To explore this hypothesis, we compared the acute toxicity of silver and zinc oxide nanoparticles (nAg and nZnO) to zebrafish larvae that were either germ-free or colonized by microbiota. Over two days of exposure, germ-free zebrafish larvae were more sensitive to nAg than microbially colonized larvae, whereas silver ion toxicity did not differ between germ-free and colonized larvae. Using response addition modeling, we confirmed that the protective effect of colonizing microbiota against nAg toxicity was particle-specific. Nearly all mortality among germ-free larvae occurred within the first day of exposure. In contrast, mortality among colonized larvae increased gradually over both exposure days. Concurrent with this gradual increase in mortality was a marked reduction in the numbers of live host-associated microbes, suggesting that bactericidal effects of nAg on protective microbes resulted in increased mortality among colonized larvae over time. No difference in sensitivity between germ-free and colonized larvae was observed for nZnO, which dissolved rapidly in the exposure medium. At sublethal concentrations, these particles moreover did not exert detectable bactericidal effects on larvae-associated microbes. Altogether, our study shows the importance of taking host-microbe interactions into account in assessing toxic effects of nanoparticles to microbially colonized hosts, and provides a method to screen for microbiota interference with nanomaterial toxicity.PURPOSE Merkel cell carcinoma (MCC) is a rare, aggressive skin cancer commonly driven by the Merkel cell polyomavirus (MCPyV). The programmed death-1 (PD-1)/programmed death-ligand 1 (PD-L1) immunosuppressive pathway is often upregulated in MCC, and advanced metastatic MCC frequently responds to PD-1 blockade. We report what we believe to be the first trial of anti-PD-1 in the neoadjuvant setting for resectable MCC. METHODS In the phase I/II CheckMate 358 study of virus-associated cancer types, patients with resectable MCC received nivolumab 240 mg intravenously on days 1 and 15. Surgery was planned on day 29. Tumor regression was assessed radiographically and microscopically. Tumor MCPyV status, PD-L1 expression, and tumor mutational burden (TMB) were assessed in pretreatment tumor biopsies. RESULTS Thirty-nine patients with American Joint Committee on Cancer stage IIA-IV resectable MCC received ≥ 1 nivolumab dose. Three patients (7.7%) did not undergo surgery because of tumor progression (n = 1) or adverse indings is warranted.Objectives Patients receiving long-term anticoagulants were reported with varied health-related quality of life (HrQoL). We assessed HrQoL in patients receiving either warfarin or dabigatran from a tertiary care hospital.Methods A cross-sectional study was carried out following consent from patients on oral anticoagulants. Demographics, prothrombin time international normalized ratio (PT-INR), and drug-related details were collected. A validated Arabic version of the perception of anticoagulant treatment questionnaire (PACT-2) was used to assess HrQoL under three dimensions convenience; burden of disease; and treatment satisfaction.Results One-hundred and fifty patients were recruited. Overall good quality of life was observed as indicated by the average score of 80.3 in the warfarin group and, moderate in the dabigatran (average score of 68). Highly adherent and elderly patients receiving warfarin were significantly more likely to have good quality of life. Therapeutic PT-INR and high medication adherence were the primary domains significantly associated with good quality of life amongst patients with warfarin.Conclusion We observed good quality of life in patients receiving warfarin particularly those in the categories of elderly, with therapeutic PT-INR and high medication adherence. Small sample size with dabigatran precludes any firm conclusions.PURPOSE Like other malignancies, GI stromal tumors (GIST) are highly heterogeneous. This not only applies to histologic features and malignant potential, but also to geographic incidence rates. Several studies have reported GIST incidence and prevalence in Europe and North America. In contrast, GIST incidence rates in South America are largely unknown, and only a few studies have reported GIST prevalence in Latin America. PATIENTS AND METHODS Our study was part of a collaborative effort between Chile and Mexico, called Salud con Datos. We sought to determine GIST prevalence and patients' clinical characteristics, including survival rates, through retrospective analysis. RESULTS Overall, 624 patients were included in our study. Our results found significant differences between Mexican and Chilean registries, such as stage at diagnosis, primary tumor location, CD117-positive immunohistochemistry status, mitotic index, and tumor size. Overall survival (OS) times for Chilean and Mexican patients with GIST were 134 and 156 months, respectively. No statistically significant differences in OS were detected by sex, age, stage at diagnosis, or recurrence status in both cohorts. As expected, patients categorized as being at high risk of recurrence displayed a trend toward poorer progression-free survival in both registries. CONCLUSION To the best of our knowledge, this is the largest report from Latin America assessing the prevalence, clinical characteristics, postsurgery risk of recurrence, and outcomes of patients with GIST. PD-1/PD-L1 Inhibitor 3 clinical trial Our data confirm surgery as the standard treatment of localized disease and confirm a poorer prognosis in patients with regional or distant disease. Finally, observed differences between registries could be a result of registration bias.
