Degradome sequencing is commonly used to generate high-throughput information on mRNA cleavage sites mediated by small RNAs (sRNA). read more In our datasets of potato (Solanum tuberosum, St) and Phytophthora infestans (Pi), initial predictions generated high numbers of cleavage site predictions, which highlighted the need of improved analytic tools. Here, we present an R package based on a deep learning convolutional neural network (CNN) in a machine learning environment to optimize discrimination of false from true cleavage sites. When applying smartPARE to our datasets on potato during the infection process by the late blight pathogen, 7.3% of all cleavage windows represented true cleavages distributed on 214 sites in P. infestans and 444 sites in potato. The sRNA landscape of the two organisms is complex with uneven sRNA production and cleavage regions widespread in the two genomes. Multiple targets and several cases of complex regulatory cascades, particularly in potato, was revealed. We conclude that our new analytic approach is useful for anyone working on complex biological systems and with the interest of identifying cleavage sites particularly inferred by sRNA classes beyond miRNAs.Ergotism is a common and increasing problem in Saskatchewan's livestock. Chronic exposure to low concentrations of ergot alkaloids is known to cause severe arterial vasoconstriction and gangrene through the activation of adrenergic and serotonergic receptors on vascular smooth muscles. The acute vascular effects of a single oral dose with high-level exposure to ergot alkaloids remain unknown and are examined in this study. This study had two main objectives; the first was to evaluate the role of α1-adrenergic receptors in mediating the acute vasocontractile response after single-dose exposure in sheep. The second was to examine whether terazosin (TE) could abolish the vascular contractile effects of ergot alkaloids. Twelve adult female sheep were randomly placed into control and exposure groups (n = 6/group). Ergot sclerotia were collected and finely ground. The concentrations of six ergot alkaloids (ergocornine, ergocristine, ergocryptine, ergometrine, ergosine, and ergotamine) were determined using HPLC/MS t exposed group compared to the control group at two of the three concentrations of TE (TE 30 nM, p = 0.36; TE 100 nM, p less then 0.001; TE 300 nM, p less then 0.001). Similar to chronic exposure, acute exposure to ergot alkaloids results in increased vascular sensitivity to PE. TE is a more potent dose-dependent antagonist for the PE contractile response in sheep exposed to ergot compared to the control group. This study may indicate that the dry gangrene seen in sheep, and likely other species, might be related to the activation of α1-adrenergic receptor. This effect may be reversed using TE, especially at early stages of the disease before cell death occurs. This study may also indicate that acute-single dose exposure scenario may be useful in the study of vascular effects of ergot alkaloids.This research seeks to explore the impact of COVID-19 on depressive symptoms, analyzing discrepancies of sexual orientation in a Portuguese-speaking sample. 1590 individuals participated, of which 63% were women, and 88% self-identified as straight. Participants responded to the depression sub-scale of the Beck Symptoms Iventory-18, the fear of COVID-19 scale and the COVID-19 negative impact scale. Depressive symptoms observed were higher than expected, and several significant differences were obtained women and self-identified bisexual participants had higher levels of depressive symptoms compared to male and straight and gay or lesbian participants. Depressive symptoms negatively correlated with age and positively correlated with COVID-19 aggravated responses, fear of COVID-19, and negative impact of COVID-19. Hierarchical linear regression analysis showed that age, gender and sexual orientation explained 6% of the variance of depressive symptoms, and when fear and the negative impact of COVID-19 was added, the model explained 23% of results. This study provides an important contribution to the understanding of factors arising from the pandemic that may have an impact on the mental health of sexual minorities.Recently two peptides isolated from the Cuban freshwater snail Pomacea poeyana (Pilsbry, 1927) were described to have antimicrobial activity against bacterial pathogens. Here we show considerable activities of Pom-1 and Pom-2 to reduce the viability of C. albicans, C. parapsilosis and the less common species C. auris measured as the decrease of metabolic activity in the resazurin reduction assay for planktonic cells. Although these activities were low, Pom-1 and Pom-2 turned out to be highly potent inhibitors of biofilm formation for the three Candida species tested. Whereas Pom-1 was slightly more active against C. albicans and C. parapsilosis as representatives of the more common Candida species Pom-2 showed no preference and was fully active also against biofilms of the more uncommon species C. auris. Pom-1 and Pom-2 may represent promising lead structures for the development of a classical peptide optimization strategy with the realistic aim to further increase antibiofilm properties and other pharmacologic parameters and to generate finally the first antifungal drug with a pronounced dedication against Candida biofilms.The impact of lockdown due to the COVID-19 pandemic in April-June 2020 on UV exposure of office workers was assessed using an online survey on time spent outdoors and environmental data for different locations in the UK. Without the need for commuting and with the flexibility of homeworking, weekday time spent outdoors was higher in the 2020 lockdown than in the same period in 2017. The weekday erythema effective radiant exposure was higher in 2020 due to an additional 45 min outdoors in the late afternoon that was not observed in 2017 and high UV levels due to extremely sunny weather in spring. The lockdown did not impact the frequency of time spent outdoors around midday, which was still governed by work commitments, and at the weekends, no difference between 2020 and 2017 was observed. In 2020, responders felt that time outdoors was very important for their health and well-being.
