH in A. parasiticus. These results show that, even though Aspergillus section Flavi growth did not increase, AFB1 production increased on maize grains at GBH concentrations similar to those of field recommended doses under favorable water availability and temperature conditions.Alkaline wetlands distributed in arid or semi-arid areas are hotspots of methane (CH4) emissions. Periods of drought and flood, although regular, are stressful events encountered by methanogenic anaerobes in alkaline wetlands. To investigate the response of the CH4 cycle of alkaline wetlands to such stresses, we take Zhalong wetland as an example, then determined the CH4 flux and soil microbiomes in the wetland during wet, dry, and flooded periods. The in-situ CH4 flux in the wet period was 9.55-17.29 mg‧m-2‧h-1, but sharply degraded to 3.37-6.61 mg‧m-2‧h-1 in the dry period. It resumed to 4.51-20.80 mg‧m-2‧h-1 when the wetland was flooded again, which indicated that methanogenesis is quite resilient to drought. Syntrophic acetogenesis, and subsequently aceticlastic methanogenesis, were the dominant methanogenic pathways and resisted drought. Members belonging to Syntrophobacterales were the dominant syntrophic acetogens. They enter a viable but non-culturable (VBNC) state to resist drought. The dominant Methanosarcinales have the ability to repair reactive oxygen species damage during dry periods. The community of CH4 sink was governed by anaerobic methanotrophs, which entered a VBNC state or used repair systems to survive dry periods. This study revealed the responses of the CH4 cycle and microbial functional genes to drought and flood in alkaline wetlands.Bisphenol A (BPA), an environmental pollutant, has received considerable attention worldwide for its hazardous effects of promoting atherosclerosis and increasing the risk of cardiovascular diseases (CVDs). However, the mechanisms involved are unclear. We aimed to investigate the mechanisms underlying BPA-aggravated atherosclerosis and potential preventive treatments. Four-week-old male Ldlr-/- C57BL/6 mice were administered 250 μg/L BPA via drinking water for 30 weeks with or without a Western diet and/or resveratrol (RESV) for 12 weeks. Chronic BPA exposure significantly aggravated atherosclerosis, enhanced the production of inflammatory cytokines but not lipid levels, promoted macrophage infiltration into plaque areas. Moreover, peritoneal macrophages isolated from BPA-exposed mice exhibited a more pro-inflammatory phenotype in response to cholesterol crystal treatment than those from control mice. The comet assay revealed that the DNA repair capacity of BPA-exposed macrophages was impaired, and western blotting showed that sirtuin 1 and Nijmegen breakage syndrome 1 (NBS1) expression was reduced.
